Human immune memory to yellow fever and smallpox vaccination.

BACKGROUND: Establishment of immunological memory is a hallmark of adaptive immune responses and the biological mechanism for the success of vaccines. However, in humans, much of our knowledge about adaptive immune responses derives from studies of chronic viral infections. OBJECTIVE: Here, we summarize the work of our laboratory and others on B and T cell responses and the establishment and maintenance of immune memory after acute viral infections induced by vaccination with two of the most successful vaccines to date, the yellow fever and the smallpox vaccines.

Beyond Disease Categories: The Adaptive Hierarchical Domain Network (AHDN) for Precision Phenotyping and Personalized Management of Chronic Orofacial Pain.

Background/Objectives: Chronic orofacial pain disorders are highly heterogeneous and therapeutically challenging. Although current classifications such as the International Classification of Orofacial Pain (ICOP) and the concept of nociplastic pain have improved diagnostic consistency, patients with the same diagnosis may show substantial differences in symptom severity, underlying biology, and treatment response. This suggests that disease classifications may describe clinical phenotypes more effectively than the biological mechanisms sustaining persistent pain. This article proposes the Adap

Peripheral and Central Adaptations After a Median Nerve Neuromobilization Program Completed by Individuals With Carpal Tunnel Syndrome: An Exploratory Mechanistic Study Using Musculoskeletal Ultrasound Imaging and Transcranial Magnetic Stimulation.

OBJECTIVE: Neuromobilization exercises are increasingly advocated in the conservative management of individuals with carpal tunnel syndrome (CTS), as they may mitigate CTS-related signs and symptoms via potential peripheral (ie, musculoskeletal) and central (ie, neurophysiological) adaptations. However, the mechanisms underlying these adaptations have not been studied extensively. Hence, this exploratory and mechanistic study aims to evaluate the potential peripheral and central adaptations that may result in individuals with CTS who have completed a neuromobilization program. METHODS: Fourte

Multi-immersion oblique plane microscope (miOPM): a reconfigurable platform for high-resolution light-sheet fluorescence microscopy.

Light-sheet fluorescence microscopy (LSFM) enables gentle, rapid and efficient volumetric imaging of biological specimens. Despite its potential, LSFM has fragmented into multiple variants, each narrowly optimized for specific samples or imaging regimes, limiting its broad applicability. Here we introduce the multi-immersion oblique plane microscope (miOPM), an adaptable LSFM platform for high-resolution imaging ranging from subcellular dynamics to whole organisms and cleared tissues. miOPM supports seamless interchangeability of oil, water and air objectives and maintains diffraction-limited

Functional roles of Syk in macrophage-mediated inflammatory responses.

Inflammation is a series of complex biological responses to protect the host from pathogen invasion. Chronic inflammation is considered a major cause of diseases, such as various types of inflammatory/autoimmune diseases and cancers. Spleen tyrosine kinase (Syk) was initially found to be highly expressed in hematopoietic cells and has been known to play crucial roles in adaptive immune responses. However, recent studies have reported that Syk is also involved in other biological functions, especially in innate immune responses. Although Syk has been extensively studied in adaptive immune respo

From insect behavior to transferable robot locomotion: inferring embodied locomotor principles from limited data via adversarial inverse reinforcement learning.

Insect locomotion exhibits remarkable adaptability and flexibility despite the limited scale of its nervous system. However, the underlying principles that govern leg coordination remain difficult to extract and model computationally. Understanding how insects achieve stable and adaptive locomotion has long provided important inspiration for the development of control strategies in bio-inspired robotics. Nevertheless, many existing approaches rely on predefined coordination rules, manually tuned parameters, or hand-crafted reward functions, which limit the flexibility and transferability of th

Treatment of Ewing sarcoma family of tumors: current status and outlook for the future.

BACKGROUND: The Ewing sarcoma family of tumors (ESFT) comprises a group of well-characterized neoplasms with aggressive behavior. Despite significant progress with the use of intensive multiagent chemotherapy and local control measures, a significant proportion of patients die of disease progression. Chemotherapy dose intensification and autologous hematopoietic stem cell transplant (HSCT) have been explored by many institutions without obvious benefit in high-risk patients. Our current understanding in the biology and treatment of ESFT suggests that a more rational approach to the development

Tendon arthroplasty of the first metatarsophalangeal joint in hallux rigidus: preliminary communication.

A biological arthroplastic interposition is described for hallux rigidus, in which the rolled plantaris tendon is positioned at the base of the first phalanx. Eleven patients, for a total of 12 feet, underwent surgery using this technique. The average follow-up was 21 months. In all patients, pain and supination gait disappeared. The average metatarsal motion increased 44 degrees in dorsiflexion. The tendon packet seems to be a good biological spacer, because it adapts and models itself to the new joint function.

Recent Advances in the Synthesis of Acyclic Nucleosides and Their Therapeutic Applications.

DNA is commonly known as the "molecule of life" because it holds the genetic instructions for all living organisms on Earth. The utilization of modified nucleosides holds the potential to transform the management of a wide range of human illnesses. Modified nucleosides and their role directly led to the 2023 Nobel prize. Acyclic nucleosides, due to their distinctive physiochemical and biological characteristics, rank among the most adaptable modified nucleosides in the field of medicinal chemistry. Acyclic nucleosides are more resistant to chemical and biological deterioration, and their adapt

Early Biological Orchestration in Fracture Healing: Decoding the Crosstalk between Inflammation, Coagulation, and Metabolism.

