Lens-specific conditional knockout of Msx2 in mice leads to ocular anterior segment dysgenesis via activation of a calcium signaling pathway.

Ocular anterior segment dysgenesis (ASD) is a failure of normal development of anterior structures of the eye, leading to lens opacification … t, while other eye structures were not significantly affected. Multiple lines of evidence show that calcium signaling pathways are involved in this pathogenesis. Our study demonstrates that Msx2 plays an essential role in lens development by activating a yet undetermined calcium signaling pathway.

Inhibition of calcium-calmodulin kinase restores nitric oxide production and signaling in submandibular glands of a mouse model of salivary dysfunction.

Nitric oxide is an intracellular and diffusible messenger of neurotransmitters involved in salivary secretion, as well as an inflammatory mediator in salivary gland diseases. It is synthesized by three different isoforms of nitric oxide synthase (NOS), each subject to a fine transcriptional, post-transcriptional and/or post-translational regulation. Our purpose was to study the possible mechanisms leading to NOS downregulation in submandibular glands of normal mice and in the nonobese diabetic (NOD) mouse model of salivary dysfunction with lower NOS activity. NOS activity and cGMP accumulation

Combined calcium chloride and heat treatment delays softening of fresh-cut jackfruit by activating the Ca2+ signaling-NAC29 transcriptional regulatory module.

Fresh-cut jackfruit is highly susceptible to rapid softening during storage, representing a major quality deterioration that limits its commercial distribution. This study elucidated the potential mechanisms by which combined calcium chloride and heat treatment (CH) mitigates softening in fresh-cut jackfruit. CH treatment mitigated cell wall degradation, inhibited the accumulation of water-soluble pectin (WSP), and delayed the degradation of chelator-soluble pectin (CSP) and cellulose, while significantly suppressing the activities of key cell wall-degrading enzymes, including polygalacturonas

GAS5 mediates notch signaling to exacerbate calcium oxalate-induced renal injury through the NF-κB/NLRP3 axis by promoting macrophage polarization.

The contribution of macrophage polarization dysregulation to calcium oxalate (CaOx) nephrolithiasis remains poorly defined. Here, we identify the long non-coding RNA growth arrest-specific transcript 5 (GAS5) as a critical driver of pro-inflammatory M1 macrophage polarization through the Notch/NF-κB/NLRP3 axis, thereby aggravating CaOx-induced renal injury. In human macrophages, calcium oxalate monohydrate (COM) stimulation upregulates GAS5 expression, which functions as a competing endogenous RNA by sequestering miR-449c-5p, leading to derepression of Notch1 and amplification of downstream in

Dual Roles of Voltage-gated Calcium Channels and γ-Aminobutyric Acid-mediated Signaling in Modulating Neurotensin Receptor Type 2-induced Antinociception.

BACKGROUND: The potential to mitigate pain by targeting a single receptor while simultaneously modulating peripheral and spinal circuits, offers an exciting nonopioid therapeutic strategy. Neurotensin receptor type 2 (NTSR2) is a promising yet underexplored pathway for nonopioid analgesia. The authors investigated the antinociceptive effects of NTSR2 activation and its mechanisms in rodent models of perioperative and chronic pain. METHODS: Using NT79, a selective NTSR2 agonist, the authors assessed pain behaviors in male and female rats and mice. Animals were randomly assigned to receive sali

Tellurium nanoparticles produced by laser ablation induce selective anticancer effects via ROS-mediated apoptosis and calcium signaling pathways: In vitro screening.

Over the past decade, increasing attention has been directed toward the development and evaluation of novel anticancer agents based on nanot … y of L-929 fibroblasts, but only after 48 h of exposure. We further demonstrate that TeNPs activate calcium signaling pathways in a dose-dependent manner in both cancerous and non-cancerous cells. However, the effective concentration (EC50, half-maximal effective concentration) and the amplitude and morphology of calcium responses varied considerably between cell t …

P2RX1 promotes mitochondrial apoptosis via calcium/CaM KII-mediated suppression of PI3K/Akt signaling in Philadelphia chromosome-positive acute lymphoblastic leukemia.

INTRODUCTION: Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) continues to pose a major clinical challenge. Although tyrosine kinase inhibitor (TKIs)-based regimens form the cornerstone of therapy, treatment outcomes are compromised by difficulties in achieving remission, high relapse rates, and the critical issue of drug resistance. This study aims to elucidate the role of the purinergic receptor P2RX1 in the pathogenesis of and therapeutic response in Ph+ ALL. METHODS: By analyzing P2RX1 expression levels in an online patient database and its association with prognos

The tryptophan-aspartate (WD) repeat domain of bovine Coronin-1A promotes mycobacterial survival by inhibiting calcium signaling-mediated phagosome-lysosome fusion.

