Imbalanced intrahepatic cytokine expression of interferon-gamma, tumor necrosis factor-alpha, and interleukin-10 in patients with acute-on-chronic liver failure associated with hepatitis B virus infection.

GOALS: This study attempts to determine expressions of intrahepatic proinflammatory and anti-inflammatory cytokines and their secreting immunocytes to evaluate their roles in the pathogenesis of acute-on-chronic liver failure (ACLF) in chronically hepatitis B virus (HBV)-infected patients. BACKGROUND: ACLF generally affects patients with established, compensated chronic liver diseases who develop an acute deterioration in liver function. In China, HBV-associated ACLF patients account for more than 80% of ACLF patients owing to a high prevalence of chronic HBV infection. Clinical observation s

Membrane and transmembrane signaling in Herpesvirus saimiri-transformed human CD4(+) and CD8(+) T lymphocytes is ATM-independent.

In the genetic disorder ataxia telangiectasia (AT), humoral (B) and cellular (T) immunological abnormalities are frequently observed. As a consequence, AT patients are predisposed to life-threatening sinopulmonary infections. The pathogenic mechanisms remain unknown, but a role for ATM in signal transduction from membrane receptors has been proposed. We have explored the effects of a defective ATMgene on isolated human T-lineage cells from 13 AT patients with proven T cell dysfunction by transforming their CD4(+) and CD8(+) T lymphocytes with Herpesvirus saimiri, and analyzing their signaling

In situ characterization of the human host response to Leishmania panamensis.

Both host and parasite determinants influence the outcome of Leishmania infections. Human host responses in cutaneous leishmaniasis of limited duration caused by a single species of the Viannia (V) subgenus were studied in skin biopsies obtained from lesions caused by Leishmania (V) panamensis in 31 male patients from the Colombian Pacific Coast. Dermal infiltrates and histopathologic changes were characterized using monoclonal antibodies and an indirect immunoperoxidase method. Dermal distribution of T-cell subpopulations and B-lymphocytes was nonrandom: CD4+ and CD8+ T cells were most freque

Study of LDL and acetylated LDL endocytosis by mononuclear cells in HIV infection.

Activated lymphocytes have a high level of low density lipoprotein (LDL) uptake as compared to resting lymphocytes, whereas scavenger receptors for acetylated LDL (Ac-LDL) are expressed on limited number of immune cells, i.e., monocytes/macrophages. The endocytosis of LDL and Ac-LDL by mononuclear cells was studied during in vitro and in vivo HIV infection, in order to use LDL and Ac-LDL as carriers of antiviral and/or immunomodulatory drugs towards lymphocytes and monocytes. The uptake of LDL and Ac-LDL was analyzed by cytofluorimetry. LDL endocytosis in PHA/IL2-activated lymphocytes was high

Correlation of the Gut Microbiota and Antitumor Immune Responses Induced by a Human Papillomavirus Therapeutic Vaccine.

Human papillomavirus (HPV) is the most common sexually transmitted pathogen worldwide and the major risk factor for cervical cancer. According to our previous study, antitumor immune responses induced by a therapeutic vaccine based on HPV E7 peptide are highly variable among individuals. Many studies have demonstrated that the discrepancy in the gut microbiota is an important factor in the development and regulation of the immune system. Therefore, we performed a systematic comparative analysis of gut microbiota in two groups of mice with significant differences in antitumor effects induced by

Improved CD4 T cell profile in HIV-infected subjects on maraviroc-containing therapy is associated with better responsiveness to HBV vaccination.

BACKGROUND: Maraviroc-containing combined antiretroviral therapy (MVC-cART) improved the response to the hepatitis B virus (HBV) vaccine in HIV-infected subjects younger than 50 years old. We aimed here to explore the effect of this antiretroviral therapy on different immunological parameters that could account for this effect. METHODS: We analysed baseline samples of vaccinated subjects under 50 years old (n = 41). We characterized the maturational subsets and the expression of activation, senescence and prone-to-apoptosis markers on CD4 T-cells; we also quantified T-regulatory cells (Treg)

Induction of donor specific transplantation tolerance to cardiac allografts following treatment with nondepleting (RIB 5/2) or depleting (OX-38) anti-CD4 mAb plus intrathymic or intravenous donor alloantigen.

The nondepleting monoclonal antirat CD4 antibody, RIB 5/2, has been shown to modulate, but not eliminate, the CD4+ T cells and to prolong survival of rat skin, renal, or cardiac allografts when serially administered after transplantation. In the present study, we compared the efficacy of recipient pretreatment with a single dose of nondepleting RIB 5/2 or depleting OX-38 anti-CD4 monoclonal antibody plus donor alloantigen given intravenously or intrathymically 21 days before transplantation on the survival of completely MHC-mismatched rat cardiac allografts. Chemokine receptors and leukocyte trafficking in the mucosal immune system.

