15 patients with acute myeloblastic leukaemia (AML) in remission were given immunotherapy and 17 similar patients were given immunotherapy plus chemotherapy with the drugs used to induce remission. Median remission length was 245 days for both groups and median survival was 465 days for patients given chemoimmunotherapy and 476 days for patients given only immunotherapy. The failure of remission chemotherapy in AML cannot be attributed to induced resistance because the same drugs induced a second remission in 60% of the patients. From laboratory studies with human AML cells it is suggested tha

BACKGROUND: Diabetes mellitus is a chronic condition caused by insufficient insulin secretion or impaired insulin action. Traditionally, pancreatic β-cell loss has been attributed to hyperglycemia-induced apoptosis. However, more recent evidence indicates that β-cells can also undergo dedifferentiation, a process in which they lose their specialized identity and revert to a precursor-like state. ALDH1A3 has been reported as a marker associated with dedifferentiated β-cells in diabetes, making it a potential molecular target for studies of β-cell identity and their redifferentiation.

A unique feature of the germinal center B cell-derived Hodgkin and Reed/Sternberg cells of classical Hodgkin lymphoma is their lost B cell phenotype and the aberrant expression of factors of other hematopoietic cell types, including ID2 and NOTCH1. As cellular dedifferentiation and upregulation of ID2 and NOTCH1 are typical consequences of a hypoxic response, we wondered whether hypoxia may impose an HRS cell-like phenotype in B cells. Culturing normal B cells or cell lines of germinal center-type diffuse large B-cell lymphoma under hypoxic conditions caused partial downregulation of several B

UNLABELLED: PROPERTIES OF THE ENDOTHELIUM: The endothelium lines all vessels of the body and is the most important communication structure between flowing blood on one hand and the vessel wall, composed of smooth muscle cells and matrix proteins, on the other. Endothelial cells release factors that affect the differentiation and growth of vascular smooth muscle cells; dedifferentiation and subsequent proliferation of vascular smooth muscle cells are important early mechanisms in the pathogenesis of chronic vascular disease.

Cancer cell invasion is modulated by their interaction with the tumor microenvironment (TME). In this article we have analyzed the cooperation of one of the TME cellular components, adipocytes, and breast tumor cells. Co-culture of these two types of cells increase tumor cell invasion and migration. This effect is associated to the de-differentiation of adipocytes that lose lipids and experience a transition to a mesenchymal phenotype. Furthermore, tumor cells are activated by adipocytes and undergo a partial epithelial-to-mesenchymal transition (EMT), which is characterized by a slow upregula

Expression of kidney injury molecule-1 (Kim-1) is rapidly upregulated following tubular injury, constituting a biomarker for acute kidney damage. We examined the renal localization of Kim-1 expression in PKD/Mhm (polycystic kidney disease, Mannheim) (cy/+) rats (cy: mutated allel, +: wild type allel), an established model for autosomal dominant polycystic kidney disease, with chronic, mainly proximal tubulointerstitial alterations. For immunohistochemistry or Western blot analysis, kidneys of male adult heterozygously-affected (cy/+) and unaffected (+/+) littermates were perfusion-fixed or dir

Cytophotometric, cytogenetic, and autoradiographic studies were performed in cells of three patients suffering from clinically diagnosed Sézary's syndrome with erythroderma and the presence of abnormal lymphoid cells in the peripheral blood, skin, bone marrow and lymph-nodes. Feulgen DNA cytophotometry of cells in the peripheral blood and skin lesions showed marked aneuploidy and tetraploidy. Multiple translocations were identified with a G-banding technique. The chromosomal abnormalities varied widely between the patients, but C and D group chromosomes were more frequently involved than other

Cervical biopsies from 97 women with malignant cells in cervical scrape smears have been studied. Forty-eight patients had invasive squamous carcinoma, and 49 had intraepithelial lesions. Of these, six had dedifferentiated carcinoma in situ, nine had the differentiated lesions generally known as ;severe dysplasia', and 29 had both; the severity of the dysplasia remained doubtful in five patients. The ;severe dysplasia' was compared with invasive carcinoma. A cellular analysis of the biopsies showed that the two categories have a number of features in common, chiefly the presence of atypical an

Human primordial germ cell-like cells (hPGCLCs) can be specified from human-induced pluripotent stem cells (hiPSCs), offering a valuable model for human germ cell development. However, further maturation steps of hPGCLCs rely on mouse feeders, or co-culture with mouse gonadal somatic cells. Exposure of hPGCLCs to human cellular niche has not been attempted. Here, we co-cultured female hPGCLCs in two distinct niche compartments. In reconstituted fetal ovary (rOv) culture, human fetal germ cells proliferated and initiated meiosis, while hPGCLCs upregulated gonadal germ cell markers such as DDX4.

