Dengue virus at the landscape scale in Cambodia: Linking climate and land use dynamics with Dengue ecology in Southeast Asia.

Dengue remains a major public health challenge in Cambodia, where transmission dynamics are driven by interactions among human infections, Aedes vector populations, land use and climatic variability. Hence, longitudinal evidence linking laboratory-confirmed dengue infections with entomological and environmental indicators remains limited. This study aimed to investigate the dengue incidence and Aedes vector populations in relation to climate and land use dynamics. Adult Aedes mosquitoes have been collected across six major land-use types using BG-Sentinel traps every 2 months during 4 years. I

Chikungunya outbreak in eastern Senegal, 2023.

BACKGROUND: Chikungunya virus (CHIKV) is a mosquito-borne Alphavirus that has caused several epidemics around the world in recent years. CHIKV is endemic in eastern Senegal, particularly in Kédougou, where epidemiological and virological surveillance is implemented in combination with the 4S network. In August 2023, an outbreak was reported in Kédougou, which rapidly spread to Tambacounda. Epidemiological and virological investigations were carried out in both regions by teams from the Institut Pasteur de Dakar, the WHO, the Africa CDC and the Senegalese Ministry of Health. METHODS: The team f

Hemozoin: a waste product after heme detoxification?

BACKGROUND: Hemozoin is considered a waste byproduct of heme detoxification following hemoglobin digestion; consequently, the biological functions of hemozoin in hemozoin-producing organisms have often been overlooked. However, recent findings indicate that Schistosoma hemozoin facilitates the transfer of iron from erythrocytes to eggs through its formation and degradation, thereby increasing interest in the role of malarial hemozoin. METHODS: Using transmission electron microscopy, we compared the formation of Schistosoma hemozoin and malaria hemozoin. Through transcriptome analysis of diffe

Charting the Proteins of Oropouche Virus.

Oropouche virus (OROV) is an emerging arbovirus responsible for Oropouche fever, also known as sloth fever, a febrile illness that can lead to recurrent outbreaks in affected regions. Endemic to parts of South and Central America, OROV is primarily transmitted by biting midges (Culicoides paraensis), although mounting evidence implicates mosquitoes, particularly the Culex and Aedes species, as additional vectors. Recent ecological disturbances-such as deforestation, urbanization, and climate change-have driven significant shifts in vector population dynamics, contributing to the expanded geogr

Seroprevalence study for selected zoonotic vector-borne pathogens in sheep from endemic areas of Croatia.

Surveillance is crucial in controlling and preventing vector-borne zoonotic diseases (VBDs). We analyzed the seroprevalence of selected vector-borne zoonotic pathogens in sheep from endemic areas and their role as possible sentinels for VBDs. A total of 300 sheep from seven farms at three micro-locations were tested for the presence of IgG antibodies against tick-borne encephalitis virus (TBEV), West Nile virus (WNV), Usutu virus (USUV), Borrelia burgdorferi s.l., and Crimean-Congo hemorrhagic fever virus (CCHFV) using ELISA with confirmation of borderline/positive results by VNT. Seropositivi

Comparative vector competence of post-2015 St. Louis encephalitis virus in Culex tarsalis and Culex quinquefasciatus mosquitoes.

The human pathogenic orthoflavivirus St. Louis encephalitis virus (SLEV) reemerged in the western United States in 2015 after more than a decade of apparent absence and has since expanded throughout California with sustained, interannual transmission. This shift from the historically sporadic pattern of SLEV activity prior to 2003 raises the question of whether contemporary SLEV strains differ in fitness in Culex vectors compared with earlier strains. To determine whether reemerging SLEV possess augmented infectivity and transmissibility that may have facilitated reestablishment, we compared t

Recent Advances in the Role of Different Nanoparticles in the Various Biosensors for the Detection of the Chikungunya Virus.