Fracture healing is a complex process driven by endogenous regenerative mechanisms, with early biological responses playing a pivotal role in determining healing outcomes. During this critical phase, the body establishes a dynamic equilibrium across multiple systems, akin to the precise calibration of a biological clock. The inflammatory response is tightly regulated through the interplay of pro- and anti-inflammatory signals, ensuring efficient immune cell recruitment for necrotic tissue clearance while preventing excessive inflammation that could compromise surrounding tissues. ZeCardioAI: combining zebrafish, AI, and xAI for an in-depth cardiac phenotyping platform.

Artificial intelligence (AI) integrated with high-throughput assays offers a powerful route to accelerate discovery in relevant biological models. Functional cardiac imaging is a prime application, where deep learning (DL) and explainable AI (xAI) can overcome limitations of traditional phenotyping methods, such as manual analysis, subjective interpretation, and low scalability. In cardiovascular research, the zebrafish model is highly valuable due to its translational relevance and accessibility for high-throughput applications. Here, we present ZeCardioAI, a computational platform combining

Treatment of mandibular condylar process fractures: biological considerations.

The topic of condylar injury in adults has generated more discussion and controversy than any other in the field of maxillofacial trauma. It is an important subject because such injuries are common and complications of trauma to the temporomandibular joint (TMJ) are far-reaching in their effects. Why are there so many different methods to treat this injury? How can seemingly disparate treatment options all produce satisfactory outcomes in the majority of patients? The reason lies with the biological adaptations that occur within the masticatory system that are poorly understood, not readily qu

The various meanings and uses of bone "remodeling" in biological anthropology: A review.

OBJECTIVES: In modern bone biology, the term "remodeling" generally refers to internal bone turnover that creates secondary osteons. However, it is also widely used by skeletal biologists, including biological anthropologists as a catch-all term to refer to different skeletal changes. In this review, we investigated how "remodeling" is used across topics on skeletal biology in biological anthropology to demonstrate potential problems with such pervasive use of a generalized term. METHODS: Using PubMed and Google Scholar, we selected and reviewed 205 articles that use the term remodeling to de

Is the whole the sum of its parts? Agent-based modelling of wastewater treatment systems.

Agent-based models (ABMS) simulate individual units within a system, such as the bacteria in a biological wastewater treatment system. This paper outlines past, current and potential future applications of ABMs to wastewater treatment. ABMs track heterogeneities within microbial populations, and this has been demonstrated to yield different predictions of bulk behaviors than the conventional, "lumped" approaches for enhanced biological phosphorus removal (EBPR) completely mixed reactors systems. Current work included the application of the ABM approach to bacterial adaptation/evolution, using

Extremophile Poeciliidae: multivariate insights into the complexity of speciation along replicated ecological gradients.

BACKGROUND: Replicate population pairs that diverge in response to similar selective regimes allow for an investigation of (a) whether phenotypic traits diverge in a similar and predictable fashion, (b) whether there is gradual variation in phenotypic divergence reflecting variation in the strength of natural selection among populations, (c) whether the extent of this divergence is correlated between multiple character suites (i.e., concerted evolution), and (d) whether gradual variation in phenotypic divergence predicts the degree of reproductive isolation, pointing towards a role for adaptat

Global adaptation to a lipid environment triggers the dormancy-related phenotype of Mycobacterium tuberculosis.

UNLABELLED: Strong evidence supports the idea that fatty acids rather than carbohydrates are the main energy source of Mycobacterium tuberculosis during infection and latency. Despite that important role, a complete scenario of the bacterium's metabolism when lipids are the main energy source is still lacking. Here we report the development of an in vitro model to analyze adaptation of M. tuberculosis during assimilation of long-chain fatty acids as sole carbon sources. The global lipid transcriptome revealed a shift toward the glyoxylate cycle, the overexpression of main regulators whiB3, dos

Menstrual variation of autonomic balance may be a factor in exacerbations of certain diseases during the menstrual cycle.

Exacerbation of certain medical conditions during specific phases of the menstrual cycle has long been recognized. Mechanisms of the cyclic variations are poorly understood, but are often attributed to fluctuations in reproductive hormones. We hypothesize that normal variations in autonomic balance during the menstrual cycle, which likely evolved as adaptations for reproduction, may contribute to catamenial variations in diseases independent of hormonal variations. Emerging evidence suggests that autonomic balance shifts towards sympathetic bias during the second half of the menstrual cycle. T

Intensity over duration in neurological rehabilitation: exploring evidence for optimised recovery paradigms.

BACKGROUND: Contemporary stroke rehabilitation protocols traditionally emphasise session frequency and treatment duration over intervention intensity-yet emerging evidence suggests we may be preparing patients for therapeutic marathons when their brains demand neuroplastic sprints. Across neuroscientific, behavioural, and clinical domains, convergent data indicate that repetition density, metabolic load, engagement, and temporal compression-not cumulative minutes-constitute the biologically meaningful drivers of neuroplastic and myoplastic adaptation. OBJECTIVE: This Perspective re-examines c
