The Effect of Myrtacine and Celastrol-Enriched Extract on Cutibacterium acnes Extracellular Vesicles.

BACKGROUND: Commensal bacteria interact with each other and their environment, secreting anti-microbial peptides, bacteriocins, and extracellular vesicles (EVs). Cutibacterium acnes (C. acnes), a commensal bacterium that plays a major role in acne, is classified into different phylotypes (IA1, IA2, IB, IC, II, and III), where overabundance of phylotype IA1 relative to other phylotypes is found in acne lesions. EVs secreted by C. acnes phylotype IA1 extracted from an acne lesion were previously found to induce a pro-inflammatory response in skin models. Here, we investigated the effects of Myrt…

Microbiota and parasite relationship.

The diversity of microbiota is different in each person. Many health problems such as autoimmune diseases, diabetes, cardiovascular diseases … ystis infection without IBS. Lactobacillus species reduce the presence of Giardia, and the produced bacteriocins prevent parasite adhesion. The presence of helminths has been strongly associated with the transition from Bacteroidetes to Firmicutes and Clostridia. Contrary to Ascaris, alpha diversity in the intestinal microbiota decreases in chronic Trichuris m …

Streptococcus salivarius K12 Limits Group B Streptococcus Vaginal Colonization.

Streptococcus agalactiae (group B streptococcus [GBS]) colonizes the rectovaginal tract in 20% to 30% of women and during pregnancy can be t … regnant women. Inhibition was dependent on the presence of megaplasmid pSsal-K12, which encodes the bacteriocins salivaricin A and salivaricin B; however, in coculture experiments, GBS growth was impeded by K12 independently of the megaplasmid. We also demonstrated that K12 adheres to and invades human vaginal epithelial cells at levels comparable to GBS. Inhi …

Snake Gut Microbiota as a Source of Anti-Inflammatory Probiotics: Isolation and Functional Characterization of Two Novel Strains.

The intestinal microbiome is fundamental to host physiological homeostasis, while deviations from its balanced state have been linked to inflammatory bowel diseases (IBD). To address the limitations of conventional antibiotic therapies, this study explored snake gut microbiota as a novel source of anti-inflammatory probiotics. We explored the gut microbiota of five snake species (Deinagkistrodon acutus, Trimerodytes annularis, Trimerodytes percarinatus, Lycodon rufozonatus, and Trimeresurus stejnegeri) through metagenomic sequencing. Community composition analysis revealed that the phylum-leve

Analysis of a collection of Escherichia coli isolates recovered from beach waters on the coast of the Río de la Plata in Montevideo.

Antibiotic resistance has become a global and multidimensional problem, where the capacity for environmental dispersion is one of the most important aspects. Estuaries are usually hotspots for anthropogenic impact because they receive effluents from wastewater treatment plants. This constitutes a potential health risk when they are used for recreational activities. Río de la Plata is an estuarine system in South America, located south of Uruguay, on whose western margin lies Montevideo, the capital of this country. In the coastal area of Montevideo, the estuary receives effluents from the city

Probiotic potential of autochthonous Enterobacter mori ASP8 and Bacillus stercoris ASP10 isolated from gut microbiota of healthy stinging catfish, Heteropneustes fossilis (Bloch).

This study characterizes two bacterial isolates Enterobacter mori ASP8 and Bacillus stercoris ASP10, derived from the stinging catfish (Heteropneustes fossilis) and evaluates in vitro probiotic efficacy for enhancing sustainability in freshwater aquaculture. Rigorous biosafety assessments confirmed that both strains were non-hemolytic, were non-blood coagulant, lack DNase/RNase activity, and were non-biogenic amine producers. Both isolates exhibited high gastrointestinal resilience and survival under stimulated stomach duodenum passage (SSDP) with a value of 81.98% for E. mori ASP8 and 84.89%

Expression and Characterization of Recombinant Triple Laterosporulin in Chlamydomonas reinhardtii.

Antimicrobial peptides with broad-spectrum antibacterial properties have emerged as promising candidates for combating bacterial infections. Laterosporulin (LS), a class II bacteriocin initially isolated from Brevibacillus sp. strain GI-9, has demonstrated broad-spectrum antibacterial activity and notable biochemical characteristics. However, the limited expression levels of LS in Brevibacillus sp. GI-9 restrict its potential for large-scale production and therapeutic and food preservation applications. In this study, a triple peptide comprising three tandem repeats of LS (3 × LS), tagged with

Reference Whole Genome Sequence Analyses and Characterization of a Novel Carnobacterium maltaromaticum Distinct Sequence Type Isolated from a North American Gray Wolf (Canis lupus) Gastrointestinal Tract.

We hypothesize that bacteria isolated from free-ranging animals could potentially be useful for practical applications. To meet this objective a Gram-positive bacterium was isolated from the gastrointestinal (GI) tract of a Gray Wolf (Canis lupus) using Brucella broth with hemin and vitamin K (BBHK). By small ribosomal RNA (16S) gene sequencing the bacterium was initially identified as a novel Carnobacterium maltaromaticum strain. The bacterium could be propagated both anaerobically and aerobically and was both catalase/oxidase negative and negative by the starch hydrolysis as well as negative

Integron-Mediated Antimicrobial Resistance and Virulence Factors in Salmonella Typhimurium Isolated from Poultry.