Bovine tuberculosis is a chronic consumptive zoonosis, causing significant economic losses and critical public health risks. Coronin-1A is a host cytoskeleton-associated protein that is crucial for understanding the inhibition of phagosome-lysosome fusion by Mycobacterium tuberculosis (M.tb) to evade host innate immune clearance. However, the involvement of bovine Coronin-1A (bCoronin-1A) in M.tb infection and whether it can be manipulated so as to enhance host resistance against bovine tuberculosis remains to be seen. Here, we explored the role of bCoronin-1A in phagosome-lysosome fusion in M

Effects of exogenous hydrogen sulfide and honokiol intervention on the proliferation, apoptosis, and calcium signaling pathway of rat enteric glial cells.

Hydrogen sulfide (H2S) is a gaseous signaling molecule that influences digestive and nervous system functions. Enteric glial cells (EGCs) are integral to the enteric nervous system and play a role in regulating gastrointestinal motility. This study explored the dual effects of exogenous H2S on EGCs and the influence of apoptosis-related pathways and ion channels in EGCs. We also administered honokiol for further interventional studies. The results revealed that low-concentration H2S increased the mitochondrial membrane potential (MMP) of EGCs, decreased the whole-cell membrane potential, downr

NCX1 disturbs calcium homeostasis and promotes RANKL-induced osteoclast differentiation by regulating JNK/c-Fos/NFATc1 signaling pathway in multiple myeloma.

Although several types of calcium channels abnormalities have been shown to promote myeloma bone disease (MBD), the relationship between Na+/Ca2+ exchanger 1 (NCX1) and MBD remains unexplored. Here, we examined the role of NCX1 in the development of multiple myeloma (MM), with a special focus on the underlying effects involved osteoclast differentiation. Firstly, we detected NCX1 protein highly expressed in BM tissues of MM patients, and its expression was positively correlated with serum calcium and the percentage of BM CD138+ cells. In vitro, NCX1 suppression with the inhibitor KB-R7943 redu

Design of an Imaging Probe to Monitor Real-Time Redistribution of L-type Voltage-Gated Calcium Channels in Astrocytic Glutamate Signaling.

PURPOSE: In the brain, astrocytes are non-excitable cells that undergo rapid morphological changes when stimulated by the excitatory neurotransmitter glutamate. We developed a chemical probe to monitor how glutamate affects the density and distribution of astrocytic L-type voltage-gated calcium channels (LTCC). PROCEDURES: The imaging probe FluoBar1 was created from a barbiturate ligand modified with a fluorescent coumarin moiety. The probe selectivity was examined with colocalization analyses of confocal fluorescence imaging in U118-MG and transfected COS-7 cells. Living cells treated with 5

Melatonin Attenuates Calcium Deposition from Vascular Smooth Muscle Cells by Activating Mitochondrial Fusion and Mitophagy via an AMPK/OPA1 Signaling Pathway.

Mitochondrial fusion/mitophagy plays a role in cardiovascular calcification. Melatonin has been shown to protect against cardiovascular disease. This study sought to explore whether melatonin attenuates vascular calcification by regulating mitochondrial fusion/mitophagy via the AMP-activated protein kinase/optic atrophy 1 (AMPK/OPA1) signaling pathway. The effects of melatonin on vascular calcification were investigated in vascular smooth muscle cells (VSMCs). Calcium deposits were visualized by Alizarin Red S staining, while calcium content and alkaline phosphatase (ALP) activity were used to

Inhibition of acid‑sensing ion channel 1a attenuates acid‑induced activation of autophagy via a calcium signaling pathway in articular chondrocytes.

Acid‑sensing ion channel 1a (ASIC1a), member of the degenerin/epithelial sodium channel protein superfamily, serves a critical role in various physiological and pathological processes. The aim of the present study was to examine the role of ASIC1a in the autophagy of rat articular chondrocytes. Autophagy was induced by acidic stimulation in rat articular chondrocytes and the extent of autophagy was evaluated via the expression levels of microtubule‑associated protein 1 light chain 3II, Beclin1 and uncoordinated‑51 like kinase1. Suppression of ASIC1a was achieved using small interfering RNA tec

mGlu1 tonically regulates levels of calcium-permeable AMPA receptors in cultured nucleus accumbens neurons through retinoic acid signaling and protein translation.

