Phenotypic and genetic analysis of a large family with migraine-associated vertigo.

OBJECTIVES: To describe a large multigenerational family with migraine-associated vertigo (MAV) combining a detailed phenotypic and genetic analysis. BACKGROUND: Migraine-associated vertigo is said to be highly prevalent in the general population and, like other migraine syndromes, its etiology is felt to have a strong genetic component. However, so far, there have been no reports of large families with MAV. METHODS: Detailed clinical study was conducted on a large multigenerational family with MAV. Genetic study using identical-by-descent analysis with dense single nucleotide polymorphism (

Gene flow among Calomys musculinus (Rodentia, Muridae) populations in Argentina.

Calomys musculinus is a Sigmodontinae rodent inhabiting periodically disturbed habitats in the central eastern plains of Argentina. It is the natural reservoir host of Junin virus, the etiological agent of Argentine Hemorrhagic Fever (AHF). In order to analyze the levels of gene flow among populations of this species, allozymic variability at 26 loci was studied in 291 individuals from the endemic zone of AHF and localities outside it. All populations showed high levels of polymorphism (He between 0.107 and 0.144; P95% between 38 and 54%). Individual loci f values were in most cases negative,

Association between Alzheimer's disease and the NOS3 gene.

Alzheimer's disease (AD) is the most common form of neurodegenerative disorder of later life. Genetic studies have demonstrated that the apolipoprotein E (ApoE) gene is an important susceptibility locus; however, other environmental and genetic factors operating alone or in combination with ApoE must also be involved. Among candidate genes that may contribute to this residual risk is the endothelial nitric oxide synthase (NOS3) gene. NO release from vascular endothelium accounts in large part for endothelium-derived relaxing factor bioactivity. Abnormalities of cerebral small vessels occur ear

CYP17 gene polymorphic sequence variation is associated with hyperandrogenism in Kashmiri women with polycystic ovarian syndrome.

OBJECTIVE: Polycystic ovarian syndrome is a complex reproductive as well as endocrinological disorder characterized by anovulatory dysfunction, androgen excess and polycystic ovarian morphology. Hyperandrogenism is regarded as a cardinal feature of the disease. It is believed that the excess androgens are produced due to abnormality in steroid biosynthesis pathway wherein cytochrome P450, 17α-hydroxylase (CYP17) plays an imperative role. Therefore the objective of the present study was to analyze the T/C polymorphism in 5'UTR of CYP17 gene for its association with PCOS and hyperandrogenism in

Adaptive landscape and genetic divergence of Karnah sheep revealed by selection scans across multiple ovine breeds.

Indigenous sheep constitute vital reservoirs of adaptive genetic diversity, yet their genomic signatures of adaptation are poorly understood. The present study provides a comprehensive genomic characterization of the endangered Karnah sheep (Ovis aries) native to North Kashmir, India. A total of 41,812 high-confidence SNPs were used across 266 individuals representing nine geographically and functionally diverse breeds (i.e., Karnah, Garole, Deccani, Changthangi, Tibetan, Australian Merino, Rambouillet, Suffolk, and Texel). Population structure analyses revealed pronounced genetic stratificati

Triploidy in a pair of hybridizing salamanders at the far end of the speciation continuum.

Ecological and genetic interactions of species at the far end of the speciation continuum can often be studied in sympatry. Triturus cristatus (C) and Triturus marmoratus (M) are two deeply differentiated yet hybridizing salamander species that engage in a mosaic distribution over a wide zone of range overlap in the west of France. Interspecies hybrids are easy to distinguish from both parentals and occur at ca. 3% of the total breeding population. Most hybrids are thought to be F1 diploids (CM). We aim to identify triploid F1 hybrids with single nucleotide polymorphism (SNP) data and determin

Association of glutathione S-transferase polymorphisms (GSTM1 and GSTT1) with primary open-angle glaucoma: an evidence-based meta-analysis.

Studies investigating the associations between glutathione S-transferase (GST) genetic polymorphisms and primary open-angle glaucoma (POAG) have reported controversial results. Therefore, a meta-analysis was performed to clarify the effects of GSTM1 and GSTT1 polymorphisms on POAG risk. Published literatures from PubMed, EMBASE, ISI Web of Science and CBM databases were retrieved. All studies evaluating the association between GSTM1/GSTT1 polymorphisms and POAG were included. Pooled odds ratio (OR) and 95% confidence interval (CI) were calculated using fixed- or random-effects model. Eleven st

Reduced efficacy of natural selection on codon usage bias in selfing Arabidopsis and Capsella species.

Population genetic theory predicts that the efficacy of natural selection in a self-fertilizing species should be lower than its outcrossing relatives because of the reduction in the effective population size (N(e)) in the former brought about by inbreeding. However, previous analyses comparing Arabidopsis thaliana (selfer) with A. lyrata (outcrosser) have not found conclusive support for this prediction. In this study, we addressed this issue by examining silent site polymorphisms (synonymous and intronic), which are expected to be informative about changes in N(e). Two comparisons were made:

Cultivation of the rhabditid Poikilolaimus oxycercus as a laboratory nematode for genetic analyses.

