Comparative study of excretory-secretory proteins released by Schistosoma mansoni-resistant, susceptible and naïve Biomphalaria glabrata.

BACKGROUND: Schistosomiasis is a harmful neglected tropical disease caused by infection with Schistosoma spp., such as Schistosoma mansoni. Schistosoma must transition within a molluscan host to survive. Chemical analyses of schistosome-molluscan interactions indicate that host identification involves chemosensation, including naïve host preference. Proteomic technique advances enable sophisticated comparative analyses between infected and naïve snail host proteins. This study aimed to compare resistant, susceptible and naïve Biomphalaria glabrata snail-conditioned water (SCW) to identify pote

Parasites and Microbiota: Dual Interactions and Therapeutic Perspectives.

The human gut hosts a diverse and active community of bacteria that symbiotically support the physiology, metabolism, and immunity of the intestinal lining. Nevertheless, a dynamic community of parasites (helminths and protozoa) may share a habitat with gut-dwelling microbiota. Both microbiota and parasites can significantly change the physical and immunological environment of the gut, thus generating several mechanisms of interaction. Studying this field is crucial for understanding the pathogenesis of parasitic diseases. Additionally, intestinal microbiota and gut-dwelling parasites may inte

Generalized herd effects and vaccine evaluation: impact of live influenza vaccine on off-target bacterial colonisation.

Interactions between pathogens and commensal microbes are major contributors to health and disease. Infectious diseases however are most often considered independent, viewed within a one-host one-pathogen paradigm and, by extension, the interventions used to treat and prevent them are measured and evaluated within this same paradigm. Vaccines, especially live vaccines, by stimulating immune responses or directly interacting with other microbes can alter the environment in which they act, with effects that span across pathogen species. Live attenuated infl uenza vaccines for example, while safe

Silencing of OCH1 unveils the role of Sporothrix schenckii N-linked glycans during the host-fungus interaction.

BACKGROUND: Sporothrix schenckii is a neglected fungal pathogen for the human being and other mammals. In several fungal systems, Och1 is a Golgi α1,6-mannosyltransferase with a key function in the synthesis of N-linked glycans; which are important elements during the host-fungus interplay. The role of OCH1 in fungal virulence seems to be species-specific, being an essential component for Candida albicans virulence and dispensable during the interaction of Aspergillus fumigatus with the host. METHODS: Here, we silenced S. schenckii OCH1 and characterized the phenotype of the mutant strains.

Genomic insights into M. phaseolina: Multifactorial virulence via secreted effectors, carbohydrate enzymes and secondary metabolites.

Macrophomina phaseolina, a soilborne necrotrophic pathogen causing charcoal rot across more than 500 host plant species, poses a significant and escalating threat to global food security under climate change-driven abiotic stresses. Despite its destructive importance, the molecular architecture underlying its broad host range and virulence versatility remains poorly characterized. De novo whole-genome sequencing, assembly, and multi-platform functional annotation were performed using BUSCO v6.0.0, eggNOG-mapper, Gene Ontology, KEGG, COG, PHI-base, SignalP 6.0, DeepTMHMM, EffectorP 3.0, CAZy, a

Landscape structure affects the prevalence and distribution of a tick-borne zoonotic pathogen.

BACKGROUND: Landscape structure can affect pathogen prevalence and persistence with consequences for human and animal health. Few studies have examined how reservoir host species traits may interact with landscape structure to alter pathogen communities and dynamics. Using a landscape of islands and mainland sites we investigated how natural landscape fragmentation affects the prevalence and persistence of the zoonotic tick-borne pathogen complex Borrelia burgdorferi (sensu lato), which causes Lyme borreliosis. We hypothesized that the prevalence of B. burgdorferi (s.l.) would be lower on isla

CFEM proteins: unique to fungi with essential roles in plant host interactions.

