ESRRA-ATG5-Mediated mitophagy enhances arginine metabolism to alleviate diabetic kidney disease.

Diabetic kidney disease (DKD) is increasingly recognized as a consequence of impaired mitochondrial quality control in renal tubular epithelial cells (TECs). In this study we show that the nuclear receptor ESRRA (estrogen related receptor alpha) transcriptionally activates ATG5 (autophagy related 5) to sustain PINK1 (PTEN induced kinase 1)-dependent mitophagy and preserve tubular homeostasis. ESRRA and ATG5 expression were markedly reduced in human DKD biopsies, and their abundance correlated positively with estimated glomerular filtration rate and inversely with albuminuria. Conditional delet

MCL-1 and BCL-xL-dependent resistance to the BCL-2 inhibitor ABT-199 can be overcome by preventing PI3K/AKT/mTOR activation in lymphoid malignancies.

Overexpression of anti-apoptotic BCL-2 family members is a hallmark of many lymphoid malignancies, including chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma (NHL) that can be targeted with small molecule inhibitors. ABT-199 is a rationally designed BCL-2 homology (BH)-3 mimetic that specifically binds to BCL-2, but not to MCL-1 and BCL-xL. Although the thrombocytopenia that occurs with navitoclax treatment has not been a problem with ABT-199, clinical trials in CLL could benefit by lowering the ABT-199 concentration through targeting other survival pathways. In this study, we inves

A novel regulatory mechanism of Pim-3 kinase stability and its involvement in pancreatic cancer progression.

UNLABELLED: Translationally controlled tumor protein (TCTP/TPT1) was identified from a yeast 2-hybrid screen and shown to interact with Pim-3, a member of the proto-oncogene Pim family with serine/threonine kinase activity. TCTP was aberrantly expressed in human pancreatic cancer cells and malignant ductal epithelial cells, but not in normal pancreatic duct epithelial cells adjacent to tumor foci of human pancreatic cancer tissue. Moreover, TCTP colocalized with Pim-3 both in human pancreatic cancer cells and in clinical tissues. Mapping studies revealed that the interaction between Pim-3 and

Role of cAMP-phosphodiesterase 1C signaling in regulating growth factor receptor stability, vascular smooth muscle cell growth, migration, and neointimal hyperplasia.

RATIONALE: Neointimal hyperplasia characterized by abnormal accumulation of vascular smooth muscle cells (SMCs) is a hallmark of occlusive disorders such as atherosclerosis, postangioplasty restenosis, vein graft stenosis, and allograft vasculopathy. Cyclic nucleotides are vital in SMC proliferation and migration, which are regulated by cyclic nucleotide phosphodiesterases (PDEs). OBJECTIVE: Our goal is to understand the regulation and function of PDEs in SMC pathogenesis of vascular diseases. METHODS AND RESULTS: We performed screening for genes differentially expressed in normal contractil

The E3 ubiquitin ligase NEDD4 protects against nonesterified fatty acid-induced hepatic inflammatory injury in ketotic cows.

Cows with ketosis frequently exhibit liver injury, primarily driven by increased influx of nonesterified fatty acids (NEFA) into the liver. This excess NEFA induces lipotoxicity, which triggers hepatic inflammatory responses and ultimately results in liver damage, further exacerbating ketosis. Neural precursor cell expressed developmentally downregulated 4 (NEDD4), an E3 ubiquitin ligase, plays a crucial role in regulating anti-inflammatory processes, yet its role in the hepatic injury in ketotic cows remains unclear. This study aimed to investigate the link between NEDD4 and hepatic inflammat

MARVELD1 regulates hepatocyte transformation via maintaining NF-κB signaling homeostasis mediated by the CCL5/CCR5 axis between hepatic stellate cells and hepatocytes.

