RATIONALE: Several studies have demonstrated that males with a family history of alcoholism (FHP) show less of a response to alcohol (e.g. lower ratings of intoxication) than males without a family history of alcoholism (FHN). The purpose of this pilot study was to determine if FHP females also showed a reduced sensitivity to alcohol compared to FHN females.
OBJECTIVES: To determine if FHP females (n=16) were less sensitive to the subjective effects and performance-impairing effects of alcohol compared to FHN females (n=16).
METHODS: The effects of placebo and alcohol (0.25, 0.50, 0.75 g/kg,

Cognitive impairment is frequently observed in patients with alcohol misuse or alcohol addiction. Multiple cognitive functions are reduced in these patients. Frontal lobe functions, as planning, abstract thinking, set shifting or continuous performance are most frequently affected. Alcohol amnestic syndrome, alcohol dementia and the Wernicke-Korsakow-Syndrome constitute distinct entities. Alcohol dementia follows the diagnostic criteria of dementia with clear evidence for alcohol abuse or alcohol addiction. The diagnostic procedure of alcohol-induced cognitive impairment includes medical histo

Scientists and policymakers have explored numerous strategies to prevent alcohol abuse and dependence as well as the adverse social, legal, and medical consequences of alcohol use. Many of these efforts have focused on reducing alcohol-impaired driving and the associated injuries and fatalities. As reported in this article, such efforts have included general deterrence laws (e.g., reduced minimum legal drinking age, administrative license revocation, and lower legal limits for blood alcohol concentrations), measures targeted at repeat offenders, and measures to control alcohol availability (e.

Fetal alcohol syndrome is often associated with severe physical and neuropsychiatric maldevelopment. On the other hand, some offspring of women who drank during pregnancy appear to be affected in minimal ways and function relatively well within society. We questioned whether this effect of prenatal alcohol in the adult is generally minimal. To bear on this, we determined whether we could distinguish alcohol-exposed from nonexposed individuals in a population of male veterans, selected because of both their accepted level of function within society (e.g., honorable discharge from the military)

Recent human genetic studies suggest that a predisposition to alcohol abuse and/or to develop alcoholism may be inherited. Pedigree analysis, linkage, and association studies have helped to detect marker loci and candidate genes that may prove useful in identifying individuals at risk. In particular, molecular genetic research into the causes of alcoholism has drawn attention to the potentially important role of alcohol- and acetaldehyde-metabolizing enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH). Functional polymorphisms have been observed at various genes encoding the

Ethanol blood levels are the result of alcohol absorption and the process of its distribution, metabolism and excretion. Kinetics are complex and not yet well known. They can be influenced by acquired factors (type of alcohol ingested, association with fasting or eating, induction or inhibition of ethanol metabolism) or by genetically determined differences in the activity of alcohol and of acetaldehyde dehydrogenase. The presence of ethanol in the organism leads to various consequences. On the one hand, hydropic changes on membranes modify their function and thus that of membrane proteins (pa

In view of conflicting results concerning differences between sons of alcoholics and sons of nonalcoholics in response to a single alcohol challenge (with a given dose), we exposed these high and low risk groups to several sessions in which they drank alcohol at the same dose in order to measure the development of chronic tolerance or sensitization with repeated doses. Sons of alcoholics and sons of nonalcoholics received a moderate dose of alcohol (0.5 g/kg) in three sessions with alcohol, followed by a placebo session. Sons of alcoholics developed reverse tolerance or chronic sensitization t

An empirical biobehavioral research approach to the conditions generally identified as alcohol abuse and alcoholism emphasizes the temporal ordering of participating biochemical, physiological, and behavioral events that provide an operational basis for characterizing the functional aspects of this complex disorder and identifying distinguishable features of the alcohol abuse and dependence process. The available evidence suggests that alcoholism is a condition determined by a host of continuous variables rather than an entity possessing static qualities that imply intractability. The challeng

In this study 444 chronic alcoholic patients, hospitalized at the beginning of this study, were followed up for 4 to 7 years (31). During this time 101 patients died (23.2%), with whom severeness of disease as well as the extent of social depravation could be identified as factors influencing mortality. Beginning in January of 1982 we investigated mortality of the population of a village with a similar socio-cultural background (working situation, rural population, wine-growing area). As it was possible to cooperate with the local general practitioner, who has been living and working in this v

Ultrastructure of alcoholic hyaline (AH) in alcoholic hepatitis was observed to demonstrate the relationship between AH filaments and intermediate filaments (IFs). Type 1, Type 2 and Type 3 AH were noted in hepatocytes, and the filaments in these types of AH showed similar ultrastructural findings as already reported. Two different kinds of filaments were noted in the Type 2 AH. The large on showed 160 to 200 A in a diameter and the small one showed around 100 A in diameter. Of the 29 cases with alcoholic hepatitis, a new type of AH was noted in the rosette forming hepatocytes in 2 cases of al

The existence of FAS can no longer be denied; reports of greater than 200 cases which fall into a phenotypically recognizable continuum justify its existence. At this point, however, it is probably prudent to withhold judgment of the specific human teratogenicity of alcohol until more refined information regarding dose-risk relationships are available not only from humans, but also from animal models, including primates. It may be that Fetal Alcohol Syndrome, at this point, should be considered the Fetal Alcoholism Syndrome. At any rate, until much more is known, it would seem advisable to obs

A toxic effect of alcohol is the principal cause of the development of liver disease in alcoholism. Fatty infiltration of the liver is a consequence of ethanol metabolism due mainly to an increased synthesis and decreased degradation of fatty acids. Mechanisms that have been suggested for ethanol-induced hepatocellular necrosis include centrolobular hypoxia due to an increased oxygen requirement and intracellular accumulation of protein, fat, and water which results in increased cell size. Hepatocellular necrosis, however, may not be a necessary stage in the development of cirrhosis. Chronic e

Selective oxidation of hydrocarbons requires highly selective catalysts toward desired reaction products. In this study, SiO2-supported Ti catalysts were synthesized featuring isolated Ti oxide species (0.2 Ti atoms/nm2) that serve as active Lewis acid sites for the selective oxidation of cyclohexene with H2O2. To investigate the effect of the surface microenvironment on catalytic performance, post-modification was performed using alcohols with different terminal groups (octyl (O), triglycol (TG), and fluorous (F)). FT-IR spectroscopy confirms that the deposited organic groups were successfull…

When [Cp*Ru(MeCN)3]PF6 is used as the catalyst for the trans-hydrosilylation of propargyl alcohol derivatives, the silyl group is transferred with appreciable levels of regioselectivity to the alkyne C-atom distal to the protic substituent. A copper-mediated 1,4-Brook rearrangement converts the resulting alkenylsilanes into alkenylcopper species, which can be intercepted with a multitude of carbon- or heteroatom-based electrophiles; this formal ipso-substitution of the R3Si-group allows a variety of trisubstituted olefins to be formed in stereodefined format, including fluoroalkenes and olefin