This study investigates antimicrobial-resistant (AMR) Salmonella Typhimurium in poultry, focusing on how class I integrons contribute to AMR and virulence. Using whole genome sequencing, researchers analyzed 26 S. Typhimurium isolates from U.S. poultry, finding that three isolates contained integrons (1000 base pairs each). These integron-positive isolates exhibited significantly higher resistance to beta-lactams, phenicols, and tetracyclines compared to integron-free isolates (p = 0.004, 0.009, and 0.02, respectively) and harbored genes like ges, imp, and oxa, which are linked to extended-spe

Linker-Improved Chimeric Endolysin Selectively Kills Staphylococcus aureus In Vitro, on Reconstituted Human Epidermis, and in a Murine Model of Skin Infection.

Staphylococcus aureus causes a broad spectrum of diseases in humans and animals. It is frequently associated with inflammatory skin disorders such as atopic dermatitis, where it aggravates symptoms. Treatment of S. aureus-associated skin infections with antibiotics is discouraged due to their broad-range deleterious effect on healthy skin microbiota and their ability to promote the development of resistance. Thus, novel S. aureus-specific antibacterial agents are desirable. We constructed two chimeric cell wall-lytic enzymes, Staphefekt SA.100 and XZ.700, which are composed of functional domai

Interactions between probiotic and oral pathogenic strains.

More than 6 billion bacteria and other microorganisms live in the adult oral cavity. As a result of any deleterious effect on this community, some microorganisms will survive better than others, which may trigger pathogenic processes like caries, halitosis, gingivitis or periodontitis. Oral dysbiosis is among the most frequent human health hazards globally. Quality of life of patients deteriorates notably, while treatments are often unpleasant, expensive and irreversible, e.g. tooth loss. In the experiments reported here, we investigated the individual interactions between 8 pathogenic and 8 p

A Rapid Lysostaphin Production Approach and a Convenient Novel Lysostaphin Loaded Nano-emulgel; As a Sustainable Low-Cost Methicillin-Resistant Staphylococcus aureus Combating Platform.

Staphylococcus aureus is a Gram-positive pathogen that is capable of infecting almost every organ in the human body. Alarmingly, the rapid emergence of methicillin-resistant S.aureus strains (MRSA) jeopardizes the available treatment options. Herein, we propose sustainable, low-cost production of recombinant lysostaphin (rLST), which is a native bacteriocin destroying the staphylococcal cell wall through its endopeptidase activity. We combined the use of E. coli BL21(DE3)/pET15b, factorial design, and simple Ni-NTA affinity chromatography to optimize rLST production. The enzyme yield was up to

An approach on the potential use of probiotics in the treatment of skin conditions: acne and atopic dermatitis.

Acne and Atopic Dermatitis (AD) are chronic inflammatory skin conditions with severe impact on a patient's life. Current treatments are related to adverse effects and do not represent a definitive cure. The present paper reviews the alterations in skin microbiome, specifically in acne and AD, and aims in searching for potential treatments based on benefic microorganisms, called probiotics. The review was made through bibliographic search of the main databases (Science Direct, PubMed, Scielo, Medline) between September 2015 and June 2016. Acne lesions create an environment that facilitates the

Influence of Oral Probiotic Streptococcus salivarius K12 on Ear and Oral Cavity Health in Humans: Systematic Review.

Traditionally, probiotics are linked to the good health of the intestine and most clinical studies focus on that field. Evidence of oral probiotic use for ear and oral cavity disease prevention with impact on human health is limited. This work reviews existing studies and literature on Streptococcus salivarius K12 as an oral probiotic and effects of S. salivarius K12 on human ear and oral cavity human health. The studies were accessed via database searches: MEDLINE, PubMed, and Elsevier. The search included/focused on/encompassed publications from 2003 to 2016 with keywords related to K12 Stre

The outcome of H. influenzae and S. pneumoniae inter-species interactions depends on pH, nutrient availability and growth phase.

Haemophilus influenzae and Streptococcus pneumoniae exist together as common commensals of the healthy human nasopharynx, but both are important aetiological agents of different diseases, including the paediatric disease otitis media. It was recently shown that the formation of a multispecies biofilm of H. influenzae and S. pneumoniae is the cause of chronic forms of otitis media. However, the interactions between the two species are not clearly defined. Using a defined and kinetic analysis, our study has shown that while co-existence of the two species occurs, S. pneumoniae is also able to co

[Microbiology of streptococcal infections].

Since the discovery of streptococci by the surgeon of Vienna, Theodor Billroth, more than 100 years ago, they have proved to be a bacterial group of great medical and epidemiological importance. The classification in growth-characteristics on blood culture mediums (alpha-, beta- and gamma-hemolysis) has been detached by the evidence of group specific cell wall antigens. The antigene extraction described by Lancefield can distinguish at least 21 serogroups (A-T). They have also taken over the historical names (S. pyogenes, S. agalactiae etc.). In addition to group relationship the antigen struc
