Effects of remote ischemic preconditioning on myocardial injury and endothelial function and prognosis after percutaneous coronary intervention in patients with acute coronary syndrome.

In marine ecosystems, most invertebrates possess diverse microbiomes on their external surfaces, such as those found in the pedal mucus of grazing gastropods and chitons that aids displacement on different surfaces. The microbes are then transported around and placed in contact with free-living microbial communities of micro and other macro-organisms, potentially exchanging species and homogenizing microbial composition and structure among grazer hosts. Here, we characterize the microbiota of the pedal mucus of five distantly related mollusk grazers, quantify differences in microbial community

Multi Omics Analysis Reveals That Compound Radix Pulsatillae and Lactic Acid Bacteria Reprogram the Microbiome Metabolome Network in Oat Silage.

Oat (Avena sativa L.) silage fermentation often fails due to insufficient lactic acid bacteria (LAB) and low water-soluble carbohydrate content. We investigated the effects of Compound Radix Pulsatillae (CRP; 40 g/kg FM) alone or combined with a commercial LAB inoculant (containing L. plantarum, L. buchneri, and Enterococcus faecium, CRP_LA) on oat silage after 60 days. Compared to control (CK), both CRP and CRP_LA increased dry matter and water-soluble carbohydrate retention while reducing fiber components and ammonia nitrogen (p < 0.05). CRP_LA exhibited superior fermentation quality (lowest

Infant Botulism and Carbohydrate Blood Group Antigens: Preliminary Results from a Discordant Twin Study.

OBJECTIVE: We investigated if specific carbohydrate blood group antigens were associated with infant botulism (IB) among discordant twins and triplets. STUDY DESIGN: Infants in the US and Canada from 2015 to 2022 who had laboratory-confirmed IB; were members of a multiple gestation birth; and had unaffected sibling(s), were eligible to participate. Blood specimens were tested for ABO, Lewis, H, and P1 carbohydrate blood antigens. FUT2 and FUT3 genes were sequenced to determine secretor and Lewis antigen status, respectively. In addition, characteristics of IB cases in the study were compared

Sugar metabolism by Brucellae.

The metabolic capabilities of the species of Brucella were originally of interest as a means of distinguishing them from each other and from other genera. Certain unusual characteristics, especially erythritol utilization, were studied in the hopes they would shed light on the pathogenicity. With the advent of modern genetic methods and genomic sequencing, it is now possible to get a good idea of the total capabilities of the organism and to do tests to confirm these deductions. Brucella appears to be a fairly normal member of the alpha-proteobacteria, but with some differences. A few question

Multi-omics profiling reveals gut segment-specific and age-associated microbial restructuring linked to host phenotypic maturation in Taihu geese.

Taihu geese are valued for efficient utilization of fibrous feed resources and for desirable meat quality, yet how gut microbial organization varies across intestinal regions and physiological stages, and how these changes relate to host phenotype, remains unclear. We integrated 16S rRNA gene sequencing, ITS sequencing, shotgun metagenomics, and host phenotyping to characterize intestinal microbial ecology in Taihu geese at 160 days of age (D160; physiological stability stage) and 450 days of age (D450; late-laying stage). Spatial profiling of the D160 group across the duodenum, jejunum, ileum

Comparative genomics of Sneathia species: insights into pathogenicity and adaptation to the human niche.

BACKGROUND: Sneathia are urogenital anaerobes linked to bacterial vaginosis, preterm birth, chorioamnionitis, urethritis, and HPV-mediated cervical carcinogenesis. Two species have been identified: Sneathia vaginalis and Sneathia sanguinegens. Yet, due to their fastidious nature, our knowledge of their pathogenicity is limited. METHODS: Comparative genomic analysis of 12 Sneathia genomes, including two newly sequenced via whole-genome sequencing, was used to evaluate the pathogenic potential of S. vaginalis and S. sanguinegens. RESULTS: Sneathia species have small genomes (1.25-1.35 Mbp) wit

A somatically mutated human antiganglioside IgM antibody that induces experimental neuropathy in mice is encoded by the variable region heavy chain gene, V1-18.

IgM paraproteins associated with autoimmune peripheral neuropathy and anti-Pr cold agglutinins react with sialic acid epitopes present on disialylated gangliosides including GD1b, GT1b, GQ1b, and GD3. A causal relationship between the paraprotein and the neuropathy has never been proven experimentally. From peripheral blood B cells of an affected patient, we have cloned a human hybridoma secreting an antidisialosyl IgM mAb, termed Ha1, that shows identical structural and functional characteristics to its serum counterpart. Variable region analysis shows Ha1 is encoded by the same VH1 family he

Pyruvate oxidase, as a determinant of virulence in Streptococcus pneumoniae.

