Chiral benzylic alcohols serve as key intermediates in the pharmaceutical industry. Through structure-guided semirational engineering, a mutant library of ketoreductase KR8 was developed that catalyzes the stereoselective reduction of aryl ketones to chiral benzylic alcohols with high enantioselectivity. In combination with KR14, these ketoreductases form a stereocomplementary biocatalytic tool box featuring a broad substrate scope. For 12 aryl ketone substrates, the corresponding enantiomers of benzylic alcohols were all obtained with high stereoselectivity and conversion efficiency. This wor

Enzymes represent promising biocatalysts; however, their industrial application is often limited by instability under harsh conditions, particularly their low tolerance to organic solvents. Although immobilization within porous organic frameworks (POFs) offers a potential solution, systematic investigations remain limited. In this work, Aspergillus niger lipase (ANLP) was immobilized in three POFs (ZIF-8, COF-LZU1, and HOF-101), and the tolerance of these composites was evaluated in six common organic solvents, including n-hexane and dichloromethane. The underlying mechanisms governing solvent

Non-canonical amino acids (ncAAs) are privileged building blocks for the synthesis of natural products, biocatalysts, and drug molecules. Among them, γ-tertiary-nitro-α-amino acids are highly valuable yet remain scarcely explored due to challenges in synthetic accessibility. Although pyridoxal-5'-phosphate (PLP)-dependent enzymes are powerful biocatalysts for ncAA production, no enzymatic platform has, to the best of our knowledge, enabled efficient access to γ-tertiary-nitro-α-amino acids. Here, we report a PLP-dependent enzymatic platform that selectively couples O-acetyl-serine with seconda

BACKGROUND: β-Galactosidase (β-Gal) serves as an important biomarker for primary ovarian cancer and cellular senescence, as well as a key indicator for Escherichia coli (E. coli). Besides, β-Gal serves as a common reporter enzyme in biosensing systems and is widely applied to improve lactose digestion in food industry. Current β-Gal detection strategies mainly rely on probe-based one-step reactions, which often suffer from limited sensitivity. Therefore, the development of highly sensitive β-Gal detection methods based on signal amplification is urgently needed and remains a great challenge.

Nicotinic acid, a bioactive form of niacin (vitamin B3), is an essential nutrient involved in various metabolic pathways. Its deficiency can lead to severe health issues, emphasizing the need for effective production methods. Traditional chemical methods for synthesizing nicotinic acid often require harsh reaction conditions and produce environmentally hazardous byproducts. This has prompted increasing interest in more sustainable biocatalytic alternatives. In this study, we investigated the bioconversion of 3-methylpyridine (3-MP) to nicotinic acid using resting cells of recombinant Escherich

Pro-Xylane is a biologically active C-glycoside compound in cosmetic products. The conventional method involves converting d-xylose from beech trees to 1-C-(β-d-xylopyranosyl)-acetone and reducing it with sodium borohydride. This method produced borate salts as byproducts, requiring a tedious removal process. In addition, (S)-Pro-Xylane shows superior biological activity compared to (R)-Pro-Xylane. Hence, with focus on the second step of the reduction reaction, the carbonyl reductase (CR) from Candida orthopsilosis Co 90-125 (CoCR13) was engineered in this study. Variant R129E/D210F showed a 2

Methanol is an ideal one-carbon (C1) feedstock for bioconversion into multicarbon value-added compounds. Biocatalytic approaches to methanol conversion provide sustainable and environmentally friendly alternatives to conventional methods. This process is facilitated by methanol-oxidizing enzymes, including alcohol oxidase (AOx). Here, we report an AOx from Pestalotiopsis fici (PfAOx) with the highest methanol oxidation activity and efficient heterologous expression compared to other AOxs. To investigate the bioconversion of a multicarbon compound (C4 chemical, 2-keto-4-hydroxybutyrate, 2-KHB)

This study presents the potential role of deep eutectic solvents (DESs) in a lipase-catalyzed hydrolysis reaction as a co-solvent in an aqueous solution given by a phosphate buffer. Ammonium salts, such as choline chloride, were paired with hydrogen bond donors, such as urea, 1,2,3-propanetriol, and 1,2 propanediol. The hydrolysis of p-nitrophenyl laureate was carried out with the lipase Candida antarctica Lipase B (CALB) as a reaction model to evaluate the solvent effect and tested in different DES/buffer phosphate mixtures at different % w/w. The results showed that two mixtures of different

Hypertrophic cardiomyopathy is one of the most common forms of genetic cardiomyopathy. Mavacamten is a first-in-class myosin modulator that was identified via activity screening on the wild type, and it is FDA-approved for the treatment of obstructive hypertrophic cardiomyopathy (HCM). The drug selectively binds to the cardiac β-myosin, inhibiting myosin function to decrease cardiac contractility. Though the drug is thought to affect multiple steps of the myosin cross-bridge cycle, its detailed mechanism of action is still under investigation. Individual steps in the overall cross-bridge cycle