Fungal diseases pose a serious threat to agriculture, forestry, and human health sectors and are major contributors to global economic losses. Plant-pathogenic fungi rely on a wide array of molecules that contribute to pathogenicity and virulence; thus, it is critical to understand their roles in plant-fungal interactions to propose control strategies. Among these, CFEM domain-containing proteins, which are unique to fungi primarily within the phyla Ascomycota and Basidiomycota, have diverse functions in the interaction with the host plant. The CFEM domain is characterized by the presence of e

Metal-based nanoparticles in antibacterial application in biomedical field: Current development and potential mechanisms.

The rise in drug resistance in pathogenic bacteria greatly endangers public health in the post-antibiotic era, and drug-resistant bacteria currently pose a great challenge not only to the community but also to clinical procedures, including surgery, stent implantation, organ transplantation, and other medical procedures involving any open wound and compromised human immunity. Biofilm-associated drug failure, as well as rapid resistance to last-resort antibiotics, necessitates the search for novel treatments against bacterial infection. In recent years, the flourishing development of nanotechno

A virus-induced circular RNA maintains latent infection of Kaposi's sarcoma herpesvirus.

Non-coding RNAs (ncRNAs) play important roles in host-pathogen interactions; oncogenic viruses like Kaposi's sarcoma herpesvirus (KSHV) employ ncRNAs to establish a latent reservoir and persist for the life of the host. We previously reported that KSHV infection alters a novel class of RNA, circular RNAs (circRNAs). CircRNAs are alternative splicing isoforms and regulate gene expression, but their importance in infection is largely unknown. Here, we showed that a human circRNA, hsa_circ_0001400, is induced by various pathogenic viruses, namely KSHV, Epstein-Barr virus, and human cytomegaloviru…

Characterization of microRNA candidates at the primary site of infectious bronchitis virus infection: A comparative study of in vitro and in vivo avian models.

Infectious bronchitis virus (IBV) is an important avian pathogen with a positive-sense single-stranded RNA genome. IBV is the causative agent of infectious bronchitis (IB), a primarily respiratory disease affecting chickens, with the ability to disseminate to other organ systems, such as the gastrointestinal, renal, lymphoid, and reproductive systems. Tracheal epithelial cells are the primary target of IBV, and these cells play a vital role in the effective induction of the antiviral response and eventual clearance of IBV. The host immune system is regulated by a number of different molecular

Spotted Fever Group Rickettsia Trigger Species-Specific Alterations in Macrophage Proteome Signatures with Different Impacts in Host Innate Inflammatory Responses.

The molecular details underlying differences in pathogenicity between Rickettsia species remain to be fully understood. Evidence points to macrophage permissiveness as a key mechanism in rickettsial virulence. Different studies have shown that several rickettsial species responsible for mild forms of rickettsioses can also escape macrophage-mediated killing mechanisms and establish a replicative niche within these cells. However, their manipulative capacity with respect to host cellular processes is far from being understood. A deeper understanding of the interplay between mildly pathogenic ri

Treatment with the Mycoparasite Pythium oligandrum Triggers Induction of Defense-Related Reactions in Tomato Roots When Challenged with Fusarium oxysporum f. sp. radicis-lycopersici.

ABSTRACT The influence exerted by the mycoparasite Pythium oligandrum in triggering plant defense reactions was investigated using an experimental system in which tomato plants were infected with the crown and root rot pathogen Fusarium oxysporum f. sp. radicis-lycopersici. To assess the antagonistic potential of P. oligandrum against F. oxysporum f. sp. radicis-lycopersici, the interaction between the two fungi was studied by scanning and transmission electron microscopy (SEM and TEM, respectively). SEM investigations of the interaction region between the fungi demonstrated that collapse and

Assessment of Survival and Replication of Brucella spp. in Murine Peritoneal Macrophages.

Brucella spp. are bacteria that naturally infect a variety of domesticated and wild animals leading to abortions and infertility, and these bacteria are also capable of causing debilitating human infections, which often result from human exposure to infected animals and animal products. The brucellae are intracellular pathogens that reside in host cells, including macrophages and dendritic cells, and it is paramount for the pathogenesis of Brucella that the bacteria are able to survive and replicate in these host cells. The methods outlined in this chapter can be employed to study the interact