Pneumococcus has been shown to bind to epithelial cells of the nasopharynx and lung, and to endothelial cells of the peripheral vasculature. To characterize bacterial elements required for attachment to these cell types, a library of genetically altered pneumococci with defects in exported proteins was screened for the loss of attachment to glycoconjugates representative of the nasopharyngeal cell receptor, type II lung cells (LC) and human endothelial cells (EC). A mutant was identified which showed a greater than 70% loss in the ability to attach to all cell types. This mutant also showed de

Genomic and functional characterisation of exopolysaccharide biosynthesis in marine bacterium Alteromonas sp. PRIM-28 with emulsifying and anti-inflammatory activities.

Bacterial exopolysaccharides (EPS) are structurally heterogeneous biopolymers governed by species- and strain-specific biosynthetic pathways and with diverse biotechnological applications. The study investigated EPS biosynthesis in a marine bacterium, Alteromonas sp. PRIM-28 through an integrated genomic and experimental approach. Whole-genome sequencing and functional annotation were performed to identify the gene clusters and regulatory elements associated with EPS biosynthesis. The salt tolerance of the bacterium was studied at salt concentrations of up to 25%. EPS production was assessed u

Evaluation of Indigenous Bacillus Strains from Asian Fermented Foods for Probiotic Properties.

Bacillus species hold particular importance due to their versatile enzymatic repertoire and ability to synthesize diverse bioactive metabolites. In this study, two fermented food-derived strains, Bacillus siamensis BB3 (from douchi) and Bacillus velezensis TMA10 (from tapai) were evaluated for their probiotic, anti-microbial and functional potentials. Both strains exhibited desirable probiotic characteristics, including desirable tolerance to simulated gastric and intestinal conditions, with BB3 showing greater acid tolerance than TMA10. Safety assessments confirmed the absence of hemolytic ac

Comprehensive nutritional metabolomics and transcriptomics reveal varietal differences and drying effects on health-related metabolites in Amomum tsao-ko, a traditional medicinal and culinary plant.

Amomum tsao-ko, a valued traditional medicinal and culinary plant, lacks a systematic characterization of its primary nutritional metabolites across different varieties and processing methods. This study employed widely-targeted metabolomics (UPLC-MS/MS) combined with transcriptome sequencing to comprehensively profile amino acids, vitamins, carbohydrates, and lipids in fresh and dried fruits of three morphologically distinct A. tsao-ko accessions. We identified 474 nutritional metabolites, uncovering significant varietal specificity; the round-fruited accession consistently accumulated higher

Congenital disorder of glycosylation due to DPM1 mutations presenting with dystroglycanopathy-type congenital muscular dystrophy.

Congenital disorders of glycosylation (CDG) are rare genetic defects mainly in the post-translational modification of proteins via attachment of carbohydrate chains. We describe an infant with the phenotype of a congenital muscular dystrophy, with borderline microcephaly, hypotonia, camptodactyly, severe motor delay, and elevated creatine kinase. Muscle biopsy showed muscular dystrophy and reduced α-dystroglycan immunostaining with glycoepitope-specific antibodies in a pattern diagnostic of dystroglycanopathy. Carbohydrate deficient transferrin testing showed a pattern pointing to a CDG type I

Impact of the resident microbiota on the nutritional phenotype of Drosophila melanogaster.

BACKGROUND: Animals are chronically infected by benign and beneficial microorganisms that generally promote animal health through their effects on the nutrition, immune function and other physiological systems of the host. Insight into the host-microbial interactions can be obtained by comparing the traits of animals experimentally deprived of their microbiota and untreated animals. Drosophila melanogaster is an experimentally tractable system to study host-microbial interactions. METHODOLOGY/PRINCIPAL FINDINGS: The nutritional significance of the microbiota was investigated in D. melanogaste

Cystic fibrosis and bacterial colonization define the sputum N-glycosylation phenotype.

Although mucin O-glycosylation of sputum from individuals suffering from cystic fibrosis (CF) is known to be altered relative to their unaffected counterparts, protein N-glycosylation of CF sputum remains structurally and functionally under-characterized. We report the first N-glycome of soluble proteins in sputum derived from five CF patients, two pathogen-free and two pathogen-infected/colonized non-CF individuals suffering from other pulmonary conditions. N-Glycans were profiled using porous graphitized carbon-liquid chromatography-negative ion-tandem mass spectrometry following enzymatic r

Biosynthesis of a novel 3-deoxy-D-manno-oct-2-ulosonic acid-containing outer core oligosaccharide in the lipopolysaccharide of Klebsiella pneumoniae.

