Optimized detection of caspase-6 activation in a murine inflammation model to inform neurodegenerative disease therapies.

Caspase-6 (casp-6), is a member of the apoptotic family of caspases (cysteine aspartic proteases) and has been shown to be involved in several neurodegenerative diseases, including Alzheimer's disease. Drug discovery efforts have focused on specific inhibition of casp-6, and it is important that lead compounds demonstrate engagement and inhibition of the protease in vivo. A casp-6 overexpressing mouse model was reported, but is no longer available to the scientific community. We developed alternative means of activating casp-6 in commercially available mice as a platform to test potential casp…

Pyroptosis in chronic obstructive pulmonary disease: Beyond the canonical NLRP3/Caspase-1/GSDMD pathway.

Chronic obstructive pulmonary disease (COPD), the third leading cause of death worldwide, is characterized by persistent airway inflammation that remains incompletely controlled in some patients despite current anti-inflammatory therapies. Pyroptosis, a pro-inflammatory, lytic form of programmed cell death mediated by the gasdermin family of proteins, has emerged as a key amplifier of inflammation in COPD. Although most evidence centers on the NLR family pyrin domain-containing 3 (NLRP3)/caspase-1/gasdermin D (GSDMD) axis, COPD-relevant studies increasingly implicate non-canonical caspase-4/5/

Inverse expression of ubiquitin-specific peptidase 19 and caspase 7 correlates with gastric neoplastic transformation.

BACKGROUND: Ubiquitin-specific protease 19 (USP19) is a deubiquitinylase (DUB) that is part of the USP family, the largest group of DUBs in humans. Growing evidence has indicated that USP19 is involved in tumor progression and serves as a new prognostic marker for various malignant disorders. Interestingly, USP19 has been shown to have both promoting and inhibiting effects on the onset and development of different neoplasms, depending on the specific tissue type. DUBs including USP19 affect a variety of cell functions including apoptotic cell death. Herein, caspase 7 acts as a key executioner

Caspase-1-mediated pyroptosis drives secondary thalamic neurodegeneration after focal cerebral infarction.

BACKGROUND: Secondary neurodegeneration in brain regions remote from the primary infarct contributes substantially to long-term neurological dysfunction after ischemic stroke. Although pyroptosis has been implicated in acute ischemic injury, its contribution to delayed secondary thalamic degeneration remains poorly understood. This study investigated whether canonical inflammasome-mediated pyroptosis contributes to secondary thalamic injury following focal cerebral infarction. METHODS AND RESULTS: A permanent distal middle cerebral artery electrocoagulation model was established in male C57BL

Electroacupuncture alleviates synovial inflammation in rheumatoid arthritis rats by suppressing caspase-1-mediated pyroptosis.

Rheumatoid arthritis (RA) causes inflammatory bone and cartilage damage. While electroacupuncture (EA) alleviates joint inflammation, its anti-inflammatory mechanisms remain unclear. Sprague-Dawley rats were randomly assigned to control, collagen-induced arthritis (CIA), EA (EA+CIA), and sham-EA (sham-EA+CIA) groups. Rats in the EA group received EA at unilateral ST36 (Zusanli) every other day from day 1. Paw swelling and paw withdrawal latency (PWL) were measured during the treatment. The levels of caspase-1, GSDMD, IL-1β, and IL-18 in the synovium were assessed after 28 days of EA treatment.

Differential Expression of Caspase-3 and Ki-67 in Hereditary Gingival Fibromatosis and Inflammatory Gingival Hyperplasia.

OBJECTIVE: The nuclear protein Ki-67 and caspase-3 regulate cell proliferation and apoptosis as well as the preservation of tissue homeostasis in response to genetic and inflammatory signals that have a role in the pathogenesis of different conditions such as gingival overgrowth. Yet, the distinct pathogenesis of these lesions could be determined by variations in cellular homeostasis mechanisms. The aim of this study was to assess and compare the immunohistochemical expression of Ki-67 and caspase-3 in hereditary gingival fibromatosis (HGF) and inflammatory gingival hyperplasia (IGH), measure

Japanese encephalitis virus activates the NLRP3/caspase-1/GSDMD signaling pathway in dopaminergic neurons.

Japanese encephalitis virus (JEV) preferentially targets brain regions rich in dopaminergic neurons, including the thalamus, midbrain, and striatum, leading to severe neuroinflammation. However, the underlying pathological mechanisms remain unclear. This study hypothesizes that JEV-induced pyroptosis exacerbates neuroinflammation by activating the NLRP3/caspase-1/GSDMD signaling pathway. In vivo experiments using JEV-infected mice revealed virus-specific targeting of dopaminergic neurons, concurrent activation of the NLRP3 inflammasome, and induction of inflammatory responses. In vitro studies

Porcine caspase-8 and 13 cleave GSDMD to generate products can cause cell death.

Pyroptosis is a form of programmed cell death mediated by gasdermin family proteins (GSDMs), characterized by the disruption of the cell membrane and the release of cytoplasmic contents. Protease-mediated cleavage is essential for activating the pyroptosis-mediating function of GSDMs. Therefore, identification of proteases that are capable of processing GSDMs is crucial for elucidating the mechanisms of pyroptosis in different hosts. To date, it has been determined that porcine caspase-1 (pCASP1) can cleave porcine GSDMD (pGSDMD) to activate the pyroptosis-mediating ability of pGSDMD. However,

Caspase 6 deficiency exacerbates inflammatory bowel disease via enterocyte necroptosis and bacterial translocation.

