RSK2 is a modulator of craniofacial development.

BACKGROUND: The RSK2 gene is responsible for Coffin-Lowry syndrome, an X-linked dominant genetic disorder causing mental retardation, skeletal growth delays, with craniofacial and digital abnormalities typically associated with this syndrome. Craniofacial and dental anomalies encountered in this rare disease have been poorly characterized. METHODOLOGY/PRINCIPAL FINDINGS: We examined, using X-Ray microtomographic analysis, the variable craniofacial dysmorphism and dental anomalies present in Rsk2 knockout mice, a model of Coffin-Lowry syndrome, as well as in triple Rsk1,2,3 knockout mutants. W

Roberts syndrome with normal cell division.

Roberts-SC phocomelia syndrome (RS) is an autosomal recessive disorder of symmetric limb defects, craniofacial abnormalities, pre- and postnatal growth retardation, and mental retardation. Patients with RS have been reported to have premature separation of heterochromatin of many chromosomes and abnormalities in the cell-division cycle. We report an infant whose clinical and radiologic findings resemble those of RS but who lacks the cytogenetic and cell division abnormalities reported in RS. This patient may represent a variant of RS or a new syndrome.

Artificial Intelligence Applications in Pediatric Craniofacial Surgery.

Artificial intelligence is rapidly transforming pediatric craniofacial surgery by enhancing diagnostic accuracy, improving surgical precision, and optimizing postoperative care. Machine learning and deep learning models are increasingly used to analyze complex craniofacial imaging, enabling early detection of congenital anomalies such as craniosynostosis, and cleft lip and palate. AI-driven algorithms assist in preoperative planning by identifying anatomical abnormalities, predicting surgical outcomes, and guiding personalized treatment strategies. In cleft lip and palate care, AI enhances pre

Complex Facial Clefts.

The embryology, classification, evaluation, and reconstructive management of complex craniofacial clefts, a group of exceedingly rare and highly variable congenital anomalies, is presented. Using the Tessier system as a structural framework, the characteristic features of each cleft subtype and highlights, key diagnostic considerations, including assessment of soft-tissue deficiency, skeletal disruption, and associated functional concerns are described. Emphasis is placed on the application of fundamental reconstructive principles: respect for facial subunits, restoration of symmetry, and stag

Craniofacial features associated with Hutchinson - Gilford progeria syndrome - A case report.

Hutchinson - Gilford Progeria syndrome (HGPS) is a very rare syndrome characterized by early onset senescence. It is manifested as premature aging with involvement of hair, skin, nail, cardiovascular and bone manifestations. This syndrome has also been reported to be associated with craniofacial anomalies. With regard to these craniofacial phenotypes of Hutchinson - Gilford Progeria syndrome several studies are being undertaken all around the world. Here we present one such rare case of HGPS associated with craniofacial anomaly along with cleft lip and cleft palate in an eleven year old boy.

Heritage, Collaboration, and Innovation: The Developmental Trajectory of Chinese Craniofacial Surgery.

The journey of Chinese craniofacial surgery is a testament to resilience, innovation, and interdisciplinary collaboration. Its origins trace back to the historic spring of 1977, when Professor Zhang Disheng performed China's first intracranial approach orbital hypertelorism correction at Shanghai Ninth People's Hospital. Operating with rudimentary manual instruments in the absence of electric saws or drills, the metallic clinks against bone symbolized not only medical ingenuity but also the courage to break ideological constraints. This high-risk procedure, conducted under basic anesthetic mon

Craniofacial syndromes and class III phenotype: common genotype fingerprints? A scoping review and meta-analysis.

Skeletal Class III (SCIII) is among the most challenging craniofacial dysmorphologies to treat. There is, however, a knowledge gap regarding which syndromes share this clinical phenotype. The aims of this study were to: (i) identify the syndromes affected by the SCIII phenotype; (ii) clarify the involvement of maxillary and/or mandibular structures; (iii) explore shared genetic/molecular mechanisms. A two-step strategy was designed: [Step#1] OMIM, MHDD, HPO, GeneReviews and MedGen databases were explored; [Step#2]: Syndromic conditions indexed in [Step#1] were explored in Medline, Pubmed, Scop

Epidemiological Characteristics of Children With Non-Cleft Lip/Palate Craniofacial Anomalies.

Non-cleft craniofacial anomalies are not as common as cleft lip and palate but resultant disability can be very severe. Although there are epidemiological studies of the clefts in the medical literature, the non-cleft group is still not well known. This study was to examine the epidemiological characteristics of the non-cleft craniofacial anomalies. Patients younger than 18 years old were included during a 1-year period. Patient information was retrieved from medical records and a questionnaire filled by primary caregivers. There were 139 patients included in the study with an average age of 6

Right-Sided Complete Third Branchial Cleft Fistula.

Branchial cleft anomalies occur due to insufficient closure of cavities during embryogenesis. These anomalies consist of cysts, sinuses, and fistulas, with the rarest type being fistulas. A 29-year-old male presented at the authors' clinic with a right-sided complete third branchial cleft fistula. Fistula track excision surgery was successfully performed and no recurrence was observed in the 12-month follow-up after the surgery. While second branchial cleft fistula is the most common, third and fourth brancial cleft fistulas are extremely rare. In addition, they are usually incomplete and almo

Cleft and Craniofacial Coding in ICD-10.

OBJECTIVE: With the gradual replacement of the ICD-9 coding system by the newer ICD-10 system, there is a need to critically evaluate how the system improves-or fails to improve-the coding of specific cleft and craniofacial anomalies. The following review examines the new system and the key components for practitioners who treat patients with such anomalies. CONCLUSIONS: While seemingly beneficial in certain respects, the newer ICD-10 system remains far from ideal for cleft and craniofacial anomalies. With future iterations of the system, a more concerted effort to precisely code such anomali

The use of piezosurgery in cranial surgery in children.

Piezosurgery is an alternative surgical technique, now widely tested, that uses ultrasounds for bone cutting. This device uses ultrasounds to section hard tissues without harming surrounding soft tissues. The authors analyzed their experience in craniomaxillofacial procedures with piezosurgery. A comparison between operation timing and complication rates between piezosurgery and traditional cutting instruments has been performed. A total of 27 patients were examined (15 females and 12 males; average age, of 5.5 months) affected by craniosynostosis. The aim of this study was to analyze the adva

A novel EFNB1 mutation in a patient with craniofrontonasal syndrome and right hallux duplication.

Craniofrontonasal syndrome (CFNS, MIM #304110) is a rare X-linked dominant developmental disorder that shows paradoxically greater severity in affected females than in affected males. Our female patient with frontonasal dysplasia, craniosynostosis and additional malformations was consistent with CFNS. EFNB1, which encodes a member of the ephrin family of transmembrane ligands for Eph receptor tyrosine kinases, is the only gene in which mutation is known to cause CFNS. Here, we describe 402T>C, a novel de novo mutation on EFNB1. This mutation results in substitution of highly conserved isoleuci

Bifid nose - a mild degree of frontonasal dysplasia. A case report.

Frontonasal dysplasia is an unusual congenital condition with a wide phenotypic range. Because of this, only a small number of cases and their management have been reported in the literature. The ideal surgical procedures to correct mild cases of frontonasal dysplasia, and the time to perform them, are still controversial. The case of a 9-month-old girl with a mild form of this condition (a congenital bifid nose and a duplicated frenulum), and its surgical management, is presented. The surgery achieved an early improvement of the patient's appearance and she had no complications. In the future
