Human intestinal organoid-derived epithelium reveals donor-dependent antiviral responses to therapeutics against enteric picornaviruses.

Human picornavirus infections remain without approved antiviral therapies. Antiviral testing performed in immortalised cell lines often fails to predict antiviral efficacy in vivo and subsequently clinical outcomes. Here, we assessed how antiviral responses to clinically approved compounds differ between immortalised intestinal cell lines and a physiologically relevant human intestinal organoid-derived epithelium (HIE) model. Posaconazole, itraconazole, ribavirin, remdesivir, and pocapavir, compounds with reported anti-picornaviral activity in cell lines, were evaluated against enterovirus A71

Macropinocytosis and Fast Endophilin-Mediated Endocytosis Mediate Absorption of Garlic Chive-Derived Vesicle-like Nanoparticles in Human Intestinal Epithelial Cells.

Dietary extracellular vesicles (EVs) or vesicle-like nanoparticles (VLNs) have been shown to exert beneficial functions in a wide range of d … arlic chive-derived VLNs (GC-VLNs) using C57BL/6J mice and a human intestinal epithelial cell line, Caco-2 cells. We found that orally administered GC-VLNs crossed the epithelial layer of the small intestine and entered the underlying lamina propria. GC-VLNs were taken up and transported across the fully differentiated Caco-2 epithelial monolayer. Proteins and …

Bacteriocins attenuate Listeria monocytogenes-induced intestinal barrier dysfunction and inflammatory response.

Bacteriocins have the potential to effectively improve food-borne infections or gastrointestinal diseases and hold promise as viable alterna … t causing any cytotoxicity. Bacteriocins inhibited the adhesion and invasion of L. monocytogenes on Caco-2 cells, lactate dehydrogenase (LDH), trans-epithelial electrical resistance (TEER), and cell migration showed that bacteriocin improved the permeability of Caco-2 cells. These results were attributed to the promotion of tight junction proteins (TJP) assemb …

Microvesicles released from Giardia intestinalis disturb host-pathogen response in vitro.

Giardia intestinalis (G.I), is an anaerobic protozoan and the aetiological agent of giardiasis, a diarrhoea present worldwide and associated … rdiasis. MVs from log phase were able to increase the attachment of G. intestinalis trophozoites to Caco-2 cells. Moreover, MVs from G. intestinalis could be captured by human immature dendritic cells, resulting in increased activation and allostimulation of human dendritic cells. Methodological Considerations in Small-Sample Multitask QSPR Modeling for PAMPA Permeability.

Formanek et al. reported a multitask PAMPA dataset of 143 drugs and drug candidates and compared multiple molecular descriptors and regression models for predicting passive membrane permeability. This Letter further examines two conclusions drawn from those results, with the aim of qualifying rather than overturning them. First, the external test set contains only 29 compounds, making small differences in test-set R2 difficult to interpret reliably. The test-set R2 and correlation metrics can also diverge because R2 is sensitive to scale and bias, whereas correlation primarily reflects orderin

Biosafety measures for Alicyclobacillus spp. strains across various levels of biohazard.

Alicyclobacillus bacteria are important contaminants in the beverage industry because their spores remain in the product after usual pasteurization. At the same time, their impact on human health has yet to be characterized, as it is generally assumed to be low or non-existent. However, these bacteria are causing quality concerns mainly due to odor and taste changes of the product. Since potential health effects are not precisely known, an experimental assessment was performed, including a biosafety assessment of six viable and non-viable vegetative and spore forms of Alicyclobacillus spp. str

Detecting host responses to microbial stimulation using primary epithelial organoids.

The intestinal epithelium is constantly exposed to microbes residing in the lumen. Traditionally, the response to microbial interactions has been studied in cell lines derived from cancerous tissues, e.g. Caco-2. It is, however, unclear how the responses in these cancer cell lines reflect the responses of a normal epithelium and whether there might be microbial strain-specific effects. To address these questions, we derived organoids from the small intestine from a cohort of healthy individuals. Culturing intestinal epithelium on a flat laminin matrix induced their differentiation, facilitatin

Cell penetrating peptides-functionalized Licochalcone-A-loaded PLGA nanoparticles for ocular inflammatory diseases: Evaluation of in vitro anti-proliferative effects, stabilization by freeze-drying and characterization of an in-situ forming gel.