The spatially separated immobilization of organometallic catalyst and enzymes on suitable carriers not only can reduce their mutual inhabitation but also can enhance their reusability. Here in this work, we present a hierarchical porous COFs (HP-TpBpy) that incorporated with [(Cp*RhCl2]2 to generate the metalized COF, Rh-HP-TpBpy. Thermolysin (TLN) is a microbial highly-priced thermostable metallo-endoprotease with complementary substrate specificity to those of proteases widely used in science and industry for protein digestion and milk-clotting. This study is the first to immobilize TLN on aminated superparamagnetic nanoparticles (Fe3O4@silica-NH2) aiming for higher stability, recoverability, reusability, and applicability in proteolysis and as a microbial rennet-like milk-clotting enzyme. The nanobiocatalyst developed (Fe3O4@silica-TLN) displays hydrolytic activity on a synthetic TLN substrate and, apparently, was fu

N-methylated tryptamines, such as the hallucinogenic natural products, psilocybin and N,N-dimethyltryptamine (DMT), are gaining interest from the medical community due to their potential as next generation treatments for mental health disorders. The clinical relevance of these compounds has driven scientists to develop biosynthetic production routes to a number of tryptamine drug candidates, and efforts are ongoing to expand and further develop these biosynthetic capabilities. To that end, we have further characterized the substrate preferences of two enzymes involved in tryptamine biosynthesi

Transaminases have shown the ability to catalyze the amination of a series of aliphatic and (hetero)aromatic α,α-difluorinated ketones with high stereoselectivity, thus providing the corresponding β,β-difluoroamines in high isolated yields (55-82%) and excellent enantiomeric excess (>99%). It was also observed that these activated substrates could be quantitatively transformed by employing a small molar excess of the amine donor since this amination process was thermodynamically favored. Selected transformations could be scaled up to 500 mg, showing the robustness of this methodology.

Kinases mediate cell signaling pathways by catalyzing protein phosphorylation. Irregularities in kinase activity are directly associated with disease conditions. Therefore, methods to identify substrates of a particular kinase are needed to understand signaling cascades in normal and diseased states. Photocrosslinking ATP analogs provide powerful tools to study kinases by covalently linking kinases with substrates. However, the involvement of UV light and nonspecific reactivity of current ATP-photocrosslinkers challenge kinase-substrate identification. We report here an affinity-based crosslin

Aldehydes are a class of carbonyl compounds widely used as intermediates in the pharmaceutical, cosmetic and food industries. To date, there are few fully enzymatic methods for synthesizing these highly reactive chemicals. In the present work, we explore the biocatalytic potential of an amino oxidase extracted from the etiolated shoots of Lathyrus cicera for the synthesis of value-added aldehydes, starting from the corresponding primary amines. In this frame, we have developed a completely chromatography-free purification protocol based on crossflow ultrafiltration, which makes the production

Deacetoxycephalosporin C synthase (DAOCS) is a nonheme iron(II) and 2-oxoglutarate (α-KG)-dependent oxygenase that catalyzes the oxidative ring expansion of penicillin N (penN) to deacetoxycephalosporin C (DAOC). Earlier reported crystal structures of DAOCS indicated that the substrate penicillin binds at the same site of succinate, leading to the proposal of the unusual "ping-pong" mechanism. However, more recent data provided evidence of the formation of ternary DAOCS·α-KG·penN complex, and thus DAOCS should follow the usual consensus mechanism of α-KG-dependent nonheme iron(II) oxygenases.

Microgels are an emerging class of "ideal" enzyme carriers because of their chemical and process stability, biocompatibility, and high enzyme loading capability. In this work, we synthesized a new type of permanently positively charged poly(N-vinylcaprolactam) (PVCL) microgel with 1-vinyl-3-methylimidazolium (quaternization of nitrogen by methylation of N-vinylimidazole moieties) as a comonomer (PVCL/VimQ) through precipitation polymerization. The PVCL/VimQ microgels were characterized with respect to their size, charge, swelling degree, and temperature responsiveness in aqueous solutions. P45

Human cytochrome P450 (P450) 11B2 catalyzes the formation of aldosterone, the major endogenous human mineralocorticoid. Aldosterone is important for the regulation of electrolyte homeostasis. Mutations and overexpression of P450 11B2 (also known as aldosterone synthase) can lead to hypertension, congestive heart failure, and diabetic nephropathy. The enzyme is therefore a target for drug development to manage these various disorders. P450 11B2 catalyzes aldosterone formation from 11-deoxycorticosterone through three distinct oxidation steps. It is currently unknown to which degree these reacti

Ongoing advances in chemical proteomic methods have facilitated detection and quantification of enzymatic activity, a highly informative parameter that is not captured in protein abundance measurements. However, some biological questions remain unanswered, since current gel- or LC-MS/MS-based detection methods suffer from limitations stemming from sample homogenization, signal-averaging, and an inherent bias toward abundant proteins. To address these shortcomings, we recently developed an activity-based proximity ligation (ADPL) platform to capture and quantify enzyme activity on the level of