Humans contract the Chikungunya virus (CHIKV), an alphavirus transmitted by mosquitoes that induces acute and chronic musculoskeletal discomfort and fever. Millions of cases of the disease have been attributed to CHIKV in the Indian Ocean region since 2004, and the virus has since spread to Europe, the Middle East, and the Pacific. The exponential proliferation of CHIKV in recent times underscores the critical nature of implementing preventative measures and exploring potential control strategies. The principal laboratory test employed to diagnose infection in serum samples collected over six

Malaria is a parasitic infection caused by bites from Plasmodium falciparum (P. falciparum)-infected mosquitoes with a present scale of symptoms ranging from moderate fever to neurological disorders. P. falciparum is the most lethal of the five strains of malaria, and is a major case of morbidity and mortality in endemic regions. Recent advancements in malaria diagnostic tools and prevention strategies have improved conjugation antimalarial therapies using fumigation and long-lasting insecticidal sprays, thus lowering malarial infections. Declines in the total number of infected individuals ha

Functional Characterization of the m6A-Dependent Translational Modulator PfYTH.2 in the Human Malaria Parasite.

Posttranscriptional regulation of gene expression is central to the development and replication of the malaria parasite, Plasmodium falciparum, within its human host. The timely coordination of RNA maturation, homeostasis, and protein synthesis relies on the recruitment of specific RNA-binding proteins to their cognate target mRNAs. One possible mediator of such mRNA-protein interactions is the N6-methylation of adenosines (m6A), a prevalent mRNA modification of parasite mRNA transcripts. Here, we used RNA protein pulldowns, RNA modification mass spectrometry, and quantitative proteomics to id

IMRAS-Immunization with radiation-attenuated Plasmodium falciparum sporozoites by mosquito bite: Cellular immunity to sporozoites, CSP, AMA1, TRAP and CelTOS.

BACKGROUND: Immunization with radiation-attenuated sporozoites (RAS) by mosquito bites provides >90% sterile protection against Plasmodium falciparum malaria in humans. We conducted a clinical trial based on data from previous RAS clinical trials that suggested that 800-1200 infected bites should induce ~50% protective vaccine efficacy (VE) against controlled human malaria infection (CHMI) administered three weeks after the final immunization. Two cohorts were immunized separately. VE was 55% in Cohort 1 but 90% in Cohort 2, the cohort that received a higher first dose and a reduced (fractiona

Meta-analysis of the use of sterilized mosquito net mesh for inguinal hernia repair in less economically developed countries.

BACKGROUND: Inguinal hernias are common in less economically developed countries (LEDCs), and associated with significant morbidity and mortality. Tension-free mesh repair is the standard treatment worldwide. Lack of resources combined with the high cost of commercial synthetic mesh (CSM) have limited its use in LEDCs. Sterilized mosquito net mesh (MNM) has emerged as a low-cost, readily available alternative to CSM. The aim of this systematic review and meta-analysis was to evaluate the safety and efficacy of MNM for the use in hernia repair in LEDCs. METHODS: A systematic review and data me

Zika Virus Proteins NS2A and NS4A Are Major Antagonists that Reduce IFN-β Promoter Activity Induced by the MDA5/RIG-I Signaling Pathway.

Zika virus (ZIKV) is a mosquito-transmitted, emerging Flavivirus that causes Guillain-Barré syndrome and microcephaly in adults and fetuses, respectively. Since ZIKV was first isolated in 1947, severe outbreaks have occurred at various places worldwide, including Yap Island in 2007, French Polynesia in 2013, and Brazil in 2015. Although incidences of ZIKV infection and dissemination have drastically increased, the mechanisms underlying the pathogenesis of ZIKV have not been sufficiently studied. In addition, despite extensive research, the exact roles of individual ZIKV genes in the viral evas

From research to phase III: preclinical, industrial and clinical development of the Sanofi Pasteur tetravalent dengue vaccine.

Dengue vaccine development has reached a major milestone with the initiation, in 2010, of the first phase III clinical trial to investigate the Sanofi Pasteur CYD tetravalent dengue vaccine (TDV). The CYD TDV candidate is composed of four recombinant, live, attenuated vaccines (CYD-1-4) based on a yellow fever vaccine 17D (YFV 17D) backbone, each expressing the pre-membrane and envelope genes of one of the four dengue virus serotypes. The vaccine is genetically and phenotypically stable, non-hepatotropic, less neurovirulent than YFV 17D, and does not infect mosquitoes by the oral route. In vit