In several brain regions, ongoing metabotropic glutamate receptor 1 (mGlu1) transmission has been shown to tonically suppress synaptic levels of Ca2+ -permeable AMPA receptors (CP-AMPARs) while pharmacological activation of mGlu1 removes CP-AMPARs from these synapses. Consistent with this, we previously showed in nucleus accumbens (NAc) medium spiny neurons (MSNs) that reduced mGlu1 tone enables and mGlu1 positive allosteric modulation reverses the elevation of CP-AMPAR levels in the NAc that underlies enhanced cocaine craving in the "incubation of craving" rat model of addiction. Calcium phosphate particles induce interleukin-8 expression in a human gingival epithelial cell line via the nuclear factor-κB signaling pathway.

BACKGROUND: Dental calculus is calcified plaque composed primarily of calcium phosphate mineral salts, and there is a clear association between the presence of calculus and the initiation/progression of periodontitis. However, it is still inconclusive whether dental calculus can be a direct causative factor. The authors examined the effect of nano/microsized calcium phosphate particles, which may be generated in the process of early precipitation and/or dissolution of calcium phosphate mineral, on the expression of interleukin (IL)-8 in human gingival epithelial cells. METHODS: Primary human

Calcium input potentiates the transforming growth factor (TGF)-beta1-dependent signaling to promote the export of inorganic pyrophosphate by articular chondrocyte.

Transforming growth factor (TGF)-β1 stimulates extracellular PP(i) (ePP(i)) generation and promotes chondrocalcinosis, which also occurs secondary to hyperparathyroidism-induced hypercalcemia. We previously demonstrated that ANK was up-regulated by TGF-β1 activation of ERK1/2 and Ca(2+)-dependent protein kinase C (PKCα). Thus, we investigated mechanisms by which calcium could affect ePP(i) metabolism, especially its main regulating proteins ANK and PC-1 (plasma cell membrane glycoprotein-1). We stimulated articular chondrocytes with TGF-β1 under extracellular (eCa(2+)) or cytosolic Ca(2+) (cCa

Activation of ERK1/2 and TNF-alpha production are mediated by calcium/calmodulin, and PKA signaling pathways during Mycobacterium bovis infection.

Mycobacterium bovis bacillus Calmette-Guérin (BCG)-induced tumor necrosis factor (TNF)-alpha secretion via an extracellular signal-regulated kinase (ERK) mitogen-activated protein kinase-dependent mechanism is an important host defence mechanism against Mycobacterium tuberculosis in human monocytes. We now define distinct signaling pathways that regulate induction of TNF-alpha and activation of ERK1/2 by intracellular signaling mechanisms during M. bovis infection. We determined that M. bovis BCG-induced ERK 1/2 activation occurs through a mechanism that requires intracellular calcium and like

The role of calcium signaling in early axonal and dendritic morphogenesis of rat cerebral cortex neurons under non-stimulated growth conditions.

The effects of depolarizing stimuli on neurite outgrowth have been shown to depend on an influx of extracellular calcium. However, the role of calcium under non-stimulated growth conditions is less well established. Here we investigated the contribution of calcium signaling to early neuronal morphogenesis of rat cerebral cortex neurons at three levels by blocking L-type voltage sensitive calcium channels, by depleting intracellular calcium or by blocking myosin light chain kinase. Detailed quantitative morphological analysis of neurons treated for 1 day revealed that depletion of intracellular…

Transcriptomic and functional profiling of human intestinal organoids identifies enhanced calcium signalling and thrombospondin-2 activity in cystic fibrosis.

BACKGROUND: Although gastrointestinal symptoms of cystic fibrosis (CF) have long been overlooked, patients with CF have an unexplained 5-fold increased risk of colorectal cancer. Much of our understanding of the disease has relied on two-dimensional cell lines and animal models. However, the primary changes occurring in human CF epithelial cells remain largely unclear. METHODS: Donor-matched wild-type (WT) and CFTR knockout (KO) human intestinal organoid (HIO) lines were employed to dissect epithelial cell-intrinsic responses to CFTR dysfunction. Knockout was confirmed at the molecular level

High-Throughput Optical Controlling and Recording Calcium Signal in iPSC-Derived Cardiomyocytes for Toxicity Testing and Phenotypic Drug Screening.

Understanding how excitable cells work in health and disease and how that behavior can be altered by small molecules or genetic manipulation is important. Genetically encoded calcium indicators (GECIs) with multiple emission windows can be combined (e.g., for simultaneous observation of distinct subcellular events) or used in extended applications with other light-dependent actuators in excitable cells (e.g., combining genetically encoded optogenetic control with spectrally compatible calcium indicators). Such approaches have been used in primary or stem cell-derived neurons, cardiomyocytes, a