Vulva formation is a paradigm for evolutionary developmental biology in nematodes. Not only do the number of vulval precursor cells (VPCs) differ between members in the Rhabditidae and Diplogastridae, they are also sculpted via different developmental mechanisms, either by cell fusion in most Rhabditidae or by programmed cell death in the Diplogastridae. In this context, the species Poikilolaimus oxycercus is the only known species in the family Rhabditidae to have a subset of the Pn.p cells commit programmed cell death during the patterning of the VPCs. Our current study introduces P. oxycerc

Host Genetic Factors in Q Fever Susceptibility.

Several indirect findings suggest that host-related factors influence susceptibility to Coxiella burnetii infection. We decided to explore the influence of genetic factors related to both innate and adaptive immunity in acute Q fever susceptibility. TLR2 (Arg753Gln) and TLR4 (Asp299Gly, Thr399Ile) polymorphisms, along with HLA-DRB1 alleles, were analyzed for 38 patients with acute Q fever, 38 matched controls, and 121 blood donors. No significant associations were found for TLR polymorphisms. However, HLA-DRB1*04 was more frequent in patients. HLA-DRB1 variants may play a role in Q fever susce

The efficacy and safety of eculizumab in patients and the role of C5 polymorphisms.

Eculizumab is an orphan drug with indications for extremely rare autoimmune disorders. It is primarily prescribed for use in patients with paroxysmal nocturnal hemoglobinuria and atypical hemolytic uremic syndrome; but is also highly effective in the treatment of myasthenia gravis, among others. By binding to the C5 protein in the complement system, eculizumab effectively inhibits cellular hemolysis and autoimmune reactions. Despite this effective treatment, some patients reported no improvement in symptoms. Genetic sequencing revealed three distinct C5 mutations in the non-responders and thes

Association of insulin receptor genetic variants with polycystic ovary syndrome in a population of women from Central Europe.

To assess the role of the insulin receptor gene in polycystic ovary syndrome (PCOS) we performed a case-control study in a female population (n=226) from Central Europe by examining the genetic associations of single nucleotide polymorphisms (rs8107575, rs2245648, rs2245649, rs2963, rs2245655, and rs2962) and inferred haplotypes around exon 9 of this gene. The ancestral T allele of single nucleotide polymorphism rs2963 or the corresponding haplotype (GGTC-C) showed association with PCOS with odds ratio 2.99, 95% confidence interval 1.4-6.3, independent of obesity but related to the presence of

Association of the androgen receptor gene (AR) with ADHD and conduct disorder.

The male predominance of externalizing behaviors suggests that the X-linked androgen gene might be involved. Since the shorter alleles of the CAG and GGC polymorphisms of the AR gene are associated with increased gene expression we sought to determine whether they were also associated with externalizing behaviors. We examined 302 subjects consisting of Tourette syndrome probands and controls. ANOVA showed a significant association between the AR haplotypes and ADHD (p < 0.0001), conduct disorder (CD; p < 0.017), and oppositional defiant disorder (ODD; p < 0.004) with the lowest scores in those

Association studies of estrogen receptor polymorphisms in a Norwegian testicular cancer population.

To examine a possible influence of estrogen receptor variants on the genetic susceptibility to germ cell cancer we have analyzed the allelic frequencies of three polymorphisms within the estrogen receptor gene in testicular cancer (n = 454) and control populations (n = 672). There were no differences in allelic frequencies in cancer patients compared to controls. Subgroup analyses did not indicate differences in allele frequencies for any of the polymorphisms in those individuals most likely to be predisposed to testicular cancer (patients with a history of maldescent of testicles and/or infer

Isolation of DNA fragments from chromosome 13.

A number of patients with retinoblastoma have a deletion of chromosome 13. Comparison of the deleted segments from different individuals reveals that all deletions involve chromosome band 13q14. This observation has lead to the hypothesis that in this region is a gene of genes important in the etiology of retinoblastoma. As a first step toward understanding those genes, the authors successfully isolated five DNA fragments from chromosome 13 using recombinant DNA techniques. The DNA fragments from chromosome 13 will be useful in identifying DNA polymorphic sites that are linked to the retinobla

The Genetic Changes of Hepatoblastoma.

Hepatoblastoma is the most common malignant liver cancer in childhood. The etiology of hepatoblastoma remains obscure. Hepatoblastoma is closely related to genetic syndromes, hinting that hepatoblastoma is a genetic predisposition disease. However, no precise exposures or genetic events are reported to hepatoblastoma occurrence. During the past decade, significant advances have been made in the understanding of etiology leading to hepatoblastoma, and several important genetic events that appear to be important for the development and progression of this tumor have been identified. Advances in

Glucocorticoid Receptor Gene Polymorphisms and Femoral Head Osteonecrosis.

Background and Objectives: Non-traumatic osteonecrosis of the femoral head (ONFH) is a multifactorial disorder influenced by both environmental and genetic factors. The nuclear receptor subfamily 3 group C member 1 (NR3C1) gene encodes the glucocorticoid receptor, which plays a key role in bone metabolism, vascular regulation, and stress response. This study aimed to investigate the association between NR3C1 polymorphisms and susceptibility to ONFH, with particular emphasis on age-related genetic effects. Materials and Methods: A hospital-based case-control study was conducted using genotyping
