Structural correlates of cognitive impairment in normal pressure hydrocephalus.

OBJECTIVES: The pathological bases for the cognitive and neuropsychiatric symptoms in normal pressure hydrocephalus (NPH) have not been elucidated. However, the symptoms may indicate dysfunction of subcortical regions. Previously, volume reductions of subcortical deep grey matter (SDGM) structures have been observed in NPH patients. The present study used automated segmentation methods to investigate whether SDGM structure volumes are associated with cognitive and neuropsychiatric measures. METHODS: Fourteen NPH patients and eight healthy controls were included in the study. Patients complete

Individual differences in risk preference predict neural responses during financial decision-making.

We investigated the neural correlates of subjective valuations during a task involving risky choices about lotteries. Because expected value was held constant across all lotteries, decisions were influenced by subjective preferences, which manifest behaviorally as risk-seeking or risk-averse attitudes. To isolate structures encoding risk preference during choice, we probed for areas showing increased activation as a function of selected risk-level. Such response patterns were obtained in anterior (ACC) and posterior cingulate cortex (PCC), superior frontal gyrus, caudate nucleus, and substanti…

Basal ganglia volume and shape in anorexia nervosa.

BACKGROUND: Reward-centred models have proposed that anomalies in the basal ganglia circuitry that underlies reward learning and habit formation perpetuate anorexia nervosa (AN). The present study aimed to investigate the volume and shape of key basal ganglia regions, including the bilateral caudate, putamen, nucleus accumbens (NAcc), and globus pallidus in AN. METHODS: The present study combined data from two existing studies resulting in a sample size of 46 women with AN and 56 age-matched healthy comparison (HC) women. Group differences in volume and shape of the regions of interest were e

Test-retest reliability of dopaminergic fPET and fMRI measures during reward processing.

Reward processing is essential to human brain function, with dopamine signalling in the nucleus accumbens (NAcc) as a key element. The monetary incentive delay task is widely studied with functional magnetic resonance imaging (fMRI), measuring indirect hemodynamic changes. Functional positron emission tomography (fPET) with 6-[¹⁸F]FDOPA directly quantifies dopamine synthesis, enabling dynamic assessment during task performance within a single scan. We investigated the reliability of 6-[¹⁸F]FDOPA fPET and blood oxygenation level dependent (BOLD) fMRI during a modified monetary incentive delay t

Differences between GABA levels in Alzheimer's disease and Down syndrome with Alzheimer-like neuropathology.

Down syndrome (DS) is a genetic disease with developmental brain abnormalities resulting in early mental retardation and precocious, age-dependent Alzheimer-type neurodegeneration. Furthermore, non-cognitive symptoms may be a cardinal feature of functional decline in adults with DS. A number of amino acids [glutamate, aspartate, gamma-aminobutyrate (GABA), glycine, taurine, glutamine, serine, arginine] were investigated in post-mortem tissue samples from temporal, occipital cortex, thalamus, caudate nucleus, and cerebellum of adult patients with Down syndrome (DS) exhibiting Alzheimer-like neu…

Genetic architecture of subcortical brain structures in 38,851 individuals.

Subcortical brain structures are integral to motion, consciousness, emotions and learning. We identified common genetic variation related to the volumes of the nucleus accumbens, amygdala, brainstem, caudate nucleus, globus pallidus, putamen and thalamus, using genome-wide association analyses in almost 40,000 individuals from CHARGE, ENIGMA and UK Biobank. We show that variability in subcortical volumes is heritable, and identify 48 significantly associated loci (40 novel at the time of analysis). Annotation of these loci by utilizing gene expression, methylation and neuropathological data id…

Resolving mesoscale brainstem-prefrontal-striatal pathways underlying decisions upon salient events using submillimeter-resolution fMRI.

Value-based decision-making engages brain-wide motivational, cognitive, and motor processes. Yet, information integration and gating that culminate in immediate decisions upon salient events likely occur within small neural nuclei and cortical layers at the mesoscale not resolved with conventional human neuroimaging. Using submillimeter-resolution 7 T functional MRI with acquisition-matched anatomical references and a lottery choice task incorporating salient superhigh stakes, we dissociated mesoscale operations spanning a brainstem-prefrontal-striatal pathway during choice and outcome process

Structural and functional neuroimaging changes associated with cognitive impairment and dementia in Parkinson's disease.

This study seeks a better understanding of possible pathophysiological mechanisms associated with cognitive impairment and dementia in Parkinson's disease using structural and functional MRI. We investigated resting-state functional connectivity of important subdivisions of the caudate nucleus, putamen and thalamus, and also how the morphology of these structures are impacted in the disorder. We found cognitively unimpaired Parkinson's disease subjects (n = 33), compared to controls (n = 26), display increased functional connectivity of the dorsal caudate, anterior putamen and mediodorsal thal…

Modafinil-induced conditioned place preference via dopaminergic system in mice.

Modafinil, a psychostimulant, is used in the treatment of narcolepsy, shift work sleep disorder, and excessive daytime sleepiness associated with obstructive sleep apnea. Preclinical and clinical studies suggest that modafinil may have reinforcing effects. However, a possible rewarding property of modafinil has not been fully investigated. In this study, we assessed the potential rewarding property of modafinil using the conditioned place preference (CPP) paradigm in mice. Using radiolabeled ligands, we observed changes in dopamine, glutamate, and GABA receptor binding in the brains of mice af

Variation of selective gray and white matter atrophy in Huntington's disease.

The relationship between the extent of local gray/white matter atrophy, genetic load, and clinical impairment was studied in Huntington's di … ne fissure, and with the white matter along the fourth and lateral ventricles. While atrophy of the caudate nucleus was related to a higher number of CAG triplets and higher UHDRS-motor score, atrophy in other parts of the brain covaried with the two parameters differently: higher genetic load was associated with greater loss of cortical somatosensory projections …

Reduction in cerebral atrophy associated with ethyl-eicosapentaenoic acid treatment in patients with Huntington's disease.

Ultra-pure ethyl-eicosapentaenoic acid (ethyl-EPA), a semi-synthetic ethyl ester of eicosapentaenoic acid, is associated with clinical impro … -blind manner. Significant group-level reductions in brain atrophy were observed in the head of the caudate nucleus and the posterior thalamus. These findings show that treatment with ethyl-EPA is associated with significant reduction in brain atrophy, particularly in the caudate and thalamus. No other drug tested in Huntington's disease has shown this effect.
