Antigenic analysis of grass carp reovirus using single-chain variable fragment antibody against IgM from Ctenopharyngodon idella.

Grass carp (Ctenopharyngodon idella) is an important species of freshwater aquaculture fish in China. However, grass carp reovirus (GCRV) can cause fatal hemorrhagic disease in yearling populations. Until now, a strategy to define the antigenic capacity of the virus's structural proteins for preparing an effective vaccine has not been available. In this study, some single-chain variable fragment antibodies (scFv), which could specifically recognize grass carp IgM, were selected from a constructed mouse naïve antibody phage display cDNA library. The identified scFv C1B3 clone was shown to posse

The relationship of cytotoxic and genotoxic damage with blood aluminum levels and oxidative stress induced by this metal in common carp (Cyprinus carpio) erythrocytes.

Aluminum is one of the most abundant elements in nature and is used in diverse industrial processes. As a result, it contaminates aquatic ecosystems, inducing damage on associated biota. In fish, it has been observed to induce hypoxia, hypercapnia, metabolic acidosis and respiratory arrest. Although there is little information on Al-induced cytotoxicity and DNA damage, this type of studies are essential in order to identify the mechanisms of action of this metal. The cytotoxic and genotoxic effects induced by Al on common carp (Cyprinus carpio) erythrocytes were determined in specimens exposed

Molecular and functional characterization of grass carp squint/nodal-related 1: a potential regulator of activin signaling in teleost pituitary cells.

Nodal, a member of the transforming growth factor-β superfamily, plays important roles in embryogenesis in vertebrates, including fish. However, the functional characterization of the fish nodal-related gene in nonembryonic cells is still unclear. In teleost, three nodal-related genes, nodal-related (ndr)1/squint, ndr2/cyclops, and ndr3/southpaw have been reported. In this study, a full-length cDNA for grass carp squint (gcSqt) was cloned, and its transcript was detected in the selected organs, including pituitary, brain, heart, head kidney, kidney, spleen, and gonad. To further define its fun

Two HMGB1 genes from grass carp Ctenopharyngodon idella mediate immune responses to viral/bacterial PAMPs and GCRV challenge.

High mobility group box 1 (HMGB1) is a nuclear weapon in the immune arsenal and a master regulator of innate immunity, at the crossroads between innate and adaptive immunity. To clarify the immune characterizations of HMGB1 in fishes, two co-orthologs of HMGB1 (CiHMGB1a and CiHMGB1b) were identified in grass carp Ctenopharyngodon idella by local EST database searching and RACE techniques. mRNA expressions of the two HMGB1 genes are widespread in fifteen tissues investigated. The transcripts of CiHMGB1a and CiHMGB1b were significantly up-regulated and reached peak at 24h post GCRV challenge in

The efficacy of chromium as a growth enhancer for mirror carp (Cyprinus carpio L): an integrated study using biochemical, genetic, and histological responses.

A growth trial was conducted on juvenile mirror carp (Cyprinus carpio L.) for 8 weeks to compare the efficacy of three chromium (Cr) compounds (Cr chloride, Cr picolinate, and Cr yeast) at a level 0.5 mg/kg as a potential growth enhancer. In addition, a high level of Cr (2.0 mg/kg) as Cr chloride has also been added in parallel for comparison. All Cr fortified diets at a level 0.5 mg/kg produced superior growth for carp compared to the control group and the group fed the high level of Cr chloride (2.0 mg/kg). Metabolic indicators measured included two of the key liver enzymes (hexokinase, HK)

Seasonal development and pathological changes associated with the parasitic nematode Philometroides sanguineus in wild crucian carp Carassius carassius (L.) in England.

Pathological changes associated with the parasitic nematode Philometroides sanguineus (Rudolphi, 1819) are described for the first time from observations of infections in wild crucian carp Carassius carassius (L.) in England. The damage caused by P. sanguineus was influenced strongly by host size, parasite development and the seasonal migrations of female nematodes within host tissues. Male and unfertilized female nematodes located on the swim-bladder wall caused only minor, localized changes. In contrast, the migration of gravid female nematodes into the fins during autumn provoked an acute i

Insulin-like growth factor as a novel stimulator for somatolactin secretion and synthesis in carp pituitary cells via activation of MAPK cascades.

