A novel role for TRPM8 in visceral afferent function.

Transient receptor potential ion channel melastatin subtype 8 (TRPM8) is activated by cold temperatures and cooling agents, such as menthol and icilin. Compounds containing peppermint are reported to reduce symptoms of bowel hypersensitivity; however, the underlying mechanisms of action are unclear. Here we determined the role of TRPM8 in colonic sensory pathways. Laser capture microdissection, quantitative reverse transcription-polymerase chain reaction (RT-PCR), immunofluorescence, and retrograde tracing were used to localise TRPM8 to colonic primary afferent neurons. In vitro extracellular

Reversible blockade of vestibular evoked activity in the chick.

Vestibular evoked potentials (VsEP) were recorded from young chickens following bilateral intralabyrinthine injections of Tetrodotoxin (TTX) … e responses. Our results indicate that TTX is a useful substance for reversibly blocking vestibular afferent pathways without permanently damaging the labyrinth or neural components. In addition, VsEP is an appropriate tool to objectively evaluate vestibular system function. Their combination can be applied to study the significance of afferent influences on the de …

Neuropeptides, inflammation, and motility.

Neurogenic inflammation is a reaction which includes vasodilation, plasma extravasation, and smooth muscle contraction elicited by activation of and release of mediators from unmyelinated afferent nerve endings. Further release of inflammatory mediators follows activation of axon collaterals associated with these afferent nerve endings as axon reflexes. Substance P, somatostatin, vasoactive intestinal polypeptide, and calcitonin gene-related peptide are candidate mediators. Recent evidence suggests that several of these peptides may be colocalized either with one or more other peptides or with

An enteric-DRG pathway for interoception and visceral pain in mice.

Sensory afferents are major interoceptive pathways for organ-brain communication. Within the distal colon, dorsal root ganglia (DRGs) afferents regulate key gut physiology. Inflammation causes hypersensitivity of DRG pathways, leading to visceral pain. However, whether enteric neurons contribute to interoception and visceral pain remains unclear. Here, we surveyed the DRG innervation along the gastrointestinal tract in mice and found extensive associations between DRG terminals and enteric neurons. Optogenetic activation of different DRG terminals in the distal colon elicited variable degrees

Restoration of the nasopharyngeal response after bilateral sectioning of the anterior ethmoidal nerve in the rat.

In response to stimulation of the nasal passages with volatile ammonia vapors, the nasopharyngeal reflex produces parasympathetically mediated bradycardia, sympathetically mediated increased peripheral vascular tone, and apnea. The anterior ethmoidal nerve (AEN), which innervates the anterior nasal mucosa, is thought to be primarily responsible for providing the sensory afferent signals that initiate these protective reflexes, as bilateral sectioning causes an attenuation of this response. However, recent evidence has shown cardiovascular responses to nasal stimulation with ammonia vapors are

The role of sensory C-fibers in response of vagal afferent stimulation by gastric distension in rats with experimental chronic gastric ulcer.

There is growing evidence that gastric vagal afferent input may contribute to the altered sensations associated with gastrointestinal disorders. The aim of our study was to evaluate gastric vagal afferents (VA) activity in rats with experimental gastric ulcer and ulcer healing. The study was carried out on rats with gastric ulcer (GU), including, a group with perivagal capsaicin pretreatment (CAP), a group with capsaicin administration in gastric ulcer (CAP+GU) animals and control rats. In all rats electrical VA activity was recorded and analysed. In GU rats recordings were carried out in chro

Bladder activation by selective stimulation of pudendal nerve afferents in the cat.

Bladder contractions evoked by pudendal nerve stimulation in both spinal intact and spinal transected cats support the possibility of restoring urinary function in persons with chronic spinal cord injury (SCI). However, electrically evoked bladder responses in persons with SCI were limited to transient contractions at relatively low pressures. This prompted the present study, which presents a detailed quantification of the responses evoked by selective stimulation of individual branches of the pudendal nerve at different stimulation frequencies. In spinal intact cats anesthetized with alpha-ch

Fine structural survey of the intermediate subnucleus of the nucleus tractus solitarii and its glossopharyngeal afferent terminals.

The intermediate subnucleus of the nucleus tractus solitarii (imNTS) receives somatosensory inputs from the soft palate and pharynx, and projects onto the nucleus ambiguus, thus serving as a relay nucleus for swallowing. The ultrastructure and synaptology of the rat imNTS, and its glossopharyngeal afferent terminals, have been examined with cholera toxin-conjugated horseradish peroxidase (CT-HRP) as an anterograde tracer. The imNTS contained oval or ellipsoid-shaped, small to medium-sized neurons (18.2 x 11.4 microm) with little cytoplasm, few cell organelles and an irregularly shaped nucleus.

Afferent projections to the rostral anterior pretectal nucleus of the rat: a possible role in the processing of noxious stimuli.

The afferent inputs to the rostral pole of the anterior pretectal nucleus have been examined by utilizing the retrograde axonal transport of a fluorescent dye, Fast Blue. After unilateral injection of the dye into the rostral anterior prectectal nucleus, large numbers of labelled neuronal somata were found in the somatosensory cortex, the ventrolateral geniculate nucleus, the zona incerta, the superior colliculus, the deep mesencephalic nuclei, the pedunculopontine tegmental nucleus and the medial vestibular nucleus. In addition, the contralateral parabigeminal nucleus provided a major input t

Degenerative and regenerative changes in central projections of glossopharyngeal and vagal sensory neurons after peripheral axotomy in cats: a structural basis for central reorganization of arterial chemoreflex pathways.

