Sleep Deprivation and Neuronal Hyperexcitation Share Transcriptomic Signatures.

Although sleep deprivation (SD) is clinically associated with numerous neuropsychiatric disorders, its underlying molecular correlates remain unclear. Because extended wakefulness is accompanied by increased neuronal activity and network firing, SD may be associated with a hyperactive neural state. This study aimed to test the hypothesis that SD shares transcriptomic signatures induced by neuronal hyperexcitation and to identify the gene pathways and cell types associated with these signatures. Publicly available transcriptomic datasets were analyzed, including 32 SD and 23 neuronal hyperexcit

Sex-Stratified Machine Learning for the Prediction of Post-COVID Condition: A Longitudinal Cohort Study.

Background: Post-COVID-19 condition (PCC) affects many survivors, with evidence of sex-specific differences in prevalence and symptom profiles. However, few prediction studies have examined whether sex-stratified models improve prediction or generalize across sexes. This study aimed primarily to develop and compare sex-stratified machine learning models for PCC prediction using routinely available baseline variables, and secondarily to assess cross-sex generalizability and adversarial robustness. Methods: We analyzed a prospective longitudinal cohort of 1006 adults hospitalized with COVID-19 a

Integration of Systemic Immune-Inflammation Index and Lactate for diagnostic and early risk stratification in pediatric sepsis.

BACKGROUND: Pediatric sepsis remains a leading cause of global mortality, necessitating early and accurate risk stratification. Traditional scoring systems often fail to capture the rapid, non-linear progression of the disease involving both immune dysregulation and metabolic derangement. This study aimed to develop and validate an interpretable machine learning (ML) prognostic model by integrating novel inflammatory indices, specifically the Systemic Immune-Inflammation Index (SII) and Lactate to predict 28-day mortality in children with sepsis. METHODS: A single-center, retrospective cohort

Transdiagnostic divergent neural development of reward anticipation in early adolescents at risk for depression and psychosis.

BACKGROUND: Reward anticipation is a transdiagnostic factor relevant to both depression and psychosis. Adolescence is a critical neurodevelopmental period when these reward-related circuits-including frontostriatal, limbic/insular, and striatal circuits may contribute to shared vulnerability across disorders, yet longitudinal transdiagnostic comparisons in early adolescents remain scarce. METHODS: Longitudinal functional magnetic resonance imaging (fMRI) data acquired during the monetary incentive delay (MID) task were analyzed in 1839 adolescents from the Adolescent Brain Cognitive Developme

Enhanced detection of subtle cortical abnormalities in focal epilepsy using 7 T MRI surface-based models and graph neural networks.

PURPOSE: MRI detection of subtle focal cortical dysplasia (FCD)-like abnormalities remains challenging in focal epilepsy. Higher signal-to-noise ratio and spatial resolution offered by ultra-high-field 7T MRI and surface-based graph-neural-network (GNN) analysis may improve detection of subtle cortical abnormalities. We evaluated whether combining 7T MRI with a surface-based GNN classifier improves lesion detection in focal epilepsy of suspected structural origin. METHODS: We analyzed paired 7T and 3T MRI datasets from 87 patients with focal epilepsy (78.1% pediatric) and 10 internal healthy

Deep convolutional autoencoders for the spectral reconstruction of filter-interfered FTIR spectra in microplastic assessment.

The widespread contamination of aquatic environments by microplastics demands robust monitoring strategies, with Fourier transform infrared spectroscopy serving as a widely adopted technique for particle identification. However, the reliability of acquired spectra is frequently compromised by interference from filter substrates, especially when analyzing particles of very small sizes. This interference requires the reconstruction of high-fidelity spectra from distorted spectra to maintain the analytical integrity of environmental measurements. While traditional shallow convolutional autoencode

Machine learning-based screening of sarcopenia in elderly women using non-invasive physiological signals.

BACKGROUND: Sarcopenia is the age related loss of skeletal muscle mass and strength, which increases frailty, fall risk, and healthcare costs. Conventional imaging based diagnostics are costly and impractical in routine clinical or community settings. We investigated whether noninvasive physiological signals, namely electrocardiography (ECG), electromyography (EMG), and center of pressure (COP), can serve as biomarkers for sarcopenia when analyzed with machine learning models. METHODS: In this study, 80 Korean women aged 65 to 75 years were assessed for sarcopenia using the 2019 Asian Working

The common pathological network of inflammation, extracellular matrix imbalance, and senescence in intervertebral disc degeneration and osteoarthritis.

As representatives of degenerative orthopedic diseases, intervertebral disc degenerative disease (IVDD) and osteoarthritis (OA) involve the spine and peripheral articular cartilage, respectively. Their comorbidity rate in individuals over 60 years exceeds 40%, suggesting overlapping pathological and physiological features. Age-related "inflammaging" and cellular senescence, obesity-mediated mechanical load and metabolic disorders, and genetic/epigenetic abnormalities (e.g., COL2A1, ADAMTS5) constitute a shared risk factor network. Extracellular matrix (ECM) imbalance is a core initiating event

Radiomics and machine learning for characterizing CKD-associated cortical bone texture patterns in HR-pQCT tibia scans: a slice- and patient-level methodological framework.

Chronic kidney disease (CKD) is associated with alterations in cortical bone structure and composition that contribute to increased fracture risk but are incompletely captured by standard clinical imaging, including DXA. This study evaluated whether radiomics combined with machine learning can enhance detection of CKD-related cortical bone characteristics using high-resolution peripheral quantitative computed tomography (HR-pQCT). HR-pQCT images (60.7 μm isotropic resolution; 168-slice stacks acquired at 7.3% and 30% proximal to the tibial endplate) were analyzed from 72 participants (38 non-C

Association between chronic stress-related amygdala metabolic activity and distant metastasis in colorectal cancer.

BACKGROUND: Chronic stress has been implicated in cancer progression through neuroendocrine and inflammatory pathways, but its role in colorectal cancer (CRC) remains uncertain. The amygdala, a key stress-responsive brain structure, demonstrates measurable metabolic activity on 18F-fluorodeoxyglucose positron emission tomography/computed tomography (18F-FDG PET/CT) and may serve as a surrogate imaging biomarker of chronic stress. This study aimed to investigate whether elevated amygdala metabolic activity (AmygA) is associated with distant metastasis in patients with CRC. PATIENTS AND METHODS
