Diversity of Clinical and Environmental Isolates of Vibrio cholerae in Natural Transformation and Contact-Dependent Bacterial Killing Indicative of Type VI Secretion System Activity.

The bacterial pathogen Vibrio cholerae can occupy both the human gut and aquatic reservoirs, where it may colonize chitinous surfaces that induce the expression of factors for three phenotypes: chitin utilization, DNA uptake by natural transformation, and contact-dependent bacterial killing via a type VI secretion system (T6SS). In this study, we surveyed a diverse set of 53 isolates from different geographic locales collected over the past century from human clinical and environmental specimens for each phenotype outlined above. The set included pandemic isolates of serogroup O1, as well as s

Microenvironment Self-Adaptive Nanoarmor to Address Adhesion- and Colonization-Related Obstacles in Impaired Intestine Promote Bacteriotherapy Against Parkinson's Disease.

Intestinal localized inflammations are recognized as key contributors to the incidence and progression of diverse extraintestinal disorders. Probiotic colonization has been increasingly highlighted for its potential to modulate susceptibility and progression of such diseases. Considering the adhesion- and colonization-related challenges posed by multiple physiological and pathological characteristics in the intestine, a microenvironment self-adaptive nanoarmor is developed. Partially acetylated chitosan oligosaccharides (CS) were employed to tune the adaptability and responsiveness of nanoarmo

Prevention of HNSCC by chewing gum delivery of enzymes/antimicrobials to reduce carcinogenic acetaldehyde produced by pathogenic microbes.

Acetaldehyde, a WHO Group 1 human carcinogen, produced by oral microbes, plays a critical role at the onset of head and neck squamous cell carcinoma (HNSCC). Candida albicans isolated from different HNSCC patient saliva samples produced similar levels of acetaldehyde; the production level increased in a dose-dependent manner, with the highest level observed at 100 mM glucose or ethanol. Streptococci produced acetaldehyde in ethanol-free media, but the level increased up to 500-1,000 mM alcohol, whereas Fusobacterium nucleatum and Porphyromonas gingivalis showed ethanol-induced acetaldehyde pro

Fluid biomarkers in multiple sclerosis: Indicators of disease activity, progression, and treatment response.

Multiple sclerosis (MS) is a chronic immune-mediated CNS disorder marked by inflammation, demyelination, and neurodegeneration. Standard clinical evaluation, neurological exams, disability scoring, and MRI often miss subclinical activity and poorly predict progression. Fluid biomarkers in cerebrospinal fluid (CSF) and blood provide objective, quantifiable indicators of disease activity, progression, and therapeutic response. Essential fluid biomarkers for monitoring MS include neurofilament light chain (NfL), glial fibrillary acidic protein (GFAP), Chitinase-3-like protein 1 (CHI3L1), C-X-C Mo

Transgenic tomato strategies targeting whitefly eggs from apoplastic or ovary-directed proteins.

BACKGROUND: Transgenic plants expressing proteins that target the eggs of the ubiquitous plant pest Bemisia tabaci (whitefly) could be an effective insecticide strategy. Two approaches for protein delivery are assessed using the mCherry reporter gene in transgenic tomato plants, while accommodating autofluorescence in both the plant, phloem-feeding whitefly and pedicle-attached eggs. RESULTS: Both transgenic strategies were segregated to homozygous genotype using digital PCR. The first strategy uses a glycotransferase secretion signal peptide. Despite bright apoplastic accumulation, mCherry i

Antimycobacterial and healing effects of Pranlukast against MTB infection and pathogenesis in a preclinical mouse model of tuberculosis.

It is essential to understand the interactions and relationships between Mycobacterium tuberculosis (Mtb) and macrophages during the infection in order to design host-directed, immunomodulation-dependent therapeutics to control Mtb. We had reported previously that ornithine acetyltransferase (MtArgJ), a crucial enzyme of the arginine biosynthesis pathway of Mtb, is allosterically inhibited by pranlukast (PRK), which significantly reduces bacterial growth. The present investigation is centered on the immunomodulation in the host by PRK particularly the activation of the host's immune response t
