CpG DNA is a potent enhancer of systemic and mucosal immune responses against hepatitis B surface antigen with intranasal administration to mice.

Mucosal immunity is difficult to induce with subunit vaccines unless such vaccines are administered with a mucosal adjuvant such as cholera toxin (CT); however, CT is toxic in humans. Synthetic oligodeoxynucleotides containing immunostimulatory CpG motifs (CpG) are potent adjuvants for the induction of Th1-like systemic immune responses against parenterally delivered proteins. Here, we show in mice that intranasal delivery of hepatitis B surface Ag, which alone has no effect, elicits good immune responses when given with CpG oligodeoxynucleotides and/or CT. Overall, CpG is superior to CT for t

Autoimmune premature ovarian failure: of mice and women.

Autoimmunity is a well-established mechanism of premature ovarian failure and may be the dominant cause of reversible premature ovarian failure. Because of its strong analogy with the human disease, murine experimental post-thymectomy autoimmune oophoritis may provide insight into the pathogenesis of autoimmune premature ovarian failure in women and might open new avenues to specific diagnostic and therapeutic methods. We reviewed the literature on murine experimental post-thymectomy autoimmune oophoritis in order to compare and contrast it with human autoimmune premature ovarian failure, to w

Treatment with anti-IL-2 antibodies reduces hepatic pathology and eosinophilia in Schistosoma mansoni-infected mice while selectively inhibiting T cell IL-5 production.

Granulomas around Schistosoma mansoni eggs are a principal cause of morbidity in mice infected with this helminth. In vivo treatment of infected mice with anti-IL-2 antibodies, with or without anti-IL-2 receptor antibodies, significantly diminished the size of circumoval granulomas in the liver and decreased hepatic fibrosis to half that in untreated mice. Antibody-treated animals also displayed a marked reduction in both peripheral blood and tissue eosinophilia while IgE levels were unchanged or increased. Spleen cell cytokine production in response to Ag or mitogen stimulation was selectivel

SENP1 facilitates the adaptation of colonic non-lymphoid tissue Treg cells and restrains intestinal inflammation.

Colonic immune homeostasis is critically maintained by regulatory T (Treg) cells. Here, we identify SUMO-specific peptidase 1 (SENP1) as an important regulator of colonic Treg function and intestinal immune homeostasis. Treg-specific Senp1 deletion does not impair thymic Treg development, but selectively disrupts the homeostasis of colonic Treg subset without affecting Treg maintenance in other peripheral tissues. Mechanistically, SENP1 deficiency reduces CD25 expression on Tregs, blunting IL-2 responsiveness and compromising their expansion and survival. This is associated with a selective re

Naringin alleviates autoimmune hepatitis in mice via the gut-liver Axis through modulation of microbiota, metabolites, and immune responses.

Autoimmune hepatitis (AIH) is an immune-mediated liver disease that could be impacted by gut microbiota dysbiosis. Naringin, a flavonoid der … ntestinal barrier integrity, and decreased Th1/Th17 cell proportions without significantly altering Th2 cells, as validated by flow cytometry and immunohistochemistry. Importantly, FMT from naringin-treated donors provided hepatoprotective benefits in recipient mice. These findings shed fresh information on the gut-liver axis in AIH, highlighting naringin a …

A novel multi-epitope vaccine induces protective and therapeutic immunity against Helicobacter pylori.

The development of an effective vaccine against Helicobacter pylori remains a major global health priority, aimed at reducing infection prevalence and preventing diseases such as chronic gastritis, peptic ulcers, and gastric cancer. Despite extensive research, no vaccine has yet demonstrated durable efficacy in clinical settings. This study describes the development of a novel multi-epitope vaccine targeting H. pylori. Sixteen B- and T-cell epitopes derived from six virulence factors were identified using bioinformatics tools and assembled into a Multi-Epitope Unit (MEU) antigen. The MEU antig

Moxibustion combined with anti-PD-1 antibodies improves immunosuppression in septic mice potentially through the PD-1/PD-L1 pathway.

This study was to investigate the therapeutic effects of moxibustion (Mox) combined with anti-PD-1 antibody on septic mice and the immune regulatory mechanism based on the PD-1/PD-L1 axis. A CLP-induced sepsis mouse model was established and intervened with moxibustion and/or anti-PD-1 antibody. It was found that both Mox and anti-PD-1 monotherapy improved mouse survival rates, reduced murine sepsis score, alleviated pathological damage in the liver, lungs, and spleen, mitigated serum biochemical abnormalities, and regulated systemic and local organ inflammation. Combination therapy demonstrat

A Bispecific Antibody Blocking Both TSLP and IL-4Rα for the Treatment of Allergic Inflammatory Diseases.

Thymic stromal lymphopoietin (TSLP) works synergistically with Th2 cytokines to regulate infection, inflammation, and metabolic homeostasis. However, their aberrant activities lead to the onset and sustaining of many types of allergic inflammatory diseases. While biologics drug molecules blocking either TSLP or IL-4/IL-13 show clinical efficacies, the broader effect of simultaneously targeting these cytokines remains to be explored. We generated a bispecific antibody (BsAb) targeting both TSLP and IL-4Rα, which effectively blocked the signaling cascades driven by TSLP, IL-4, and IL-13. The BsA

CXCL16 deficiency alleviates ovalbumin-induced allergic rhinitis in mice.

