Glucose and physostigmine effects on morphine- and amphetamine-induced increases in locomotor activity in mice.

Recent findings indicate that glucose antagonizes several behavioral effects of cholinergic antagonists and augments those of cholinergic agonists. For example, scopolamine elicits increased locomotor activity, an action which is attenuated by glucose and by combined treatment with glucose and physostigmine at doses which are individually without effect. Opiate and catecholamine agonists, such as morphine and amphetamine, also elicit hyperactivity. The present study examined interactions of glucose and physostigmine with morphine- and amphetamine-induced hyperactivity. Mice received saline, mo…

Effects of d-amphetamine, morphine, naloxone, and drug combinations on visual discrimination in rats.

The effects of d-amphetamine, morphine, and naloxone on visual discrimination were investigated using a two-choice discrete-trial procedure in which rats were trained to discriminate the position of a lightflash. Morphine (0.3-5.6 mg/kg) but not amphetamine (0.1-1.0 mg/kg) caused a significant dose-dependent disruption in discriminative performance. Both amphetamine and morphine increased response latencies. Naloxone (1.0 mg/kg) prevented the disruption of any aspect of performance by up to 100 mg/kg morphine. Performance after naloxone/amphetamine co-administration was not significantly diffe…

Effects of naltrexone, and d-amphetamine, and their interaction on the stimulus control of choice behavior of rats.

The hypothesis that endogenous opioid peptides modulate attentional processes was tested. The effects of the opioid antagonist naltrexone (NALT), d-amphetamine (AMP), and their interaction were investigated in rats trained in a two-choice task in which the position of a short-duration light served as a cue for food-reinforced responses. NALT (0.25, 1.0, 5.0, and 10.0 mg/kg) produced no significant changes in performance (accuracy, choice latency, and food retrieval time). As predicted, AMP induced dose-dependent biphasic effects. Low doses of AMP (0.25 and 0.5 mg/kg) significantly enhanced acc…

Liquid chromatographic determination of the enantiomeric composition of amphetamine prepared from norephedrine and norpseudoephedrine.

The stereochemical course of the synthesis of amphetamine from norephedrine and norpseudoephedrine is investigated using liquid chromatography. The results show that the chiral carbon common to both compounds remains unaffected during the reaction sequence. The presence of individual amphetamine enantiomers in the reaction products is determined by reversed-phase liquid chromatographic separation on an achiral stationary phase (C18) following precolumn derivatization with 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranosyl isothiocyanate (GITC). The GITC derivatization procedure allows for the liquid…

Differential analgetic actions of amphetamine enantiomers in the mouse: a drug-drug interaction study.

A series of agents were tested for their ability to interact with the analgetic actions of either d-amphetamine (d-AMP) or l-amphetamine (l-AMP), or morphine in rats using the hot plate procedure. The analgetic action of l-AMP was potentiated by morphine and slightly antagonized by naloxone; it was antagonized by clonidine, alpha-MT, yohimbine, fluoxetine and metergoline, but enhanced by PCPA. The analgetic action of morphine was antagonized by both naloxone and PCPA. The analgetic actions of l-AMP and morphine appear to involve different biogenic amine systems.

Effect of minor tranquilizers, tryptamine antagonists and amphetamine on behavior punished by brain stimulation.

Earlier observations have shown that septal lesions released operant responding punished by foot-shock, but did not change behavior punished by electrical stimulation of the dorsal periaqueductal gray (DPAG) substance of the rat brain. In contrast, chlordiazepoxide facilitated both kinds of punished responding. In order to further study the mechanism of brain stimulation punishment, dose-response curves of two minor tranquilizers, chlordiazepoxide and pentobarbital, of two tryptamine antagonists, methysergide and cyproheptadine as well as of amphetamine on lever-pressing behavior of rats maint…

Effects of d-amphetamine and apomorphine in a new animal model of petit mal epilepsy.

d-Amphetamine is effective in controlling seizures in petit mal epilepsy. The flash-evoked afterdis-charge (FEAD) in rats has been proposed as a model of the peit mal seizure. The experiments reported here investigated the dose response relationship for the suppression of FEAD by d-amphetamine, and compared its effects with those of the dopamine-mimetic, apomorphine. Significant suppression of FEAD was observed at doses of d-amphetamine greater than 0.2 mg/kg. A maximum decrease of 60% occurred at 1.2 mg/kg. Higher doses did not result in any further suppression. In contrast, apomorphine and n…

Noradrenergic action of amphetamine following degeneration of descending monoaminergic fibers in the spinal cord.

