The timely diagnosis and treatment of post-infectious glomerulonephritis (PIGN) is currently limited by the erased and low-symptom nature of the disease, which leads to the search for informative biological markers of the disease, which can be used as immunological indicators of blood and urine. The study was carried out in order to establish the characteristic changes in the immunological parameters of blood and urine in patients with PIGN. The study included 60 patients with PIGN from among the patients, hospitalized in the nephrology department of the Republican Clinical Hospital of Health

CD3zeta is a subunit of the T-cell antigen receptor (TCR) complex required for its assembly and surface expression that also plays an important role in TCR-mediated signal transduction. We report here a patient with T(-)B(+)NK(+) severe combined immunodeficiency (SCID) who was homozygous for a single C insertion following nucleotide 411 in exon 7 of the CD3zeta gene. The few T cells present contained no detectable CD3zeta protein, expressed low levels of cell surface CD3epsilon, and were nonfunctional. CD4(+)CD8(-)CD3epsilon(low), CD4(-)CD8(+)CD3epsilon(low), and CD4(-)CD8(-)CD3epsilon(low) ce

INTRODUCTION: Associations of disturbances in innate and adaptive immunity during the clinical course of schizophrenia have been found in a number of studies. Yet, the relationship of immune parameters and systemic inflammation in relation to the clinical course of the disease and its prognosis, remains poorly understood, which highlights an interesting topic for further research. The goal of this study was to research the immuno-inflammatory changes in patients with clinical continuous and episodic paranoid schizophrenia, to assess the pathogenetic significance of these changes.

Lymphoma is the most common tumour of domestic cats, developing most frequently in the small intestine. Feline small intestinal lymphoma predominantly demonstrates a T-cell immunophenotype identified by standard immunopositivity for T cells with CD3 or immunopositivity for B cells with CD20. In contrast, a wide spectrum of immunohistochemical antibodies are applied in humans to diagnose the various specific lymphoma subtypes according to the WHO classification. Our aim was to augment our knowledge of immunophenotypes in feline non-B-cell lymphomas forming macroscopic masses in the intestinal t

We report a case of T cell prolymphocytic leukemia (T-PLL) involving blast transformation. At the initial diagnosis, most peripheral blood cells demonstrated proliferation of indolent T cell small cell variants, i.e., small to medium prolymphocytes with inconspicuous nucleoli and a normal karyotype. These cells were positive for surface CD4, CD5, and CD7, and cytoplasmic CD3, but negative for surface CD3 and CD8 and cytoplasmic terminal deoxynucleotidyl transferase (TdT). The T cell receptor (TCR) Cβ1 gene was rearranged in the cells. Large prolymphocytes with prominent nucleoli, irregular nuc

Phenotypic expression of inflammatory cells in liver and hepatic lymph nodes (HLN) has been examined in lambs experimentally infected with Dicrocoelium dendriticum using immunohistochemical techniques. Thirty-two lambs, 12 infected with 1,000 D. dendriticum metacercariae, 12 with 3,000, and 8 controls were used. Half the lambs in each group were slaughtered on days 60 and 180 post-infection (p.i.), respectively. Primary antibodies (Abs) against T cell epitopes (CD3+, CD4+, CD8+ and WC1+ gammadelta), B cell epitopes (CD79alphacy+, CD45R+), immunoglobulin (IgG)-bearing plasma cells, macrophages

Barbiturates are frequently used for the treatment of intracranial hypertension after brain injury but their application is associated with a profound increase in the infection rate. The mechanism of barbiturate-induced failure of protective immunity is still unknown. We provide evidence that nuclear factor of activated T cells (NFAT), an essential transcription factor in T cell activation, is a target of barbiturate-mediated immunosuppression in human T lymphocytes. Treatment of primary CD3+ lymphocytes with barbiturates inhibited the PMA and ionomycin induced increase in DNA binding of NFAT,

We have previously identified a subset of common variable immunodeficiency (CVID) patients with defective T cell function associated with impaired activation of the TCR-dependent tyrosine phosphorylation cascade. Here we have assessed the structural and functional integrity of the principal components involved in coupling the TCR/CD3 complex to intracellular tyrosine kinases in two of these patients. We show that ZAP-70 fails to bind the signaling-competent CD3zeta tyrosine phosphorylation isoform and to become activated following TCR engagement, suggesting that defective recruitment of ZAP-70…

To develop an effective immunotherapy for B-lineage acute lymphoblastic leukemia (ALL), bispecific monoclonal antibodies (bsAb) were raised by cell fusion of two hybridoma cell lines secreting CD3 and CD19 antibodies. The resulting bispecific antibody contains two different specificities within a single antibody molecule. One binding site (CD3) activates the T cells via the T cell receptor complex, whereas the second binding site (CD19) targets the cytolytic T cells to malignant B cells. Leukemic blasts from children with B-lineage ALL showed stable and strong CD19 expression. CD3xCD19 bsAb we

By using several monoclonal antibodies (mAb) reacting either with the constant or variable regions of the T cell receptors (TcR) αβ and γδ or various CD molecules, differences between two clinically related entities e.g. T cell acute lymphoblastic leukemia (T-ALL) and lymphoblastic lymphoma (T-LL) have been demonstrated. We studied a panel of fifteen T-ALL and fifteen T-LL because of their cell surface expression of the CD3-TcR molecules. The results indicated that TcR γδ is more frequently expressed in T-ALL (10 out of the 15 patients tested) than TcR αβ. This is in contrast to the results ob