BACKGROUND: Hepatocellular carcinoma (HCC) involves abnormal alterations in cellular homeostasis regulation and molecular crosstalk between hepatic stellate cells (HSCs) and hepatocytes, and the underlying mechanisms are highly complex. Recent evidence suggests that HSCs exhibit obvious plasticity, are activated by stress stimuli such as carcinogens, and participate in HCC occurrence and progression through crosstalk with hepatocytes. MARVELD1 is a newly reported regulator of genomic stability closely associated with malignant tumors. However, the relationship between MARVELD1-mediated hepatoc

Preventive effects and mechanisms of yam exosome-like nanoparticles on acute liver injury.

Acute liver injury (ALI) represented a significant clinical challenge due to its high mortality rate and the absence of specific therapeutic interventions. Plant-derived exosome-like nanoparticles (ELNs) had recently garnered increasing attention in the field of disease prevention and treatment, owing to their favorable biocompatibility and potential for targeted delivery. In this study, we isolated and systematically characterized ELNs from the medicinal plant yam (Yam ELNs). Structural analysis revealed that Yam ELNs possess a homogeneous bilayer membrane structure with an average particle s

Engineering overexpressing SYNGR1 inhibited the progression of GBM cells by suppressing the intracellular FGF1-mediated LDs accumulation and cytoskeleton remodeling.

BACKGROUND: Glioblastoma (GBM) is the most common and aggressive glioma subtype in adults, characterized by disrupted lipid homeostasis. The accumulation of lipid droplets (LDs) is involved in the actin cytoskeleton remodeling in different types of cells, the intracellular fibroblast growth factor 1 (FGF1) plays an important role in this process. The stability of cytoskeleton is crucial for the proliferation, invasion and other malignant behavior of GBM. The synaptic gyrus protein 1 (synaptogyrin 1, SYNGR1) is closely correlated with the progression of various tumours, but there have been no r

Assessment of the vaginal microbiota before and after use of hyperosmolal lubricant during transvaginal ultrasound.

BACKGROUND: Vaginal lubricants are commonly used during sexual activity and clinical procedures such as transvaginal ultrasound. Epidemiologic and laboratory studies indicate hyperosmolal water-based lubricants may disrupt the vaginal microbiota, particularly the beneficial Lactobacillus spp. These bacteria play a critical role in protecting against sexually transmitted infection acquisition and other adverse gynecologic and obstetric outcomes. OBJECTIVE: We sought to evaluate changes in the composition of the vaginal microbiota before and after a single exposure to lubricant over a 10-week p

Effect of folic acid-targeted triptolide-loaded microbubbles combined with ultrasound on apoptosis of human MCF-7 breast cancer xenografts in nude mice.

BACKGROUND: Breast cancer is a leading cause of mortality among women, with its incidence and mortality rates steadily rising year after year. Despite the common use of chemotherapy and radiotherapy in clinical treatment, the prognosis often remains unsatisfactory. The majority of patients face the imminent risk of tumor recurrence and metastasis, resulting in a persistently low 5-year survival rate. This study aims to develop and evaluate a novel folic acid-targeted triptolide-loaded ultrasound microbubble (FA-TLUM) system for the targeted treatment of breast cancer. METHODS: FA-TLUM were pr

Low IgE and absence of sensitization in non-T2 asthma: a transcriptomic and cytokine study.

BACKGROUND: Asthma exhibits heterogeneity, including type 2 (T2-high) and non-T2 phenotypes. This study aimed to elucidate the inflammatory mechanisms that drive non-type 2 asthma, a subtype characterized by low immunoglobulin E levels and negative allergic sensitization. This context considers the complex processes of allergic sensitization. METHODS: We performed gene expression analysis in non-T2 (n = 11) versus T2-high (n = 17) pediatric patients using public datasets GSE145505 for comparison (non-atopic and high-atopic datasets). We applied Ingenuity Pathway Analysis (IPA) to identify can

Time-dependent gene expression patterns in epidermal wound healing: Mechanistic insights into Cayratia japonica hydrogel intervention after anal fistula surgery.

