EP3 receptor mediates calcium oxalate kidney stone formation via Ca²⁺/AKT and necroinflammation.

Kidney stone disease is a prevalent urological condition with rising incidence and high recurrence rates, yet effective pharmacological interventions remain lacking [1] Prostaglandin E₂ (PGE₂) and its receptor EP3 have been implicated in inflammatory kidney injury [1-3], but their role in nephrolithiasis is unclear. This study investigates the role and mechanism of EP3 in calcium oxalate (CaOx) stone formation using in vitro and in vivo models. EP3 expression was significantly upregulated in CaOx-stimulated human renal tubular epithelial cells (HK-2) and in kidney tissues of glyoxylate-induced…

Phenanthrene disrupts calcium transients and inhibits INCX in zebrafish (Danio rerio) ventricular myocytes.

Phenanthrene is a polycyclic aromatic hydrocarbon (PAH) that is derived from fossil fuels and is present in most PAH mixtures found in water, air and soil. It has been shown to affect cardiac function of vertebrates, including fish. We investigated the effects of phenanthrene on intracellular calcium transient ([Ca2+]i) and the Na+/Ca2+ exchanger (NCX) current (INCX) in zebrafish (Danio rerio) ventricular cardiomyocytes using Fluo-4 AM imaging and whole-cell patch-clamp, respectively. Phenanthrene exposure (25 µM) reduced the amplitude and slowed the rate of decay of Ca²⁺ transients. Phenanthr…

Calcium handling properties and arrhythmia vulnerability of cardiomyocytes from dystrophin-deficient mdx mice.

Cardiomyopathy and arrhythmia development significantly contribute to mortality in patients with Duchenne muscular dystrophy (DMD), a fatal muscle disorder caused by dystrophin deficiency. Previous studies suggested that abnormal Ca handling is an important causative factor for cardiac involvement in DMD. Here, we aimed to provide an extensive analysis of Ca handling properties and arrhythmia vulnerability of cardiomyocytes from the dystrophin-deficient mdx mouse, the most common DMD animal model. Whole cell patch clamp experiments showed that currents through L-type Ca channels are similar in

Compartment-specific analysis reveals disrupted astrocytic calcium transients in anesthetized Alzheimer's disease mice.

BackgroundAlzheimer's disease (AD) is characterized by presence of extracellular amyloid plaques, intracellular tau tangles, and extensive neuronal cell death. In addition to neurons, astrocytes modulate neuronal network activity through tripartite synapses. Also, astrocytes are increasingly recognized for their involvement in AD pathology. Aberrant astrocytic calcium signaling has been implicated in AD pathological processes, including disrupted synaptic transmission, dysregulated glutamate homeostasis, and impaired vascular function via astrocytic endfeet. Previous investigations have assess…

Distinct Mechanisms of Sodium-Calcium Exchanger NCX1.4 Regulation by Ca2+, ATP, and PIP2.

Mammalian Na+/Ca2+ exchangers (NCXs) mediate inward (Ca2+-exit) or outward (Ca2+-entry) currents (INCX), with the peak current (Ipeak) declining to the steady-state current (Iss). The Ca2+-, ATP-, and PIP2-dependent activation of cardiac (NCX1.1) inward INCX has been previously documented. To investigate possible interactions among regulatory modes, we examined the effects of ATP and PIP2 on outward INCX when the CBD1 and CBD2 domains of NCX1.4 (brain variant) are Ca2+-occupied and -unoccupied. At nominally zero cytosolic Ca2+, ATP and PIP2 significantly reduced the outward Ipeak amplitude. Wh

An osteoimmunomodulatory microneedle patch targeting ER-mitochondria calcium crosstalk for periodontal tissue regeneration.

Functional reconstruction of periodontal tissue defects remains a clinical challenge, primarily owing to sustained proinflammatory responses. Endoplasmic reticulum stress (ERS) and mitochondrial dysfunction have been identified as pivotal regulators driving macrophage proinflammatory activation, yet their pathological link remains unclear. In this study, transcriptomic analyses revealed that ERS and mitochondrial dysfunction were mutually reinforced via dysregulated intracellular Ca2+ transport. To target this mechanism, we designed dual-functional polydopamine nanoparticles (PDAB NPs) by leve

Multifunctional therapeutic platform based on polydeoxyribonucleotide-loaded calcium carbonate particles for osteoarthritis management.

Osteoarthritis (OA) is associated with a mildly acidic, oxidative, and inflammatory joint microenvironment that accelerates cartilage degeneration and limits the efficacy of intra-articular therapies. However, currently available intra-articular therapies fail to simultaneously address the three key pathological drivers of acidosis, oxidative stress, and inflammation that sustain this self-reinforcing OA disease loop. Here, we developed polydeoxyribonucleotide-loaded calcium carbonate (PDRN-CaCO3) particles as a multifunctional intra-articular platform that combines pH-responsive drug delivery…

Mitochondrial protein OPA3 sustains cardiac function by regulating calcium handling in male mice.

Heart failure (HF) is a growing global health burden characterized by impaired cardiac contractility and progressive remodeling, driven in part by disrupted Ca2+ handling and mitochondrial dysfunction. However, the molecular mechanisms coordinating these processes remain incompletely understood. Here we showed that OPA3 was decreased in both human and murine HF. Cardiomyocyte-specific deletion of Opa3 in male mice led to the progressive dilated cardiomyopathy (DCM), accompanied by impaired myocardial function, calcium cycling and mitochondria function. Mechanistically, OPA3 forms multimers tha…

Renal Vdr deletion drives urinary calcium loss with intestinal and skeletal compensatory responses.