The CC chemokine receptors CCR6, CCR9, and CCR10 all contribute to the positioning of leukocytes at mucosal locations. Mucosal epithelial cells are major sources of the chemokine ligands for each of these receptors, although the pattern of expression of the individual ligands differs at distinct mucosal sites. CCR6 is expressed by most B cells, subsets of CD4 and CD8 memory T cells, and subsets of dendritic cells (DCs). Absence of CCR6 in mice leads to abnormal expansion of intestinal intraepithelial T cells and lamina propria T cells, smaller Peyer's patches, and defects in IgA-mediated respo

Severe CD8+ T Lymphopenia in WHIM Syndrome Caused by Selective Sequestration in Primary Immune Organs.

Warts, hypogammaglobulinemia, infections, and myelokathexis (WHIM) syndrome is an ultra-rare combined primary immunodeficiency disease caused by heterozygous gain-of-function mutations in the chemokine receptor CXCR4. WHIM patients typically present with recurrent acute infections associated with myelokathexis (severe neutropenia due to bone marrow retention of mature neutrophils). Severe lymphopenia is also common, but the only associated chronic opportunistic pathogen is human papillomavirus and mechanisms are not clearly defined. In this study, we show that WHIM mutations cause more severe

Localization of clonal T cells to the epidermis in cutaneous T-cell lymphoma.

BACKGROUND: Mycosis fungoides (MF) is a form of cutaneous T-cell lymphoma (CTCL) characterized by progression of clonal, epidermotropic T cells with the proliferative (Ki-67+) T-cell fraction primarily confined to the epidermis in early CTCL. OBJECTIVE: Our purpose was to determine whether the malignant clone (recognized by its clonal T-cell receptor [TCR] rearrangement) might also be localized to the epidermal compartment by differential Southern blot analysis. METHODS: A rapid heat-saline technique was used to separately isolate epidermal and dermal DNA from 11 patients with CTCL (1 with d

Comparative efficacy and safety of traditional Chinese medicine injections combined with chemotherapy for advanced non-small cell lung cancer: a Bayesian network meta-analysis.

BACKGROUND: Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality worldwide. Although chemotherapy is an important treatment strategy for advanced NSCLC, its efficacy is limited by treatment resistance and adverse events. Traditional Chinese medicine injections (TCMIs) are widely used as adjunctive therapies; however, the comparative efficacy and safety of different TCMI-based regimens remain unclear. This study aimed to compare the efficacy and safety of various TCMIs combined with chemotherapy using Bayesian network meta-analysis. METHODS: RCTs on TCMIs comb

Cancer associated fibroblast-T cell crosstalk promotes purinergic synthesis in non-small cell lung cancer (NSCLC).

The most abundant stromal cells of the tumor microenvironment (TME), cancer-associated fibroblasts (CAFs), inhibit the cytotoxic T cell response in solid tumors. CD39 and CD73 together deplete extracellular ATP in the TME to produce adenosine, which can promote tumor immune evasion. We aimed to investigate the role of CAF-T-cell crosstalk in the adenosine pathway in NSCLC. CD4+ and CD8+ T cells were flow sorted from in vitro culture with or without CAFs, and RNA-Seq was performed using the nCounter platform (NanoString). The expression of cell surface markers involved in adenosine production w

PD1+ TIGIT+ CD4+ T cells predict response to anti-TNF in rheumatoid arthritis and spondyloarthritis.

BACKGROUND: Programmed cell death protein 1 (PD-1) and T cell immunoreceptor with Ig and ITIM domains (TIGIT) are immune checkpoints expressed on T cells. Despite recent development of PD-1 agonists in rheumatoid arthritis (RA), little is known about PD-1 and TIGIT coexpression in RA and other immune-mediated inflammatory diseases (IMIDs). This study quantified PD-1 and TIGIT expression in CD4 and CD8 T cells in patients with IMID and examined associations with disease characteristics and anti-tumour necrosis factor (TNF) response. METHODS: Biologics-naïve patients with RA and spondyloarthrit

IL-10- and IL-13-Biased T Cell Responses to SARS-CoV-2 Vaccination in Diabetes.

Type 1 and type 2 diabetes are associated with increased severity and mortality from respiratory virus infections. Vaccination in the general population significantly reduces the risk of severe respiratory viral infection and triggers a strong, polyfunctional, and lasting T cell response in healthy individuals. However, vaccine effectiveness in people with type 1 diabetes is unclear. Here, we studied the magnitude and functional characteristics of vaccine-specific CD4+ and CD8+ T cell responses to vaccination in people with type 1 and type 2 diabetes and compared them to those of people living

Immune profiling-based targeting of pathogenic T cells with ustekinumab in ANCA-associated glomerulonephritis.

Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis is a life-threatening autoimmune disease that often results in kidney failure caused by crescentic glomerulonephritis (GN). To date, treatment of most patients with ANCA-GN relies on non-specific immunosuppressive agents, which may have serious adverse effects and be only partially effective. Here, using spatial and single-cell transcriptome analysis, we characterize inflammatory niches in kidney samples from 34 patients with ANCA-GN and identify proinflammatory, cytokine-producing CD4+ and CD8+ T cells as a pathogenic signature.
