Molecular surveillance of bat flies (Diptera: Nycteribiidae and Streblidae) and other ectoparasites in Ratchaburi, Thailand: Unraveling host associations and coronavirus transmission dynamics in the context of zoonotic spillover risk.

BACKGROUND: Bats act as hosts for various ectoparasites, including bat flies, bugs, fleas, ticks, and mites, which play crucial roles in the transmission of bat-borne pathogens. As obligate blood-feeding parasites, these ectoparasites can serve as direct vectors or indirectly influence pathogen dynamics in bat populations. In Thailand, molecular studies on bat ectoparasites are limited, with scarce data on their diversity, distribution, and role in zoonotic pathogen transmission. This study aims to provide the first molecular characterization of bat flies and other ectoparasites in Ratchaburi,

Dynamics of digestive proteolytic system during blood feeding of the hard tick Ixodes ricinus.

BACKGROUND: Ticks are vectors of a wide variety of pathogens causing severe diseases in humans and domestic animals. Intestinal digestion of the host blood is an essential process of tick physiology and also a limiting factor for pathogen transmission since the tick gut represents the primary site for pathogen infection and proliferation. Using the model tick Ixodes ricinus, the European Lyme disease vector, we have previously demonstrated by genetic and biochemical analyses that host blood is degraded in the tick gut by a network of acidic peptidases of the aspartic and cysteine classes. Prevalence of Borrelia miyamotoi and Neoehrlichia mikurensis in a cohort of tick-exposed individuals and population controls in Denmark 2002-2021.

Borrelia miyamotoi and Neoehrlichia mikurensis are recently discovered tick-borne pathogens. The aim of this study was to ascertain the extent of transmission among high-risk groups and healthy controls. The study design is retrospective cohort study. Blood from a total of 1180 individuals, collected between 2002-2021: (i) 180 tick-exposed individuals, (ii) 400 immunocompromised individuals and (iii) 600 blood donors was included. Infection was examined by a protein array (B. miyamotoi) and PCR. Neoehrlichia mikurensis DNA was detected in only one individual with Lyme neuroborreliosis (LNB) wi

Molecular survey of bovine vector-borne pathogens in Cebu, Philippines.

Vector-borne diseases (VBDs) continue to threaten the worldwide livestock industry, but comprehensive epidemiological surveys on such diseases have not been conducted in the Philippines. In the present study, we screened 408 bovine blood samples from 9 areas in Cebu, Philippines, for various VBD pathogens using specific PCR assays. The results revealed prevalences of 54.7, 15.4, 10.0, and 12.0% for Anaplasma spp., Babesia bigemina, Babesia bovis, and Trypanosoma (Tr.) theileri, respectively. In contrast, none of the samples were positive for Trypanosoma (Tr.) evansi, Theileria (Th.) orientalis

Vascular traffic control of neutrophil recruitment to the liver by microbiota-endothelium crosstalk.

During bloodstream infections, neutrophils home to the liver as part of an intravascular immune response to eradicate blood-borne pathogens, but the mechanisms regulating this crucial response are unknown. Using in vivo imaging of neutrophil trafficking in germ-free and gnotobiotic mice, we demonstrate that the intestinal microbiota guides neutrophil homing to the liver in response to infection mediated by the microbial metabolite D-lactate. Commensal-derived D-lactate augments neutrophil adhesion in the liver independent of granulopoiesis in bone marrow or neutrophil maturation and activation…

Antibiotic use in Thailand: quantifying impact on blood culture yield and estimates of pneumococcal bacteremia incidence.

No studies have quantified the impact of pre-culture antibiotic use on the recovery of individual blood-borne pathogens or on population-level incidence estimates for Streptococcus pneumoniae. We conducted bloodstream infection surveillance in Thailand during November 2005-June 2008. Pre-culture antibiotic use was assessed by reported use and by serum antimicrobial activity. Of 35,639 patient blood cultures, 27% had reported pre-culture antibiotic use and 24% (of 24,538 tested) had serum antimicrobial activity. Pathogen isolation was half as common in patients with versus without antibiotic us…

Using Digital PCR to Unravel the Occurrence of Piroplasmids, Bartonella spp., and Borrelia spp. in Wild Animals from Brazil.

