INTRODUCTION: Although often self-limited, acute pericarditis recurs in up to 30% of patients. Recurrent pericarditis (RP) causes substantial morbidity (pain, fatigue, and impaired quality of life) and can lead to constriction. Traditional treatments have important limitations, including corticosteroid dependence, recurrent flares, and treatment-related adverse effects. Emerging targeted immunomodulators are therefore under active investigation.
AREAS COVERED: This review summarizes the burden of pericarditis across etiologies, current therapies, and the global market for pericarditis treatme

Vascular dementia (VaD) is a debilitating cognitive disorder with limited pharmacological treatments. Naokang II decoction (NKII) is a traditional Chinese medicine (TCM) formula for VaD with significant efficacy, but its mechanisms remain unclear. This study integrated chemical profiling, network pharmacology, single-nucleus RNA sequencing (snRNA-seq) analysis, molecular docking, and in vitro validation to elucidate NKII's mechanisms against VaD. LC-MS/MS identified 412 components in NKII, of which 45 compounds were screened as active constituents. Their 1081 predicted targets overlapped with

Neutrophils are increasingly recognized as a heterogeneous lineage rather than a uniform population of short-lived innate effector cells. Low-density neutrophils (LDNs; often termed low-density granulocytes, LDGs) are operationally defined by their co-sedimentation with peripheral blood mononuclear cells after density-gradient centrifugation, but this physical definition encompasses cells with different maturation states, origins and functions. In rheumatic and autoimmune diseases, expansion of the low-density compartment is associated with disease activity, autoantibody titers, interferon sig

As the most prevalent cause of death worldwide, ischemic stroke urgently requires innovative therapeutic strategies. The present study demonstrated the therapeutic potential of human umbilical cord-derived mesenchymal stem cell-derived exosomes (hUMSC-Exos) in ameliorating hypoxia-induced cerebrovascular endothelial dysfunction through modulation of the AMPK/NLRP3 signaling pathway. Bioinformatics analysis of DisGeNET and exosomal cargo databases revealed 283 overlapping cerebral ischemia-related genes, implicating hUMSC-Exos in inflammatory regulation. In vitro experiments showed that hUMSC-E

Current therapies slow down advanced features but do not halt or reverse degeneration and neovascularization in dry and wet age-related macular degeneration (AMD). Recent research implicates the gastrointestinal microbiome as a potential critical modulator in AMD pathogenesis through the gut-retina axis. Dysbiosis, characterized by imbalanced microbial diversity, composition and function, can exacerbate systemic and retinal inflammation through microglial priming, inflammasome activation, and secretion of pro-angiogenic cytokines (IL-6, IL-1β, TNF-α, VEGF). Additionally, microbiome-derived met

The emergence of carbapenem-resistant Acinetobacter baumannii (CRAB) poses a major obstacle in treating sepsis, highlighting the urgent need for new therapeutic approaches. Recent studies have reported that disulfiram (DSF) exhibits antibacterial activity and potentiates meropenem activity against resistant A. baumannii (A. baumannii) isolates. The mechanisms by which DSF modulates inflammation during A. baumannii infection remain unclear, although DSF possesses anti-inflammatory properties by regulating the pyroptosis pathway. The present study aimed to investigate the potential effect of DSF

Hypobaric hypoxia (HH)-induced blood-brain barrier (BBB) disruption is a critical pathological event in high-altitude cerebral edema (HACE). However, the mechanistic role of extravasated immunoglobulin G (IgG) in this process remains insufficiently characterized. This study elucidates that leaked IgG actively contributes to neuroinflammation and BBB impairment under HH conditions, functioning beyond being a passive biomarker. Using both in vivo and in vitro models, we demonstrated that HH rapidly induces Caveolin-1-dependent transcytosis of IgG across endothelial cells as an early event, prece

Neuroinflammation, a central pathological mechanism in central nervous system disorders, remains challenging to treat due to the restrictive nature of the blood-brain barrier and poor drug delivery efficiency. In this study, we developed a novel intranasal brain-targeted delivery system by constructing peptide-modified quercetin liposomes (QuR-lipo). By using intranasal administration to bypass the blood-brain barrier, QuR-lipo demonstrated higher uptake and accumulation in the brain compared to unmodified liposomes. This system disrupts the vicious cycle between neuroinflammation and oxidativ

Six previously undescribed alkaloids, including four C19-diterpenoid alkaloids, aemulanines A-D, and two C18-diterpenoid alkaloids, aemulanines E-F, together with five known analogues, were isolated from the whole plant of Delphinium aemulans Nevski (Ranunculaceae). Their structures were established by comprehensive spectroscopic analysis, and the absolute configuration of aemulanine A was confirmed by single-crystal X-ray diffraction. Aemulanines A-C feature a rare C19-diterpenoid alkaloid skeleton with an opened B ring and an unusually rearranged D ring, while aemulanine E represents the fir

MXene quantum dots (MQDs) have recently emerged as a distinct zero-dimensional derivative of MXenes, exhibiting unique physicochemical and electronic properties that extend their relevance beyond conventional nanomaterial applications. Owing to their ultrasmall size, rich surface termination chemistry, and transition metal-centered electronic structure, MQDs interact dynamically with biological systems and influence immune behavior in a context-dependent manner. This review provides the first comprehensive and focused overview of MQDs as immunomodulatory nanoplatforms for inflammatory control

