Report From the International Conference on Incontinentia Pigmenti: Translating Discovery to Therapy.

Incontinentia pigmenti (IP) is a rare, X-linked dominant condition characterized by a perinatal rash as well as abnormalities of the central nervous system (CNS), eyes, hair, nails, and teeth. The CNS and ophthalmic manifestations are potentially severe and may lead to neurologic impairment and vision loss. Due to the rarity of IP, there remains a limited understanding of its physiology, and the guidelines for screening, clinical staging, and management of complications remain controversial. To address these gaps, a multidisciplinary, international conference on IP was held in Charlotte, North

The landscape of dementia research, diagnosis, treatment, and care in Latin America.

Latin America is undergoing rapid population aging alongside a rising burden of dementia. While the region holds substantial potential for dementia risk reduction, challenges remain, such as delayed diagnoses, limited access to specialized care and biomarker testing, persistent stigma, and deep-rooted structural inequities. To address these gaps and foster regionally informed solutions, the Alzheimer's Association convened the 2025 Alzheimer's Association International Conference (AAIC) Satellite Symposium in Lima, Peru, on May 14-15, in collaboration with the Global Brain Health Institute (GB

Knowledge translation for delirium superimposed on dementia care: a realist review.

BACKGROUND AND OBJECTIVES: People living with dementia are at higher risk of developing delirium during healthcare admission. While there have been advancements in delirium interventions, challenges persist in translating evidence-based knowledge into practice. This review aims to explore the contextual factors and underlying mechanisms of knowledge translation for preventing and managing delirium in people with dementia, and to understand how these processes contribute to intended outcomes. RESEARCH DESIGN AND METHODS: A realist synthesis was adopted to construct context-mechanism-outcome co

Pulsed focused ultrasound for seizure suppression: Mechanisms, models, and clinical translation.

Focused ultrasound (FUS) has emerged as a versatile, non-invasive therapeutic platform for drug-resistant epilepsy, offering both thermal effects for the permanent ablation of epileptogenic foci and non-thermal mechanical mechanisms for the reversible neuromodulation of neural activity, all without the need for surgical implantation. Pulsed focused ultrasound (pFUS) achieves seizure control through seizure suppression via non-thermal mechanical mechanisms. At the cellular level, pFUS modulates mechanosensitive ion channels, and may interrupt astrocyte-mediated excitatory signaling pathways. At

Bibliometric and graphical analysis of ferroptosis and aging research: Trends, gaps, and future directions.

Over the past 12 years, a significant body of evidence derived from extensive research has underscored the pivotal involvement of ferroptosis in the mechanisms underlying aging. Despite the growing body of literature on this topic, there remains a paucity of analytical and descriptive studies that explore its trajectory, key research directions, current trends, primary focal points, and future outlooks. This research endeavors to provide an exhaustive overview of the advancements in understanding the relationship between ferroptosis and aging over the past 12 years. The dataset utilized in thi

When the science alone is not enough: embracing our responsibility as science communicators.

The US biomedical research enterprise has led the world in driving medicine forward but now faces unprecedented threats. Systematic efforts to defund and dismantle the academic research infrastructure, compounded by misinformation and questionable public support for science, have already impeded innovation and discouraged students from pursuing careers in research and healthcare, which ultimately will harm patients. This moment calls on us, as scientists and physicians, to effectively communicate the relevance of our research to the public and elected officials and ensure that the value of bas…

Best Practice Guidelines for Collecting Microbiome Samples in Research Studies.

Microbiome research has the potential to provide valuable information regarding the complex relationship between microbial communities and the human body. To help facilitate the translation of this potentially revolutionary research to clinical medicine, common guidelines and best practices are necessary. These guidelines should be based on the key findings of recent research in the field and address the primary areas of discrepancy in the previously published literature. Based on this review's findings, future microbiome research should adopt a consistent and clear approach to nomenclature, u

Decoding orofacial pain: a translational review of mechanisms and novel therapies.

Orofacial pain encompasses a diverse group of disorders that pose a significant clinical challenge, often progressing from acute to chronic conditions due to a disconnect between clinical diagnosis and underlying pathophysiology. This review provides a synthesis of the mechanisms driving this transition and evaluates emerging therapeutic strategies that target them. First, we dissect the pathophysiological mechanisms of acute pain, including peripheral sensitization and transient central sensitization; and then detail how these processes can become maladaptive, leading to chronic pain states s

An international perspective on the future of systemic sclerosis research.

Systemic sclerosis (SSc) remains a challenging and enigmatic systemic autoimmune disease, owing to its complex pathogenesis, clinical and molecular heterogeneity, and the lack of effective disease-modifying treatments. Despite a century of research in SSc, the interconnections among microvascular dysfunction, autoimmune phenomena and tissue fibrosis in SSc remain unclear. The absence of validated biomarkers and reliable animal models complicates diagnosis and treatment, contributing to high morbidity and mortality. Advances in the past 5 years, such as single-cell RNA sequencing, next-generati