1,25-Dihydroxyvitamin D3 (1,25(OH)2D3), parathyroid hormone (PTH), and fibroblast growth factor 23 (FGF23) are endocrine regulators of calcium homeostasis. To assess the contribution of 1,25(OH)2D3-mediated renal calcium reabsorption to systemic calcium balance and bone homeostasis, we generated mice with a targeted deletion of the vitamin D receptor (VDR) in renal tubules (VdrRen- mice), weaned them onto diets containing 1% or 0.2% calcium, and examined their phenotype at 8 weeks of age. Despite higher urinary calcium excretion, VdrRen- mice maintained normocalcemia, suggesting compensatory a…

Tubular Aggregate Myopathies: Genetic Heterogeneity and Diverse Clinical Features Converging on Calcium Dysregulation.

Tubular aggregate myopathy (TAM) is a rare inherited muscle disorder characterized by the abnormal accumulation of tubular aggregates (TAs) within skeletal muscle fibers. These aggregates, composed of compacted sarcoplasmic reticulum (SR) tubules, are strongly linked to disturbances in calcium (Ca2+) homeostasis. Clinically, TAM manifests with slowly progressive proximal muscle weakness, exercise intolerance, cramps, and myalgia, frequently beginning in childhood and often present with elevated serum creatine kinase levels. These symptoms can also be associated with some additional disorders, …

Calcium Imaging and Analysis in Beta Cells in Acute Mouse Pancreas Tissue Slices.

Ca2+ ions play a central role in the stimulus-secretion coupling cascade of pancreatic beta cells. The use of confocal microscopy in conjunction with the acute pancreas tissue slice technique offers valuable insights into changes in the intracellular calcium concentration following stimulation by secretagogues. This allows the study of beta cells on a single cell level, as well as their behavior on a multicellular scale within an intact environment. With the use of advanced analytical tools, this approach offers insight into how single cells contribute to the functional unit of islets of Lange…

Nanophysiology approach reveals diversity in calcium microdomains across zebrafish retinal bipolar ribbon synapses.

Rapid and high local calcium (Ca2+) signals are essential for triggering neurotransmitter release from presynaptic terminals. In specialized bipolar ribbon synapses of the retina, these local Ca2+ signals control multiple processes, including the priming, docking, and translocation of vesicles on the ribbon before exocytosis, endocytosis, and the replenishment of release-ready vesicles to the fusion sites for sustained neurotransmission. However, our knowledge about Ca2+ signals along the axis of the ribbon active zone is limited. Here, we used fast confocal quantitative dual-color ratiometric…

Iatrogenic Hypercalcaemia Secondary to Antibiotic-Eluting Absorbable Calcium-Sulphate Beads in Orthopaedic Surgery.

Calcium-sulphate beads (CSBs) are increasingly used to manage surgical site infections, serving both as local antibiotic carriers and as fillers for dead space. Although generally well tolerated, hypercalcaemia has been described as a serious adverse effect. In this case report, we describe a patient who developed symptomatic hypercalcaemia secondary to using CSBs during a proximal femoral excision (Girdlestone procedure). This study describes an 86-year-old woman who developed decreased consciousness and altered neurological status four days post a Girdlestone procedure with placement of 20 c…

Embryo-derived trypsin-induced calcium entry is inhibited by endometrial infertility factor, LEFTY2.

INTRODUCTION: A transient window of uterine receptivity ensures that embryos implant in an optimal endometrial environment. Failure to establish or premature closure of the implantation window is thought to be a major cause of infertility, which affects many couples globally. Embryos release trypsin, which designates its developmental potential and plays a crucial role in implantation. Calcium (Ca2+) signalling participates in receptivity and is thus a prerequisite for embryo implantation. Left-right determination factor 2 (LEFTY2) is a negative regulator of endometrial receptivity and is asso…

Altered Mitochondrial Bioenergetics and Calcium Kinetics in Young-Onset PLA2G6 Parkinson's Disease iPSCs.

Parkinson's disease (PD) has emerged as a multisystem disorder affecting multiple cellular and organellar systems in addition to the dopaminergic neurons. Disease-specific induced pluripotent stem cells (iPSCs) model early developmental changes and cellular perturbations that are otherwise inaccessible from clinical settings. Here, we report the early changes in patient-derived iPSCs carrying a homozygous recessive mutation, R741Q, in the PLA2G6 gene. A gene-edited R747W iPSC line mirrored these phenotypes, thus validating our initial findings. Bioenergetic dysfunction and hyperpolarization of

Cellular prion protein and calcium ions trigger the neurotoxicity of α-synuclein aggregates.

BACKGROUND: The aggregation of α-Synuclein (αS) into amyloid fibrils and their deposition in intraneuronal Lewy bodies are hallmark features of Parkinson's disease (PD) and other synucleinopathies. Among the molecular players implicated in αS toxicity, the cellular prion protein (PrPC) has emerged as a potential modulator of αS-neuron interactions. RESULTS: Using confocal microscopy, colocalization analysis and both siRNA-induced PrPC silencing and antibody-based blockade, we investigated the contribution of PrPC to αS-induced neurotoxicity in human iPSC-derived dopaminergic neurons, primary