Caspase 6 is a pivotal executioner caspase involved in cell death; however, its role in inflammatory bowel disease (IBD) remains incompletely understood. Levels of cleaved caspase 6 were quantified in colonic tissues from IBD patients, and an IBD mouse model was established via DSS induction, incorporating both systemic (Casp6 KO) and IEC-specific knockout (Casp6 cKO) strategies. Single-cell RNA sequencing (scRNA-seq) revealed that Casp6 KO enhanced necroptosis in IECs, reducing intestinal endocrine cells and damaging intestinal stem cells. Both in vivo and in vitro studies confirmed that casp

Selective genetic inactivation of Caspase 8 in hepatocytes ameliorates progression of MASH following Jnk deficiency.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) is associated with c-Jun N-terminal kinase (JNK) activation across various cell types, but its hepatocyte-specific function in steatotic liver disease remains unclear. Our study investigates the role of JNK1/JNK2 during MASH progression and dissects its hepatocyte-specific function. APPROACH AND RESULTS: We showed that UK Biobank patients with a predicted loss-of-function variant of JNK1 presented an increased prevalence of metabolic dysfunction-associated steatotic liver disease and liver damage. Analysis of a patho

Non-genetic inactivation of caspase-3 and P53 increases cancer cell fitness by PDIA4 redistribution.

Numerous cellular pathways are known to cause resistance in cancer cells. The unfolded protein response (UPR), a signaling pathway activated during proteostasis stress in the endoplasmic reticulum (ER), is an adaptive process to increase cancer cell fitness. However, the molecular mechanism between ER stress, UPR activation, and chemoresistance is insufficiently understood. Here, we report that ER stress induction and UPR activation are necessary for chemoresistance to cisplatin and doxorubicin. Mild ER stress is a sufficient precondition for cancer cells to evade cisplatin- and doxorubicin-as

Pyroptosis Mediated by ROS/Caspase-3/GSDME Pathway in Aspergillus fumigatus-Induced Fungal Keratitis.

PURPOSE: Gasdermin E (GSDME), cleaved by (casp-3) activation, is known as a key executor in pyroptosis. However, its role in fungal keratitis (FK) remains unclear. Thus we analyzed the role of GSDME, as well as its signaling pathway in Aspergillus fumigatus (AF)-induced FK. METHODS: We conducted experiments using a mouse model and human corneal epithelial cells. Initially, we infected mice with AF at various time intervals and examined the progression of FK lesions, selecting the time point with the most severe lesions. Next, we pre-treated the subjects with various cytokine inhibitors that m

Phyllanthin mitigates doxorubicin-induced acute cardiotoxicity: role of Bax/Bcl2 and Caspase-3 regulation.

Doxorubicin (DOX)-induced toxicity impairs cardiovascular functions. Phyllanthus amarus exhibits anti-inflammatory, antiviral, anticancer, antioxidant, nephroprotective, and hepatoprotective effects. However, its potential against DOX-induced acute cardiotoxicity is lacking. This study aimed to assess the underlying mechanism of action of P. amarus against DOX-induced acute cardiotoxicity in Sprague-Dawley rats. Phyllanthin was isolated from the methanolic extract of the aerial parts of P. amarus and characterized using HPLC-ESI-MS/MS. Its cardioprotective efficacy was evaluated in DOX-intoxic

Sublethal executioner caspase activation in hepatocytes promotes liver regeneration through the JAK/STAT3 pathway.

Apoptosis has been reported to drive regeneration in many species. Executioner caspases, the key effectors in apoptosis, are responsible for production and secretion of various pro-regenerative signals from apoptotic cells to the surrounding cells. However, whether executioner caspase activation (ECA) can promote regeneration without inducing apoptosis is poorly understood. Here, by generating transgenic mice carrying a lineage tracing system for cells that have experienced ECA, we demonstrate that ECA occurs in a few hepatocytes in homeostatic livers. The fraction of hepatocytes with ECA dram…

HMGB1 activates caspase-1 and induces hepatic stellate cell activation via GABPA-ASC.

In the intricate process leading to liver fibrosis, which frequently correlates with inflammation, the activation of hepatic stellate cells (HSCs) is critical. High mobility group box 1(HMGB1), as endogenous danger signal in the extracellular environment, governs the activation of caspase-1 and hepatic stellate cell. Constructing a liver fibrosis model via intraperitoneal thioacetamide (TAA) administration unveiled excessive HMGB1 expression and serum release during the TAA-induced fibrosis progression. Intraperitoneal injection of ethyl pyruvate (EP, which inhibits the release of HMGB1) or AA

Cisplatin induces acute liver injury by triggering caspase-3/GSDME-mediated cell pyroptosis.

BACKGROUND: Cisplatin triggers Gasdermin E (GSDME) cleavage, causing membrane bubble formation, content release, and inflammation. Caspase-3 activation initiates GSDME cleavage, and thus inhibiting this pathway mitigates cisplatin-induced pyroptosis in hepatocytes. This study aimed to delve into how cisplatin induces liver injury via pyroptosis. METHODS: For animal experiments, C57BL/6J mice were divided into three groups: control, liver injury model group, and Ac-DMLD-CMK (caspase-3 inhibitor) intervention group. The liver histology was evaluated by hematoxylin and eosin staining, immunohist