Hypoxic hyperventilation in cats is a reflex normally initiated by afferent impulses originating in the carotid body and conducted to the brain stem by the carotid sinus nerves. The reflex response is abolished acutely after section of carotid sinus nerves and excision of the carotid bodies; but, chronically, there is a chemoreflex restoration which is mediated by the aortic body via the aortic depressor nerves. The restoration is associated temporally with changes in efficacy of ventilatory reflexes elicited by electrically stimulating carotid sinus and aortic depressor nerves, and these chan

Role of the parasympathetic nervous system in cardioprotection by remote hindlimb ischaemic preconditioning.

This investigation was designed to determine the participation of the vagus nerve and muscarinic receptors in the remote ischaemic preconditioning (rIPC) mechanism. New Zealand rabbits were anaesthetized, and the femoral artery was dissected. After 30 min of monitoring, the hearts were isolated and subjected to 30 min of global no-flow ischaemia and 180 min of reperfusion (non-rIPC group). The ventricular function was evaluated, considering the left ventricular developed pressure and the left ventricular end-diastolic pressure. In the rIPC group, the rabbits were subjected to three cycles of h

Vagus nerve stimulation: a new promising therapeutic tool in inflammatory bowel disease.

Inflammatory bowel disease (IBD), that is Crohn's disease (CD) and ulcerative colitis, affects about 1.5 million persons in the USA and 2.2 million in Europe. The pathophysiology of IBD involves immunological, genetic and environmental factors. The treatment is medico-surgical but suspensive. Anti-TNFα agents have revolutionized the treatment of IBD but have side effects. In addition, a non-negligible percentage of patients with IBD stop or take episodically their treatment. Consequently, a nondrug therapy targeting TNFα through a physiological pathway, devoid of major side effects and with a

Sensorimotor trigeminal unbalance modulates pupil size.

We studied whether patients affected by Temporo-Mandibular Disorder (TMD), showing asymmetric electromyographic (EMG) activity of masticatory muscles also display asymmetries in pupil size. In 30 pain free TMD patients a highly significant, positive correlation was found between left-right differences in EMG and pupil size. The asymmetry in pupil size was induced by the asymmetric sensorimotor signals arising from the orofacial region, since pupils became of about the same size following orthotic correction, which greatly reduced the EMG asymmetry. Moreover, bite wearing bilaterally increased

Evidence for differential control of posterior hypothalamic, supramammillary, and medial mammillary theta-related cellular discharge by ascending and descending pathways.

Single-unit discharge was recorded from cells in the posterior hypothalamic nucleus (PH), supramammillary nucleus (SuM), and medial mammillary nucleus (MM) during hippocampal theta (theta) elicited by stimulation of the reticular nucleus pontis oralis (RPO). In agreement with previously published work, theta-related cells in the PH (12 cells) were all classified as tonic theta-ON (increased tonic discharge rate during hippocampal theta), whereas those in the SuM (9 cells) and MM (15 cells) were all classified as phasic theta-ON (rhythmic discharge, in phase with ongoing theta). The effect of R

Electrophysiological analysis of the ascending and descending components of the micturition reflex pathway in the rat.

Electrophysiological techniques were used to examine the organization of the spinobulbospinal micturition reflex pathway in the rat. Electrical stimulation of afferent axons in the pelvic nerve evoked a long latency (136 +/- 41 ms) response on bladder postganglionic nerves, whereas stimulation in the dorsal pontine tegmentum elicited shorter latency firing (72 +/- 25 ms) on these nerves. Transection of the pelvic nerve eliminated these responses. Firing on the bladder postganglionic nerves was evoked by stimulation in a relatively limited area of the pons within and close to the laterodorsal t

Histochemical mapping of acetylcholinesterase and butyrylcholinesterase in the medulla oblongata and pons of squirrel (Funambulus palmarum).

The distribution of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) has been investigated in a series of sections passing through the medulla oblongata and pons of the squirrel brain. A comparison of the two enzymes has given an interesting picture of their selective localization in the different nuclei. Marked AChE activity has been observed in the cranial nerve nuclei. BChE activity in various nuclei of the medulla oblongata and pons is variable and occurs diffusely between the cells. Possible reasons pertaining to marked variation in AChE and BChE contents of various nuclei and

A neural geometry for forelimb proprioception in the cervical spinal cord.

Precise, real-time somatosensory feedback is essential for coordinated movement. While the anatomy and physiology of these sensory pathways is well described, their neural code and its construction remains unclear. Here we show that neurons in the cervical spinal cord generate a precise neural representation of the forelimb's kinematic state using muscle and tendon sensory afferent inputs. We identify two classes of movement responsive neurons - the first encodes speed, position and direction of the limb, while the second exhibits precise firing at specific limb positions or grid-like firing p

Hepato-vagal pathway associated with nicotine's anorectic effect in the rat.

Nicotine reduces appetite and body weight. Because the hepato-portal area senses different types of nutrients that transmit signals via vagal afferent nerves to the hypothalamus to modify food intake and feeding pattern, we investigated the effect of nicotine on a hepato-vagal-hypothalamic pathway. Low doses of nicotine (< 10 ng) injected into portal vein (i.p.v.) decreased, while high doses of nicotine increased (> or = 10 ng) electrophysiological activity of hepatic vagal afferents. Stimulatory effect of high dose of nicotine on vagal hepatic afferents was blocked by a prior i.p.v. injection

Central expression of synaptophysin and synaptoporin in nociceptive afferent subtypes in the dorsal horn.

Central sprouting of nociceptive afferents in response to neural injury enhances excitability of nociceptive pathways in the central nervous system, often causing pain. A reliable quantification of central projections of afferent subtypes and their synaptic terminations is essential for understanding neural plasticity in any pathological condition. We previously characterized central projections of cutaneous nociceptive A and C fibers, selectively labeled with cholera toxin subunit B (CTB) and Isolectin B4 (IB4) respectively, and found that they expressed a general synaptic molecule, synaptoph
