OBJECTIVE: To explore the application of collagen-binding assay for von Willebrand disease (vWD).
METHODS: We investigated the sensitivity and reproducibility of collagen-binding assays (CBA) of vWF using the enzyme-linked immunosorbent assay (ELISA) technique and compared CBA with 3 other assays for their ability to detect vWD.
RESULTS: It was found that the vWF: CBA level of plasma from patients with different types of vWD is lower than that from healthy donors, hemophilia and other bleeding disorders (P < 0.001). In general, the four assays have the capability to identify vWD from normal

A novel, label-free, fluorescent, turn-on sensor for biological thiol detection that uses highly fluorescent gold nanoclusters (AuNCs), prepared by a bovine serum albumin (BSA)-templated green synthetic route, has been developed. In this paper, we present an optical diagnostic assay consisting of a mixture of environmental-sensitive fluorescent dyes combined with multivariate data analysis for quantitative and qualitative examination of biological and clinical samples. The performance of the assay is based on the analysis of spectrum of the selected fluorescent dyes with the operational principle similar to electronic nose and electronic tongue systems. This approach has been successfully applied for monitoring of growing cell cultures and identification of gastrointestinal diseases in humans.

In order to understand precise biological roles of sulfur dioxide (SO(2)), reliable SO(2) donors, compounds that produce SO(2) under physiological conditions, are necessary. The design and development of 1-phenyl-benzosultine as an efficient SO(2) donor is reported. This compound undergoes cycloreversion to generate SO(2) upon dissolution in aqueous buffer at 37 °C with a yield of 89% and a half-life of 39 min and shows SO(2)-like biological activity in a DNA cleavage assay.

Five current methods of metallothionein (MT)-estimation in biological materials (Hg/TCA-, Cd/heme-, SH-assay, G 75-method and RIA) were compared. Uniform calibration was ensured by measuring the nitrogen-content of the standard-apoprotein after "Kjeldahl-digestion". Known amounts of rabbit standard Cd-MT I were recovered from identical rat S9-samples. In an overall rating, RIA and Cd/heme-assay performed best, whereas the other methods either underestimated (SH-, G 75) or overestimated (Hg/TCA) the 100%-expectation.

Botulism therapy relies on passive immunization with antitoxin. The mouse neutralization test is the only pharmacopeia assay to measure the potency of antitoxin preparations. Herein, we present an in vitro cell-based assay for the measurement of pharmaceutical type A antitoxin potency. Accuracy, reproducibility and compatibility with the mouse bioassay were demonstrated using different batches of standard antitoxin and toxin preparations. The established assay may substantially reduce the use of laboratory animals in the process of pharmaceutical antitoxin production.

Milk and serum were obtained from 50 mothers on the 6th day post partum and examined for inhibitory activity against bilirubin conjugation in rat liver slices. Neonatal serum bilirubin levels were also analysed at this time. There was no relation between the amount of inhibitory substance in breast milk or serum and the degree of neonatal hyperbilirubinaemia. It was observed that inhibitory activity increased in frozen breast milk but not in frozen autoclaved breast milk.