BACKGROUND: Patients with relapsed/refractory multiple myeloma (RRMM) experience poor outcomes following multiple lines of therapy. Available evidence has largely pooled disparate immunotherapy platforms. The present systematic review and meta-analysis was performed to describe, separately by target, the efficacy and toxicity spectra of BCMA×CD3-directed, GPRC5D×CD3-directed, and dual-target bispecific antibody therapy in RRMM, in order to inform toxicity monitoring and supportive-care planning.
METHODS: The Cochrane Library, Embase, Web of Science, and PubMed were systematically searched up

Chronic neurological condition Neuropathic pain is marked by sustained neuronal sensitization. Regulatory T cells are crucial for preserving immune homeostasis. however, their stability and suppressive activity are often compromised within inflammatory environments generated after nerve injury, limiting their ability to restrain neuroinflammation. This study sought to determine whether genetically modified regulatory T cells expressing an interleukin-6 receptor-CD3ζ fusion receptor could enhance Treg persistence, functional capacity, and therapeutic benefit in a rat chronic constriction injury

BACKGROUND: Gastric cancer is a leading cause of cancer-related mortality, with limited efficacy of immunotherapies due to intratumoral heterogeneity, an immunosuppressive microenvironment, and antigen-loss escape. The optimization of chimeric antigen receptor (CAR) T-cell therapy targeting PD-L1 and Claudin18.2 (CLDN18.2) is crucial for improving the antitumor response.
METHODS: We performed a stepwise optimization of CAR designs targeting PD-L1 and CLDN18.2, focusing on the costimulatory domains and intracellular signaling architecture. We engineered monospecific PD-L1 and CLDN18.2 CAR-T ce

INTRODUCTION: Autoimmune gastritis (AIG) is a chronic immune-mediated disease with potential for serious clinical consequences, yet early identification remains challenging. We aimed to assess whether deep oxyntic gland intraepithelial CD3 + T lymphocytosis may serve as a predictive histological marker of progression from potential -i.e., parietal cell antibody-positive patients without baseline gastric atrophy- to overt AIG.
METHODS: We prospectively enrolled 45 adult parietal cell antibody-positive patients without histological evidence of mucosal atrophy between 2020 and 2022. All underwen

Infections occur in up to 44% of trial participants treated with CD3/CD20 bispecific antibodies (BsAb), but real-world data are limited. In this study of 48 real-world R/R-NHL patients receiving commercial BsAbs, 50% and 23% experienced any-grade and grade ≥3 infections, respectively. The cumulative number of infections per patient at 12 months was 1.3 (95% CI: 0.81-2.1). Severe infections were mostly bacterial. Only 1 grade 5 event occurred, and no COVID-19-related deaths were observed. Severe neutropenia, lymphopenia, and hypogammaglobulinemia occurred in 39.6, 83.3, and 64.1% of patients, r

Cytokine release syndrome remains a critical challenge for clinical use of bispecific T-cell engagers. We present the preclinical development of a novel B-cell maturation antigen × CD3 bispecific antibody with the aim of reducing cytokine release while maintaining potent efficacy in the treatment of multiple myeloma. Based on the Fabs-in-tandem (FIT) geometry, bispecific molecules with two target arms in cis-configuration were constructed. A panel of anti-CD3 mAbs with varying affinities was generated, and the impact of binding arm geometry, valency, and anti-CD3 affinity on the T-cell engager

BACKGROUND: Rabbit anti-thymocyte globulin (r-ATG) is a thymocyte depleting agent for the induction of immunosuppression in kidney transplant recipients (KTRs). There is little data comparing a level of CD3+ T-cell suppression for optimal graft outcomes with minimized infectious risk.
METHODS: KTRs from January 2020 to January 2024 were included if they met these criteria: (1) < 21 years of age at transplantation, (2) no previous receipt of a solid organ or hematopoietic stem cell transplant, (3) completion of a r-ATG-based induction protocol, (4) had absolute CD3+ T-cell counts quantified at

BACKGROUND: CD3 + CD20 + T cells (TB cells) are a subset of lymphocytes in the human body that are associated with inflammation. They originate from T cells interacting with B cells, and their levels are abnormally elevated in individuals with immune disorders, as well as in some cancer patients. The interplay between tumor immunity and inflammation is intricate, yet the specific involvement of TB cells in local tumor immunity remains uncertain, with limited research on their subtypes.
METHODS: Lung cancer surgical samples were stained using multi-color immunofluorescence to study the subtype

A cytotoxicity assay was developed to measure the potency of GP101, a single chain diabody. GP101 is comprised of two linked Fv domains, with one that binds CD3 (expressed on T cells), and the other that binds CD19 (expressed on B cells). GP101 directs CD3 positive T lymphocytes to CD19 positive B lymphocytes. This immunotherapy redirects CD3+ T cells to CD19-expressing B-cell malignancies, enabling T-cell-mediated tumor cell lysis. We developed a GMP cell-based potency assay to satisfy the FDA requirements. The potency assay uses a cytotoxic T cell line, and a B cell lymphoma cell line as the