Licochalcone-A (Lico-A) PLGA NPs functionalized with cell penetrating peptides B6 and Tet-1 are proposed for the treatment of ocular anti-inflammatory diseases. In this work, we report the in vitro biocompatibility of cell penetrating peptides-functionalized Lico-A-loaded PLGA NPs in Caco-2 cell lines revealing a non-cytotoxic profile, and their anti-inflammatory activity against RAW 264.7 cell lines. Given the risk of hydrolysis of the liquid suspensions, freeze-drying was carried out testing different cryoprotectants (e.g., disaccharides, alcohols, and oligosaccharide-derived sugar alcohol)

Mercaptonicotinic acid activated thiolated chitosan (MNA-TG-chitosan) to enable peptide oral delivery by opening cell tight junctions and enhancing transepithelial transport.

Recent advances in peptide delivery and nanotechnology has resulted in emergence of several non-parenteral administration routes that replace subcutaneous injections associated with patient discomfort. Thiolated biopolymers are relatively new materials being explored to enhance mucoadhesivity and permeability in these efforts, yet their pH dependent reactivity remains an obstacle. This work focussed on improving the mucoadhesivity of thiolated chitosans by activating them with mercaptonicotinic acid, in a bid to create a novel thiomerized chitosan that can open cell tight junctions for applica

Characterization of Pro-Drug Metabolism and Drug Permeability Kinetics in a Microphysiological In Vitro Model of the Human Small Intestinal Barrier Incorporating Mucus-Generating Cells Coupled with LC-MS/MS Analysis.

A micro-physiological model of the human small intestinal barrier incorporating mucus-generating cells has been developed. The established barrier was utilized to assess the absorption kinetics of three selected therapeutic compounds: Doxycycline, Ganciclovir, and its prodrug Valganciclovir, following the implementation of a thorough LC-MS/MS validation protocol. The co-culture of Caco-2 cells, representing absorptive enterocytes, and HT-29 MTX cells, modeling goblet cells, enabled the in-situ generation of a sufficiently thick mucus layer covering the entire cell monolayer. The presence of HT…

Construction of a multiple fluorescence labelling system for use in co-invasion studies of Listeria monocytogenes.

BACKGROUND: Existing virulence models are often difficult to apply for quantitative comparison of invasion potentials of Listeria monocytogenes. Well-to-well variation between cell-line based in vitro assays is practically unavoidable, and variation between individual animals is the cause of large deviations in the observed capacity for infection when animal models are used. One way to circumvent this problem is to carry out virulence studies as competition assays between 2 or more strains. This, however, requires invasion-neutral markers that enable easy discrimination between the different s

Effect of aging on the release of additives and the toxicity of microplastics.

Microplastics (MPs) that contaminate water and food persist for a certain time in the environment and undergo an aging process that could affect their toxicity. The present study evaluates the effect of aging on the chemical composition of MPs and their toxicity. Two commercial plastics (PET and PVC) were aged under artificial and natural conditions. Their surface composition, additive contents and bioaccessibility were analyzed before and after the treatment. Toxicity was evaluated by exposing an intestinal cell model (Caco-2/HT29-MTX/THP-1) to aged and non-aged PVC MPs (50-100 µg/mL, median

Early ADME and Preliminary Toxicity Studies of I-152, an N-Acetyl-l-Cysteine/S-Acetylcysteamine Conjugate.

This study aimed to estimate the ADME properties and safety of I-152, a conjugate of N-acetyl-l-cysteine (NAC) and S-acetylcysteamine (also known as S-acetyl-β-mercaptoethylamine; SMEA), linked by an amide bond. Its potent antioxidant and pro-glutathione effects make it of interest for a range of conditions linked to oxidative stress, such as infectious diseases and inflammation. I-152 was characterized in vitro for its stability in plasma, liver microsomes, and hepatocytes; its protein binding; and its AB BA (apical-to-basolateral and basolateral-to-apical) permeability using Caco-2 cells. De…

Use of In silico Methodologies to Predict the Bioavailability of Oral Suspensions: A Regulatory Approach.

BACKGROUND: Oral suspensions are heterogeneous disperse systems, and the particle size distribution, crystalline form of the dispersed solid, and composition of the formulation can be listed as parameters that control the drug dissolution rate and its bioavailability. OBJECTIVE: The aim of this work was to develop a discriminative dissolution test, which, in association with in silico methodologies, can make it possible to safely anticipate bioavailability problems. METHODS: Nimesulide and ibuprofen (BCS class II) and cephalexin (BCS class I) oral suspensions were studied. Previously, solid-