Piroplasmids (Babesia spp., Rangelia spp., Theileria spp., Cytauxzoon spp.) are tick-borne apicomplexan protozoa that infect, depending on the species, erythrocytes and leucocytes in a wide variety of mammals and birds. The genera Bartonella and Borrelia include vector-borne bacteria that can infect and cause disease in both animals and humans. Detection of hemotropic bacteria and piroplasmids in wild animals is often challenging due to low bacteremia or parasitemia. Digital (d)PCR has proven to be an effective modality for the detection and quantification of DNA of hemotropic pathogens with l

Persistence of dengue serotype 2 viral RNA in blood cells of a returned traveler with dengue fever.

Dengue virus (DENV) is one of the most significant vector-borne pathogens worldwide. In this report, we describe clinical features and laboratory detection of dengue in a 45-year-old traveler to Nicaragua on return home to the United States in 2019. Clinical presentation was mild, with rash, headache, and fatigue, with only low-grade transient fever. Infection dynamics were documented by serology and PCR of serially collected body fluids. DENV serotype 2 was detected in whole blood 1 day after symptoms emerged, with viral RNA isolated to the red cell fraction, and remained detectable through d

Molecular evidence of co-infections with Anaplasma and Theileria species in domestic sheep and goats from Alentejo, Portugal.

Anaplasma spp. and piroplasmids are tick-borne pathogens with worldwide distribution, affecting domestic animals, wildlife, and humans. Although several tick-borne species have relevance in animal health and can cause economic losses in livestock farms, little is known about the epidemiological situation in small ruminants in Portugal. We aimed to assess the infection prevalence, molecular characterization, co-infection patterns and risk factors of Anaplasma spp. and piroplasmids in sheep and goat farms from the Alentejo region (south-central Portugal). A total of 290 blood samples collected i

Xenodiagnosis in the wild: A methodology to investigate infectiousness for tick-borne bacteria in a songbird reservoir.

A crucial factor to predict the persistence and spread of infections in natural systems is the capacity of reservoir hosts to maintain the infection and transmit it to others. This is known to greatly vary within and between species and through time, although the latter part of the variation is often less well understood in the wild. Borrelia garinii is one of the causal agents of Lyme disease in humans and is transmitted among avian hosts by the hard tick Ixodes ricinus. Great tits are known to be a reservoir in Europe for B. garinii. For tick-borne pathogens like B. garinii, infectiousness o

Identification and genetic characterization of Piroplasmida and Anaplasmataceae agents in feeding Amblyomma variegatum ticks from Benin.

In many African countries including Benin, the reluctance of some livestock owners to blood collection from their cattle makes epidemiological surveys cumbersome and prevents regular monitoring of tick-borne diseases. In the present study, Amblyomma variegatum ticks were used to find out more about bovine tick-borne pathogens. DNA extracts from 910 adult ticks collected off cattle in North East Benin were examined for Babesia bigemina, B. bovis, Theileria taurotragi, T. annulata, T. orientalis, T. parva, T. mutans, Anaplasma marginale and Ehrlichia ruminantium using pathogen-specific PCR assay

Infection control basis for recommending one-time use of sterile syringes and aseptic procedures for injection drug users.

Persons who inject drugs are at increased risk for many infectious diseases, including HIV. Reuse of syringes and needles and other equipment used to prepare and inject drugs increases the risk of infection with blood-borne and other pathogens. According to standard infection control guidelines, needles and syringes and their contents fall within the critical category of patient contact because they enter the normally sterile vascular space. Medical equipment considered critical should be treated or prepared in a manner that destroys vegetative and spore-forming microbes. The simplest way to e