PURPOSE: Research has indicated that individuals with allergic rhinitis exhibit elevated levels of CXCL16 expression in their serum. This study aims to illustrate the role of CXCL16 and its associated mechanisms in mice suffering from allergic rhinitis. METHODS: An allergic rhinitis model was established by injecting ovalbumin (OVA) into mice, and the expression of CXCL16 mRNA and protein in nasal mucosal tissue was measured. The frequency of nose rubbing and sneezing in each group of mice was recorded. Serum levels of IgE and IgG1 were also assessed. Th2 cell-related factors in the bronchoal

Bone marrow mesenchymal stem cell exosomes improve fracture union via remodeling metabolism in nonunion rat model.

BACKGROUND: Nonunion of fractures is a major unsolved problem in clinical treatment and prognosis of orthopedics. Bone marrow mesenchymal stem cell (BMSC) exosomes have been proven to be involved in mediating tissue and bone regeneration in a variety of diseases. However, the role of BMSC exosomes in fracture nonunion is unclear. METHODS: BMSC exosomes were injected into a rat model of nonunion fracture, and the fracture-healing site was detected by micro-CT and the serum metabolites were analyzed by LC-MS/MS. RESULTS: The results showed that the exosomes could be successfully isolated from

Late pregnancy is associated with an increase in regulatory cytokines and a decline in nematode-specific antibody levels in sheep infected with Teladorsagia circumcincta.

The gastrointestinal nematode Teladorsagia circumcincta is a prevalent and economically important parasite of sheep. Sheep develop acquired immunity to T. circumcincta, characterised by nematode-specific antibody production and a type 2 T-helper (Th) cell response. During late pregnancy and early lactation, ewes experience increased nematode faecal egg count (FEC) known as the peri-parturient rise (PPR). The PPR is associated with a decline in nematode-specific antibodies, but research on the role of Th cell-mediated immunity in the PPR is limited. Here, we characterised the cellular and humor

Polyopes affinis Suppressed IFN-γ- and TNF-α-Induced Inflammation in Human Keratinocytes via Down-Regulation of the NF-κB and STAT1 Pathways.

Polyopes affinis is a red algal species commonly found on the South coast and near Jeju Island, Korea. This study aimed to determine whether P. affinis extracts can inhibit the pathogenesis of T-helper-2 (Th2)-mediated inflammation in a human keratinocyte cell line of atopic dermatitis (AD). Cells were incubated with 10 ng/mL of interferon gamma (IFN-γ) and 10 ng/mL of tumor necrosis factor-alpha (TNF-α) at various concentrations of PAB (10, 30, and 60 µg/mL) and PAA (100, 500, and 1000 µg/mL) extracts. A gene-ontology (GO)-enrichment analysis revealed that PAB significantly enriched the genes

Immunomodulatory influence of HIV and EBV on Helicobacter pylori infections – a review.

Helicobacter pylori is a Gram-negative, microaerophilic rod colonizing the stomach mucosa. In most cases, the colonization of this organ is asymptomatic, while some people may develop diseases, including gastritis, peptic ulcers and gastric cancers. The infection caused by H. pylori is accompanied by the secretion of pro-inflammatory cytokines and the strong response of Th1/Th17 cells. Because this bacterium colonizes more than half of the human population, co-infections with other pathogens are a relatively common phenomenon. One of such etiological factors are viruses that have an immunomodu

Benralizumab: A New Approach for the Treatment of Severe Eosinophilic Asthma.

Eosinophilic asthma is the most common phenotype of severe asthma. It is characterized by abnormal production and release of type 2 cytokines from T helper type 2 (TH2) lymphocytes and type 2 innate lymphoid cells, such as IL-5. This leads to a persistent increase and activation of eosinophils in blood and the airways despite treatment with high-dose inhaled corticosteroids. Eosinophil differentiation, survival, and activation are preferentially regulated by IL-5, a cytokine that binds to the IL-5 receptor (IL-5R), which is located on the surface of eosinophils or basophils and plays a critica

Evaluation of the Protective Effect of Deoxyribonucleic Acid Vaccines Encoding Granule Antigen 2 and 5 Against Acute Toxoplasmosis in BALB/c Mice.

AbstractToxoplasma gondii infects a broad range of warm-blooded hosts, including humans. Important clinical manifestations include encephalitis in immunocompromised patients as well as miscarriage and fetal damage during early pregnancy. Toxoplasma gondii dense granule antigen 2 and 5 (GRA2 and GRA5) are essential for parasitophorous vacuole development of the parasite. To evaluate the potential of GRA2 and GRA5 as recombinant DNA vaccine candidates, these antigens were cloned into eukaryotic expression vector (pcDNA 3.1C) and evaluated in vaccination experiments. Recombinant DNA vaccines cons

Genes Critical for Developing Periodontitis: Lessons from Mouse Models.

Since the etiology of periodontitis in humans is not fully understood, genetic mouse models may pinpoint indispensable genes for optimal immunological protection of the periodontium against tissue destruction. This review describes the current knowledge of genes that are involved for a proper maintenance of a healthy periodontium in mice. Null mutations of genes required for leukocyte cell-cell recognition and extravasation (e.g., Icam-1, P-selectin, Beta2-integrin/Cd18), for pathogen recognition and killing (e.g., Tlr2, Tlr4, Lamp-2), immune modulatory molecules (e.g., Cxcr2, Ccr4, IL-10, Opg

Evolution of the Immune Response to Chronic Airway Colonization with Aspergillus fumigatus Hyphae.

Airway colonization by the mold Aspergillus fumigatus is common in patients with underlying lung disease and is associated with chronic airway inflammation. Studies probing the inflammatory response to colonization with A. fumigatus hyphae have been hampered by the lack of a model of chronic colonization in immunocompetent mice. By infecting mice intratracheally with conidia embedded in agar beads (Af beads), we have established an in vivo model to study the natural history of airway colonization with live A. fumigatus hyphae. Histopathological examination and galactomannan assay of lung homog