Amphetamine and methoxamine facilitated the flexor reflex in chronic spinal rats whose descending monoaminergic nerve endings were degenerated. Methoxamine, but not amphetamine, facilitated the flexor reflex in the chronic spinal rat 16-20 h after IV 6-hydroxydopamine treatment. These data indicate that amphetamine's spinal cord facilitative effects are mediated through two noradrenergic mechanisms. In the acute spinal rat, its facilitative effects in these experiments seem to be independent of the sympathetic nervous system and are probably dependent on the presence of the terminal axons of t…

Effects of acute and chronic amphetamine treatment on Purkinje neuron discharge in rat cerebellum.

The spontaneous discharge of the cerebellar Purkinje neuron in urethane-anesthetized rats is used as an indicator of central noradrenergic activity during the acute and chronic administration of d-amphetamine. Acute parenteral administration of amphetamine causes a dose-dependent slowing of the discharge. Since this effect is not seen in animals in which the catecholaminergic innervation has been selectively removed by previous treatment with 6-hydroxydopamine, it is hypothesized that the slowing is caused by the increased release and/or blocked reuptake of endogenous norepinephrine. Acute adm…

The role of serotonergic and cholinergic systems in the aggression-stereotypy complex produced by amphetamine in mice.

The aggression--stereotypy complex (ASC) was produced in mice by administration of 15 mg/kg of amphetamine. The elevation of cerebral serotonin level in the mouse brain by administration of L-tryptophan or DL-5-hydroxytryptophan suppressed the aggressiveness without affecting significantly the stereotypy. Serotonin antagonists: methysergide and cyproheptadine, did not potentiate clearly the aggressive component of ASC. Cholinomimetics, pilocarpine and physostigmine, strongly suppressed the aggressive component, while a cholinolytic, scopolamine, evidently potentiated the elements of aggressive…

A comparative study of different amphetamines on copulatory behavior and stereotype activity in the female rat.

The influence of D- and L-amphetamine, fenfluramine, and p-chloroamphetamine on female copulatory behavior (lordosis response) and the induction of stereotype activity was compared. Lordosis response in the female rat has been shown to be inhibited by increased central nervous serotonergic (5-HT) as well as dopaminergic (DA) activity. A dose-dependent inhibitory effect on the estrogen- + progesterone-induced lordosis response in ovariectomized rats was demonstrated after treatment with the four amphetamines. In contrast, only D- and L-amphetamine induced a stereotype activity, which is conside…

Aminorex (Menocil) and amphetamine: acute and chronic effects on pulmonary and systemic haemodynamics in the calf.

Reports of pulmonary hypertension in obese women treated with the appetite suppressant aminorex (Menocil) prompted this study using the calf as a model because of its very reactive pulmonary vascular bed. Acute and chronic haemodynamic effects were observed following daily intravenous injections of aminorex (0·25 mg/kg as free base) in four calves. Four other calves were similarly treated with the related sympathomimetic drug amphetamine in the same dose, and four calves served as controls. The acute effects of both these drugs were a significant increase in systemic arterial pressure and resi…

Retrospective Study on the Power and Interpretation of Chiral Amphetamine Analysis in Hair for Forensic Applications.

Chiral amphetamine analysis is performed in forensic toxicology to determine the nature of amphetamine intake (therapeutic or illicit). It is crucial to the correct interpretation of amphetamine-positive hair samples in terms of abstinence control and therapy compliance, where hair analysis is used to evaluate the average amphetamine use over a period of several months. However, interpretative challenges arise when different medication intakes and/or illicit drug consumption events contribute to the hair analysis result. Therefore, a retrospective study of authentic casework was conducted (201…

Distribution of amphetamine, benzoylecgonine, cocaethylene, cocaine, fentanyl, and methamphetamine in seven regions of the brain.

Brain tissue is a potentially valuable postmortem specimen because of its relative anatomical isolation, reduced susceptibility to postmortem degradation, and relevance as the primary site of action for many drugs. Despite the frequent detection of stimulants and synthetic opioids in medicolegal death investigations, limited data exist regarding their regional distribution within the human brain. This study evaluated the distribution of amphetamine (AMP), methamphetamine (MAMP), cocaine, benzoylecgonine, cocaethylene, and fentanyl across seven anatomically distinct brain regions (basal ganglia…