Subacute ruminal acidosis, induced by high-concentrate diets, can trigger systemic inflammation and mastitis, a pathology linked to ruminal dysbiosis and disruption of the host's circadian rhythm. In this study, 32 mid-lactating Saanen goats (41.5 ± 1.79 kg BW) were randomly allocated to 4 treatment groups (n = 8/group): a low-concentrate diet (LC; 30:70 concentrate:forage ratio), a high-concentrate diet (HC; 70:30), and their respective counterparts supplemented with 200 mg/kg of DM thiamine (LCT and HCT). Results demonstrated that the HC diet decreased ruminal pH, induced microbial dysbiosis

BACKGROUND: Ginsenoside Rg1 (GRg1), a principal neuroactive constituent of ginseng, has demonstrated promising antidepressant potential. Beyond its well-documented roles in mitigating oxidative stress, suppressing neuroinflammation, and protecting mitochondrial function, our study further demonstrates that GRg1 exerts its protective effects on the organism by upregulating the expression of connexin 43 (Cx43) in astrocytes. However, it remains unknown whether Cx43 mediates the antidepressant effects of GRg1 by regulating astrocyte pyroptosis.
OBJECTIVE: This study aimed to elucidate the role o

Celiac disease (CD) is a chronic immune-mediated enteropathy triggered by the ingestion of gluten in genetically predisposed individuals. While the adaptive immune response to deamidated gliadin peptides represents a central pathogenic mechanism, growing evidence suggests that epithelial stress and innate immune activation play a fundamental role in the onset and persistence of the disease. Heat shock proteins (Hsps), central regulators of cellular proteostasis, have emerged as potential mediators at the interface between epithelial distress and immune signaling. This review discusses the invo

PURPOSE OF REVIEW: This review examines differences between three large colchicine trials, including study design, trial populations, timing of treatment initiation, colchicine dosing regimens, geographic factors, the timing of trial conduct and inflammatory response.
RECENT FINDINGS: Colchicine inhibits neutrophil function and affects nucleotide-binding oligomerisation domain-like receptors, pyrin domain-containing 3 (NLRP3) inflammasome activity, thereby reducing the release of downstream pro-inflammatory cytokines such as mature interleukin-1β and interleukin-6. Large randomized controlled

Acute ischemic stroke (AIS), a primary cause of global mortality and disability, involves the sudden interruption of cerebral blood flow, leading to significant neurological impairment. Inflammation plays a critical role in exacerbating brain damage following stroke, making effective anti-inflammatory treatments essential. This study explored the therapeutic potential of anisodine hydrobromide (Ani), a traditional Chinese medicine, in modulating inflammatory responses and glycolysis in the lymphocytes of AIS patients. We evaluated the impact of Ani on peripheral blood mononuclear cells (PBMCs)

INTRODUCTION: Autoinflammatory diseases are inherited disorders of innate immunity, broadly classified into inflammasomopathies, interferonopathies, and complement-mediated disorders. They are characterized by dysregulated cytokine signaling - particularly IL-1, IL-6, TNF, type I interferon, and the JAK-STAT pathway-and are increasingly managed with molecular targeted therapies. Prototypical entities include familial Mediterranean fever (FMF), TNF receptor - associated periodic syndrome (TRAPS), and cryopyrin-associated periodic syndromes (CAPS). Other conditions - notably mevalonate kinase de

Salvia balansae, an endemic Algerian species, remains poorly investigated despite the recognized medicinal importance of the Salvia genus. This study comprehensively evaluated its phytochemical composition, biological activities, and safety profile. Quantitative analyses revealed high levels of total phenolics, flavonoids, flavonols, condensed tannins, and triterpenoids, while LC-MS identified a distinctive Aurès-region chemotype enriched in luteolin, rosmarinic acid, and flavonoid glycosides. The extract demonstrated strong antioxidant activity in free radical-scavenging, metal-chelating, and

Despite its widespread application as a triazine herbicide, atrazine (ATR) poses significant health threats, with the mechanisms underlying its induction of hepatic inflammation still not fully elucidated. Our research delineates a mechanistic cascade in which ATR exposure is associated with endoplasmic reticulum (ER) stress induction, which in turn stimulates the excessive generation of mitochondria-associated membranes (MAMs). This event is associated with mitochondrial calcium inundation, augmented generation of mitochondrial reactive oxygen species (mtROS), and the cytosolic translocation

Alzheimer's Disease (AD) is now understood as a biologically diverse condition, with amyloid and tau pathology evolving within dynamic neuroimmune networks. This challenges the traditional view that AD-related inflammation can be broadly suppressed therapeutically. We review evidence showing that neuroinflammation in AD is stage-dependent, cell-state-specific, spatially organized, and functionally complex. Microglia and astrocytes can aid in plaque containment, debris clearance, synaptic balance, metabolic adaptation, and tissue repair, but may also exacerbate injury through type-I interferon,