Somatolactin (SL), a member of the growth hormone/prolactin family, is a pituitary hormone unique to fish models. Although SL is known to have diverse functions in fish, the mechanisms regulating its secretion and synthesis have not been fully characterized. Using grass carp pituitary cells as a model, here we examined the role of insulin-like growth factor (IGF) in SL regulation at the pituitary level. As a first step, the antisera for the two SL isoforms expressed in the carp pituitary, SLα and SLβ, were produced, and their specificity was confirmed by antiserum preabsorption and immunohisto

Neural regulation of dark-induced abundance of arylalkylamine N-acetyltransferase (AANAT) and melatonin in the carp (Catla catla) pineal: an in vitro study.

In all the vertebrates, synthesis of melatonin and its rhythm-generating enzyme arylalkylamine N-acetyltransferase (AANAT) reaches its peak in the pineal during the night in a daily light-dark cycle, but the role of different neuronal signals in their regulation were unknown for any fish. Hence, the authors used specific agonist and antagonists of receptors for different neuronal signals and regulators of intracellular calcium (Ca(2+)) and adenosine 3',5'-cyclic monophosphate (cAMP) in vitro to study their effects on the abundance of AANAT and titer of melatonin in the carp (Catla catla) pinea

In vivo evoked specific cell mediated cytotoxicity in carp (Cyprinus carpio L.) uses mainly a perforin/granzyme-like pathway.

A specific cytotoxicity assay has been developed for carp using the Epithelioma Papulosum Cyprini (EPC) cell line as target cells and the release of lactate dehydrogenase (LDH) as read out system. Non-specific cytotoxicity against EPC was not observed, but animals immunized with EPC clearly showed specific killing by effector cells present in kidney, blood and spleen. This killing was strongly calcium dependent, suggesting the utilization of a perforin/granzyme-like pathway. However, blocking with EGTA was not complete indicating the existence of other additional killing routes.

Forskolin modulation of desensitization at GABA(A) and glycine receptors is not mediated by cAMP-dependent protein kinase in isolated carp amacrine-like cells.

The effects of forskolin on gamma-aminobutyric acid type-A (GABA(A)) and glycine receptors in amacrine-like cells of carp (Carassius auratus) retina were studied using patch-clamp techniques. Application of 50 microM forskolin markedly accelerated the desensitization of whole-cell responses induced by 100 microM GABA or glycine without changing the peak amplitude of the response. Both 8-bromoadenosine 3',5'-cyclic monophosphate (8-Br-cAMP) and 3-isobutyl-1 -methylxanthine (IBMX) (500 microM) failed to accelerate the desensitization of these two receptors. Effects of morphine and methionine-enkephalin on the smooth muscle tonus and the contraction induced by transmural stimulation in the carp (Cyprinus carpio) intestinal bulb.

The effects of morphine and methionine-enkephalin (met-enkephalin) on the smooth muscle tonus and the contraction induced by transmural stimulation were investigated in the isolated intestinal bulb of carp in vitro. Morphine (30 nM-3 microM) and met-enkephalin (3 nM-5 microM) caused dose-dependent non-sustained contraction. Naloxone (10 nM) inhibited the contraction induced by morphine or met-enkephalin in a competitive manner. Tetrodotoxin (400 nM) or atropine (500 nM) did not inhibit the contraction induced by morphine or met-enkephalin. Cooling of the bath fluid from 20 to 10 degrees C decr

Dopaminergic mechanisms in the teleost retina. I. Dopamine-sensitive adenylate cyclase in homogenates of carp retina; effects of agonists, antagonists, and ergots.