ETHNOPHARMACOLOGICAL RELEVANCE: Postoperative wound healing, especially after anal-fistula surgery, poses unique clinical challenges due to the complex anal anatomy and the need for accelerated recovery. Although the ethnomedicinal plant Cayratia japonica (Thunb.) Gagnep. has long been recognized for its wound-healing properties, its molecular basis and therapeutic potential remain poorly defined. Here, an optimized C. japonica hydrogel (CJH) markedly improved clinical outcomes, while temporal multi-omics profiling underscored the necessity of stage-specific therapeutic adaptation. Long-term, induced expression of Hand2 in peripheral sympathetic neurons ameliorates sarcopenia in geriatric mice.

BACKGROUND: The discovery of adrenoceptors, which mediate the effects of the sympathetic nervous system neurotransmitter norepinephrine on specific tissues, sparked the development of sympathomimetics that have profound influence on skeletal muscle mass. However, chronic administration has serious side effects that preclude their use for muscle-wasting conditions such as sarcopenia, the age-dependent decline in muscle mass, force, and power. Devising interventions that can adjust neurotransmitter release to changing physiological demands will require understanding how the sympathetic nervous s

Coronavirus nsp10/nsp16 Methyltransferase Can Be Targeted by nsp10-Derived Peptide In Vitro and In Vivo To Reduce Replication and Pathogenesis.

UNLABELLED: The 5' cap structures of eukaryotic mRNAs are important for RNA stability and protein translation. Many viruses that replicate in the cytoplasm of eukaryotes have evolved 2'-O-methyltransferases (2'-O-MTase) to autonomously modify their mRNAs and carry a cap-1 structure (m7GpppNm) at the 5' end, thereby facilitating viral replication and escaping innate immune recognition in host cells. Previous studies showed that the 2'-O-MTase activity of severe acute respiratory syndrome coronavirus (SARS-CoV) nonstructural protein 16 (nsp16) needs to be activated by nsp10, whereas nsp16 of fel…

MAPK-activated protein kinase-2 in cardiac hypertrophy and cyclooxygenase-2 regulation in heart.

RATIONALE: Activation of p38 mitogen-activated protein kinase (MAPK) has a significant impact on cardiac gene expression, contractility, extracellular matrix remodeling, and inflammatory response in heart. The p38 kinase pathway also has a controversial role in cardiac hypertrophy. MAPK-activated protein kinase-2 (MK2) is a well-established p38 downstream kinase, yet its contribution to p38-mediated pathological response in heart has not been investigated. OBJECTIVE: We examined the specific contribution of MK2 to the pathological remodeling induced by p38. METHODS AND RESULTS: We used a car

Identification and experimental validation of KMO as a critical immune-associated mitochondrial gene in unstable atherosclerotic plaque.

BACKGROUND: The heightened risk of cardiovascular and cerebrovascular events is associated with the increased instability of atherosclerotic plaques. However, the lack of effective diagnostic biomarkers has impeded the assessment of plaque instability currently. This study was aimed to investigate and identify hub genes associated with unstable plaques through the integration of various bioinformatics tools, providing novel insights into the detection and treatment of this condition. METHODS: Weighted Gene Co-expression Network Analysis (WGCNA) combined with two machine learning methods were

Disrupted hypothalamic transcriptomics and proteomics in a mouse model of type 2 diabetes exposed to recurrent hypoglycaemia.

AIMS/HYPOTHESIS: Repeated exposures to insulin-induced hypoglycaemia in people with diabetes progressively impairs the counterregulatory response (CRR) that restores normoglycaemia. This defect is characterised by reduced secretion of glucagon and other counterregulatory hormones. Evidence indicates that glucose-responsive neurons located in the hypothalamus orchestrate the CRR. Here, we aimed to identify the changes in hypothalamic gene and protein expression that underlie impaired CRR in a mouse model of defective CRR. METHODS: High-fat-diet fed and low-dose streptozocin-treated C57BL/6N mi