The core oligosaccharide region of Klebsiella pneumoniae lipopolysaccharide contains some novel features that distinguish it from the corresponding lipopolysaccharide region in other members of the Enterobacteriaceae family, such as Escherichia coli and Salmonella. The conserved Klebsiella outer core contains the unusual trisaccharide 3-deoxy-d-manno-oct-2-ulosonic acid (Kdo)-(2,6)-GlcN-(1,4)-GalUA. In general, Kdo residues are normally found in the inner core, but in K. pneumoniae, this Kdo residue provides the ligation site for O polysaccharide. The outer core Kdo residue can also be non-sto

Specific defect in N-acetylglucosamine incorporation in the biosynthesis of the glycosylphosphatidylinositol anchor in cloned cell lines from patients with paroxysmal nocturnal hemoglobinuria.

Paroxysmal nocturnal hemoglobinuria (PNH) is a clonal disorder arising in a multipotent hemopoietic stem cell. PNH manifests clinically with intravascular hemolysis resulting from an increased sensitivity of the red cells belonging to the PNH clone to complement-mediated lysis. Numerous studies have shown that surface proteins anchored to the membrane via a glycosylphosphatidylinositol (GPI) anchor (including proteins protecting the cell from complement) are deficient on the cells of the PNH clone, leading to the notion that GPI-anchor biosynthesis may be abnormal in these cells. To investigat

Specific intravenous carbohydrate therapy. A new concept in inhibiting antibody-mediated rejection--experience with ABO-incompatible cardiac allografting in the baboon.

Heterotopic allografting of ABO-incompatible donor hearts in recipient baboons "hyperimmunized" against the incompatible A or B antigen (n = 3) was followed by hyperacute antibody-mediated vascular rejection within a mean of 19 min. The A and B epitopes against which these antibodies are directed are carbohydrates that can be synthesized. The continuous i.v. infusion of the specific synthetic A or B trisaccharide, beginning immediately pre-transplant and continued posttransplant for several days, prolonged allograft survival to a mean of 8 days (n = 2) and prevented antibody-mediated rejection

Orally hypoglycemic activity of an insulin mimetic glycoprotein isolated from Cnidoscolus quercifolius Pohl. (Euphorbiaceae) seeds, Cq-IMP.

The genus Cnidoscolus (Euphorbiaceae) is widely distributed in tropical areas. In the Northeast of Brazil, the species C. quercifolius is endemic and has been used in traditional medicine. In this study, a novel protein was isolated from C. quercifolius seeds and characterized by its molecular weight, primary structure, isoelectric point (pI), and carbohydrate content. The hypoglycemic activity of this protein was investigated by in vitro assay with the RIN-5F glucose-responsive cell line and in vivo test using alloxan-induced diabetic mice models. In addition, safe use of the protein was also

Cloning of subunits of convulxin, a collagen-like platelet-aggregating protein from Crotalus durissus terrificus venom.

Convulxin (CVX) is a potent platelet-aggregating glycoprotein from the venom of the snake Crotalus durissus terrificus. It consists of two subunits, alpha and beta, joined by disulphide bridges in a hexameric structure. A cDNA library from venom gland was constructed in the vector pT3T7. The cloned cDNAs encoding the two chains of CVX were sequenced. Both are preceded by an identical 23-amino acid peptide signal sequence and encode sequences of 135 amino acids for the alpha chain and 125 amino acids for the beta chain. These polypeptides include a carbohydrate-recognition domain (CRD) in which

SLC39A8 Deficiency: A Disorder of Manganese Transport and Glycosylation.

SLC39A8 is a membrane transporter responsible for manganese uptake into the cell. Via whole-exome sequencing, we studied a child that presented with cranial asymmetry, severe infantile spasms with hypsarrhythmia, and dysproportionate dwarfism. Analysis of transferrin glycosylation revealed severe dysglycosylation corresponding to a type II congenital disorder of glycosylation (CDG) and the blood manganese levels were below the detection limit. The variants c.112G>C (p.Gly38Arg) and c.1019T>A (p.Ile340Asn) were identified in SLC39A8. A second individual with the variants c.97G>A (p.Val33Met) an