A specific dopamine-sensitive adenylate cyclase has been identified in homogenates of the teleost (carp) retina. Maximal stimulation by 100 microM-dopamine resulted in a 5--10-fold increase in adenylate cyclase activity with half-maximal stimulation occurring at a concentration of 1 microM. l-Noradrenaline and l-adrenaline were some 10 times less potent than dopamine whilst the alpha- and beta-adrenoreceptor agonists, l-phenylephrine and dl-isoprenaline were inactive. Apomorphine elicited a partial stimulation of adenylate cyclase activity whilst various ergot alkaloids produced mixed agonist/

The connections of the trigeminal and facial motor nuclei in the brain of the carp (Cyprinus carpio L.) as revealed by anterograde and retrograde transport of horseradish peroxidase.

The connections of the rostral and caudal parts of the trigeminal and facial motor nuclei in the carp were studied with the horseradish peroxidase technique. Following ionophoretic peroxidase injections in these motor nuclei, retrogradely labeled cells were observed together with anterogradely labeled motor cell processes. Several cellular areas in thalamus, cerebellum and medulla oblongata were shown to project to the V and VII motor nuclei. Labeled cells were found in the inferior lobe and the glomerular complex of the thalamus. In the medulla oblongata, cells in the descending trigeminal nu

Intestinal Responses to Short-Term Hexaflumuron Exposure in Grass Carp (Ctenopharyngodon idella): A Combined Physiological, Transcriptomic, and Gut Microbiota Perspective.

Hexaflumuron is a benzoylurea insecticide with potential application against arthropod ectoparasites in aquaculture, but its toxicity to non-target fish remains insufficiently characterized. This study evaluated acute toxicity and intestinal responses to sublethal concentrations of hexaflumuron in juvenile grass carp (Ctenopharyngodon idella). The 48- and 96-h LC50 values were 8.80 and 7.16 mg/L, respectively. Fish were subsequently exposed to nominal concentrations of 0.07 or 0.72 mg/L hexaflumuron for 96 h. Intestinal histopathology, antioxidant biomarkers, 16S rRNA sequencing, and transcrip

Metatranscriptomic analyses coupled with phenotypic and metabolic verification revealed competition mechanisms between grass carp spoilage bacteria in synthetic communities.

Bacterial competition is a major factor influencing microbiota succession in fish. However, the competition mechanisms among dominant spoilage bacteria in grass carp remain to be systematically investigated. Therefore, this study aimed to elucidate the mutual competition relationships and mechanisms among five representative spoilage bacteria isolated from grass carp. To this end, we constructed synthetic microbial communities (SynComs) with these five strains and employed metatranscriptomics coupled with phenotypic verification to demonstrate their interactions at both transcriptional and phe

Biological Characteristics, Pathogenicity Assessment, and Whole genome analysis of Aeromonas salmonicida Strain SGQ-1 Isolated from Crucian Carp.

Aeromonas salmonicida is an important fish pathogen, but its pathogenicity in non-salmonid hosts remains poorly characterized. In this study, strain SGQ-1 was isolated from diseased crucian carp (Carassius auratus) and subjected to phenotypic, genomic, antimicrobial susceptibility, and pathogenicity analyses. SGQ-1 was a Gram-negative, β-hemolytic, short rod-shaped bacterium. The optimal growth was observed at 28 °C and pH 7.0 in separate experiments. Analyses of average nucleotide identity, digital DNA-DNA hybridization, and core-genome phylogeny identified SGQ-1 as A. salmonicida. However, t

Comparative Hepatic Toxicity of Pesticides in Common Carp (Cyprinus carpio Linnaeus, 1758): An Integrated Histopathological, Histochemical, and Enzymatic Biomarker Approach.

The intensive use of pesticides in agriculture poses serious risks to aquatic ecosystems and non-target organisms, yet toxicological data remain limited. This study evaluated the acute effects of three widely used pesticides-pirimiphos-methyl (10 and 60 μg/L), propamocarb hydrochloride (40 and 80 μg/L), and 2,4-D (50 and 100 μg/L)-on the liver of common carp (Cyprinus carpio Linnaeus, 1758), a sentinel species in aquaculture, but also a species equally important in risk assessment and environmental monitoring. Fish were exposed for 96 h under controlled conditions, and histopathological, histo