Background/Objectives: Thyroid cancer spans indolent differentiated tumors to lethal poorly differentiated and anaplastic carcinomas, and histopathology and molecular testing do not capture the spatial organization of tumor, stromal, and immune compartments that underlies invasion, dedifferentiation, and treatment resistance. Spatial transcriptomics profiles gene expression in situ. We systematically reviewed its platforms, applications, and findings in thyroid cancer. Methods: PubMed/MEDLINE, Scopus, and Web of Science were searched, following PRISMA 2020 guidance and supplemented by citation

INTRODUCTION: Primary rectal choriocarcinoma is an extremely rare malignancy. The association of these neoplasms in patients with inflammatory bowel disease (IBD) has not been reported.
PRESENTATION OF CASE: A 34-year-old female with history of Ulcerative Colitis (UC) gave birth to a male fetus. She had postpartum bleeding and high level of beta-human chorionic gonadotropin (βhCG) was detected. Although initial investigations failed to confirm molar pregnancy, abnormal uterine bleeding and high βhCG level necessitate chemotherapy administration. She did not respond to chemotherapy sessions ac

BACKGROUND: G protein-coupled receptors are important regulators of contractility and differentiation in vascular smooth muscle cells (SMCs), but the specific function of SMC-expressed orphan G protein-coupled receptor class C group 5 member B (GPRC5B) is unclear.
METHODS: We studied the role of GPRC5B in the regulation of contractility and dedifferentiation in human and murine SMCs in vitro and in iSM-Gprc5b-KO (tamoxifen-inducible, SMC-specific knockout) mice under conditions of arterial hypertension and atherosclerosis in vivo.
RESULTS: Mesenteric arteries from SMC-specific Gprc5b-KOs sho

In adamantinoma of long bones, an osteofibrous dysplasia-like form with scattered epithelial elements and a classic form with abundant epithelium are distinguished. Osteofibrous dysplasia-like adamantinomas occur in children and adolescents and behave relatively benign, whereas classic adamantinomas predominate in adults and have a more aggressive clinical course. Because some osteofibrous dysplasia-like tumors have progressed to classic adamantinomas, it is hypothesized that the former is a potential precursor of the latter, showing mesenchymal-to-epithelial transformation. We report a new mo

Phenotypic modulation of smooth muscle cells (SMC) is a key event during the development of atherosclerotic and restenotic lesions. During this process, the composition of the cytoskeleton is substantially altered, with changes predominantly in actin expression reflecting a shift from smooth muscle alpha-actin to the non-muscle beta-isoform. We now demonstrate that yet another actin isoform, cardiac alpha-actin, is synthesized, de novo, in SMC of various atherosclerotic lesions. Using a highly specific monoclonal antibody against cardiac alpha-actin, we analyzed and compared the accumulation o

Sulfonylureas, commonly used to treat type 2 diabetes (T2D), often lose effectiveness over time when used as monotherapy; however, the underlying mechanisms remain unclear. To investigate the mechanisms of sulfonylurea failure, glibenclamide-releasing pellets were implanted in KK mice, a polygenic model that spontaneously develops T2D. KK mice receiving placebo pellets (KK-Placebo) developed hyperglycemia, hyperinsulinemia, glucose intolerance, and insulin resistance. Notably, KK mice implanted with glibenclamide (KK-Glib) showed improved blood glucose levels during the first 7 days, returning

The ability of articular cartilage to repair itself is limited because it lacks blood vessels, nerves, and lymph tissue. Once damaged, it can lead to joint swelling and pain, accelerating the progression of osteoarthritis. To date, complete regeneration of hyaline cartilage exhibiting mechanical properties remains an elusive goal, despite the many available technologies. The inflammatory milieu created by cartilage damage is critical for chondrocyte death and hypertrophy, extracellular matrix breakdown, ectopic bone formation, and progression of cartilage injury to osteoarthritis. In the infla

Anaplastic thyroid carcinoma (ATC) is a highly aggressive malignancy with a poor prognosis, characterized by dedifferentiation and aberrant angiogenesis. Through integrated analysis of TCGA and GEO transcriptomic data and single-cell RNA sequencing, this study identified significant enrichment of angiogenesis-related genes (ARGs), particularly sphingosine kinase 1 (SPHK1), in malignant cell subpopulations of ATC. Functional investigations revealed that alkaline ceramidase 3 (ACER3) cooperates with SPHK1 within the sphingolipid metabolic pathway to promote ATC progression. The SPHK1-specific in

The remnant auricular cartilage from microtia has become a valuable cell source for ear regeneration. It is important to clarify the issue of whether the genetically defective microtia chondrocytes could engineer cartilage tissue comparable to healthy ear chondrocytes. In the current study, the histology and cell yield of native microtia and normal ear cartilage were investigated, and the biological characteristics of derived chondrocytes examined, including proliferation, chondrogenic phenotype and cell migration. Furthermore, the in vivo cartilage-forming capacity of passaged microtia and no

Isoforms of the transmembrane glycoprotein CD44, which are generated by alternative splicing of nine variant exons, have been implicated in tumor cell adhesion, invasion and metastatic spread and may be indicators of the degree of tumor differentiation. Since little is known about the distribution of CD44 in non-neoplastic neuroendocrine cell types, we systematically investigated 42 samples of tissue from different organs, including the pituitary gland, thyroid, parathyroid, adrenal gland, lung, pancreas, stomach, duodenum, jejunum, ileum, appendix, and colon, immunohistochemically for the exp

In this retrospective study, the degree of differentiation of atypical lymphoid cells was assessed in pretreatment cutaneous biopsy specimens from 248 patients with cutaneous T-cell lymphomas (mycosis fungoides and Sézary syndrome) and the findings were correlated with the subsequent therapeutic results. Overall, patients with a predominance of cells with hyperchromatic nuclei (well-differentiated lymphoid cells) in cutaneous infiltrates responded better to treatment with improved survival rates than patients with infiltrates composed predominantly of cells with pale vascular nuclei (poorly di
