ε-Polylysine/Sodium Alginate Bilayer-Modified Nanoliposomes Enhancing the Stability and In Vitro Bioavailability of Epigallocatechin Gallate.

Epigallocatechin gallate (EGCG) represents the key phenolic compound in green tea, which has been verified to possess various biological effects but suffers from low stability and poor bioavailability. To address these issues, EGCG-loaded nanoliposomes (ELs) were screened and prepared using an ethanol injection-calcium acetate gradient (EtOH-CAG) method. An encapsulation efficiency of 94.61% was achieved, involving a particle size of 118.6 nm and a polydispersity index (PDI) of 0.23. Via layer-by-layer assembly, nanoliposomes modified with either ε-polylysine (ε-PL) monolayer (ELP) or ε-polyly

Modular alginate-based hydrogels embedding multilayer nanofibers: A synergistic strategy for hemostasis, antibacterial, and immunomodulatory therapy.

Non-compressible hemorrhage remains a leading cause of trauma mortality, with ineffective management triggering infection, dysregulated inflammation, and multi-organ dysfunction. Herein, we report a multilayer nanofiber-reinforced alginate hydrogel (GT-F) combining rapid hemostasis, strong tissue adhesion, high mechanical resilience, and wound healing. The hydrogel matrix, composed of dopamine-modified methacrylate alginate (AMD) and gelatin methacryloyl (GelMA), was incorporated with tranexamic acid (TAX). The biomimetic multilayer nanofiber framework is constructed from hydrophobic polycapro

Tailoring Drug Release Profiles From Electrospun Alginate Fibers Through Integration of Mesoporous Silica Nanoparticles.

Electrospun hydrogels often lack sufficient mechanical integrity for biomedical applications, while nanoparticle-reinforced systems are commonly associated with synthetic matrices or complex fabrication methods. Here, electrospun methacrylated alginate (AM) fibrous hydrogels reinforced with mesoporous silica nanoparticles (MSNs) are introduced as a simple, biocompatible platform for localized drug delivery. The mats are UV-cross-linked in ethanol to maintain fibrous morphology and allow chlorhexidine (CHX) loading from 0.2 to 5 mg ml- 1. Incorporation of 1 wt.% MSNs doubles compressive and ten

Mechanism of crosslinking temperature impacting the structure of calcium ion crosslinked sodium alginate/konjac glucomannan films.

Crosslinking temperature has a significant effect on the progress of the crosslinking reaction. This study systematically investigated the effect of crosslinking temperature (15-55 °C) on the structure of calcium ion crosslinked sodium alginate (SA)/konjac glucomannan (KGM) films as well as the packaging-related properties. The results indicated that crosslinking temperature not only influenced the ionic crosslinking between Ca2+ and SA, but also affected the intermolecular interactions between SA and KGM, leading to significantly different film properties. Specifically, at a relatively low cr

Ethanol Fermentation-based Valorization of Alginate Extraction Residue from Pelagic Brown Seaweed Sargassum siliquosum.

Brown seaweeds are the primary industrial source of alginates and are increasingly valued as marine-based feedstocks in integrated biorefineries. However, the substantial solid residue generated in the alginate extraction industry remains underexplored as a secondary source of carbon and energy. This study investigated the potential of the alginate-extracted residue (AER) from Sargassum siliquosum biomass collected from the southwestern coast of Taiwan as a substrate for bioethanol production. The alkaline extraction method yielded 39.00% alginate and 33.80% residue on a dry weight basis. Comp…

From molecular architecture to clinical translation: Structure-property-performance relationships in alginate-based nanostructured hydrogels.

Alginate-based hydrogels have emerged as versatile biomaterials because of their biocompatibility, high water content, mild aqueous processing, and tunable network architecture. However, their performance is strongly dependent on molecular characteristics, including molecular weight, mannuronic-to-guluronic acid (M/G) ratio, block distribution, degree of functionalization, and crosslinking density. This review critically examines how these parameters govern the physicochemical, mechanical, biological, and translational performance of alginate-based nanostructured hydrogels. Particular emphasis

Alginate-encapsulated enzymes mitigate deoxynivalenol-induced hepatic mitochondrial dysfunction and glycolithocholic acid accumulation in piglets.

BACKGROUND: Deoxynivalenol (DON), a mycotoxin commonly found in grains and feed, poses a serious threat to animal and public health. This study aimed to investigate the underlying mechanisms of DON-induced liver damage in piglets and to elucidate the potential of alginate-encapsulated enzyme in mitigating hepatotoxicity in vivo. RESULTS: The results indicated that DON exposure induced growth retardation and hepatic mitochondrial damage in piglets, which were effectively rescued by DON-degrading enzymes (DDE) supplementation. Hepatic transcriptomic profiling revealed that DON induced downregul

A Protocol for Colorectal Tumor Spheroid Culture in Tunable Stiffness Alginate-Based Hydrogels and Subsequent Immunohistochemical Analysis.

Tumor mechanical microenvironment, particularly extracellular matrix stiffness, plays a critical role in regulating cancer cell behavior, including proliferation, quiescence, and drug resistance. Conventional 2D culture or stiff 3D scaffolds fail to recapitulate the physiological soft (normal) or pathologically stiff (tumoral) mechanical niches. Here, we present a detailed protocol for establishing a tunable 3D tumor spheroid culture system using sodium alginate-based hydrogels crosslinked with calcium ions at different concentrations to achieve soft or stiff conditions that mimic normal colon

Alginate-based mucoadhesive nanofibrous system embedding resveratrol-loaded vesicles as a therapeutic platform for nasal disorders.

Oxidative stress and inflammation play a key role in the development and progression of several otorhinolaryngological disorders, including chronic rhinosinusitis and allergic rhinitis. Resveratrol (RSV), a natural polyphenol with antioxidant and anti-inflammatory properties, is a promising therapeutic candidate, although its clinical application is limited by poor aqueous solubility and rapid mucociliary clearance upon nasal administration. In this study, a mucoadhesive nanofibrous nasal patch based on sodium alginate and poly(ethylene oxide) embedding RSV-loaded phospholipid vesicles was dev

Injectable pH-responsive Bletilla striata polysaccharide-sodium alginate hydrogel promoting peripheral nerve regeneration.

Within the clinical realm, the complex process of nerve regeneration following traumatic peripheral nerve injury (TPNI) remains a formidable challenge. Injectable multifunctional hydrogels that align with the highly orchestrated cascades of nerve regeneration have emerged as promising biomaterial strategies for the TPNI treatment. Here, we develop an injectable pH-responsive composite hydrogel (BSP-SA) based on physical interactions between Bletilla striata polysaccharide (BSP) and sodium alginate (SA). This system responds to the mildly acidic microenvironment of the injured nerve, enabling c

In vitro and in silico studies on anticancer activities of alginate oligosaccharide in Epstein-Barr virus-positive nasopharyngeal carcinoma.

Alginate oligosaccharides (AOS) have recently shown promising activities in inhibiting tumour growth in osteosarcomas. It is, however, unknown if AOS is also effective against nasopharyngeal carcinoma (NPC). To this end, the antiproliferation activities of enzymatically derived AOS were investigated against the EBV-positive NPC cell line, C666-1. MTT cytotoxicity assays revealed an antiproliferation effect against the C666-1 cell line, albeit at concentrations above 10 mg/mL, but promoting growth at lower concentrations. As a potential heparin sulfate analog, which also demonstrates a similar

Interpenetrating polymer network microspheres based on alginate and thiolated CNF for tumor-triggered localized intratumoral therapy.

Interpenetrating polymer network (IPN) microspheres composed of alginate and thiolated TEMPO-oxidized cellulose nanofibers (Th-TCNF) were developed as reduction-responsive carriers for Mitoxantrone (MIT) delivery. TCNF was thiolated through covalent coupling with cysteamine, as confirmed by H NMR, FT-IR and XPS which successfully verified thiol grafting. Spray-dried microspheres were subsequently cross-linked via Ca2+ coordination and disulfide bond formation, generating a mechanically reinforced IPN structure. Swelling analysis showed significantly reduced equilibrium swelling in the full IPN

Multi-crosslinked sodium alginate fibers with super-hydrophilicity, high strength, flame retardancy, and antibacterial properties.

Sodium alginate (SA) fibers are promising biomass-based material for sustainable applications, but their low limiting oxygen index (LOI) and lack of antibacterial properties restrict their use in flame-retardant protection. Traditional functionalization approaches often sacrifice mechanical properties, while enhancing strength alone typically fails to meet multifunctional requirements. To address this challenge, we developed a dual-crosslinked SA fiber combining calcium ion crosslinking and chemical crosslinking using hexamethylene diisocyanate (HDI) as the cross-linking agent in an aqueous so

Cryoelectrospun Elastin-Alginate Scaffolds Support In Vitro 3D Epithelial-Stromal Cocultures for Salivary Tissue Engineering.

Bioengineered functional salivary tissues can advance regenerative therapies, preclinical drug testing, and the fundamental understanding of salivary gland dysfunction. Current salivary tissue models are typically Matrigel-based, hydrogel-based or scaffold-free organoid systems, with limited physiological relevance or mimicry of cell-cell and cell-extracellular matrix (ECM) interactions. We previously developed elastin-alginate cryoelectrospun scaffolds (CES) that resemble the topography and viscoelastic properties of healthy salivary ECM, and validated their potential for stromal cell culture

Assessing mesh size and diffusion of alginate bioinks: A crucial factor for successful bioprinting functional pancreatic islets.

Three-dimensional (3D) bioprinting has been utilised for the encapsulation of pancreatic islets for potentially treating type 1 diabetes. A crucial factor in selecting a cell compatible bioink, that maintains islet functionality, is the mesh size and diffusion capacity of the bioink. In this study, we present a screening strategy for alginate hydrogel formulations in three-dimensional bioprinting, utilizing the fluorescent recovery after photobleaching (FRAP) method and measuring the mesh size of the hydrogels. Subsequently, the 1.5 % alginate formulation that had been selected was used to bio

Preparation of TEMPO-partially oxidized sodium alginate/SF scaffolds with degradation compatibility for skin defect repair.

This study aimed to curtail the secondary damage caused by traditional dressings by cross-linking TEMPO-mediated oxidized sodium alginate (TOSA) with silk fibroin (SF) through the EDC system and preparing the scaffolds through freeze-drying and anhydrous ethanol immersion. The good adsorption, mechanical properties, high porosity, and good in vitro and in vivo biocompatibility of the scaffold can improve the healing efficiency of the wound. Additionally, sodium alginate was oxidized by the TEMPO system to lower its molecular weight and hence increase the degradation rate of the scaffold. Conse
