CX3CR1-expressing inflammatory dendritic cells contribute to the progression of steatohepatitis.

Liver monocytes play a major role in the development of NASH (non-alcoholic steatohepatitis). In inflamed tissues, monocytes can differentiate in both macrophages and dendritic cells. In the present study, we investigated the role of moDCs (monocyte-derived inflammatory dendritic cells) in experimental steatohepatitis induced in C57BL/6 mice by feeding on a MCD (methionine/choline-deficient) diet. The evolution of steatohepatitis was characterized by an increase in hepatic CD45+ / CD11b+ myeloid cells displaying the monocyte/macrophage marker F4-80(+). In the early phases (4 weeks of treatment

Characterisation of dendritic cell subsets in chronically inflamed human epididymis.

Infection and inflammation of the genital tract are thought to be a primary aetiological factor of male infertility. Chronic epididymitis appears to be more important than prostatitis or seminal vesiculitis due to the direct interaction between sperm cells and epididymal epithelium. Dendritic cells (DCs) are a heterogeneous population of antigen-presenting cells that play a crucial role in the regulation of the immune response and immunological tolerance. The aim of this study was to investigate the expression and characteristic of different DC subsets in chronic inflammation of human epididym

Novel immunotherapeutic approaches in targeting dendritic cells with virus vectors.

Viruses have evolved efficient strategies to overcome cellular membranes and transfer nucleic acid into a host cell. This property is being exploited in gene therapy which has the goal of delivering therapeutic genes into a patient tissue in order to achieve a clinically relevant effect. An interesting target for virus-mediated gene transfer is the immune system. In fact, the first human gene therapy trial performed involved the implantation of autologous bone marrow cells transduced ex vivo with gamma retrovirus vectors expressing adenosine deaminase in a patient with severe combined immunode

EphA7 signaling guides cortical dendritic development and spine maturation.

The process by which excitatory neurons are generated and mature during the development of the cerebral cortex occurs in a stereotyped manner; coordinated neuronal birth, migration, and differentiation during embryonic and early postnatal life are prerequisites for selective synaptic connections that mediate meaningful neurotransmission in maturity. Normal cortical function depends upon the proper elaboration of neurons, including the initial extension of cellular processes that lead to the formation of axons and dendrites and the subsequent maturation of synapses. Here, we examine the role of…

The vagus nerve alters the pulmonary dendritic cell response to injury.

BACKGROUND: We have shown previously that vagal nerve stimulation (VNS) protects against burn-induced acute lung injury (ALI). Although the mobilization and activation of immune cells is central to tissue injury caused by the systemic inflammatory response, the specific inflammatory cell populations that are modulated by VNS have yet to be fully defined. The purpose of this study was to assess whether VNS alters inflammatory cell recruitment to the lung after severe burn injury. MATERIALS AND METHODS: Male C57BL/6 mice were subjected to 30% total body surface area steam burn with and without

Follicular dendritic cell pattern in early lymphomas involving follicles.

This article reviews the most typical follicular dendritic cell (FDC) patterns displayed by early lymphomas involving follicles. In in situ follicular lymphoma, FDCs form a well-developed round (spherical) dendritic meshwork with a sharp outline. Other patterns that can be seen include contracted/distorted/disintegrated FDC meshworks. In early mantle cell lymphoma with mantle zone pattern, FDCs compose an ill-outlined and loosely arranged "centrifugal" meshwork. In marginal zone lymphoma, the FDC meshwork forms a relatively well-developed nodular meshwork with irregular outlines. In nodular ly

Plasmacytoid dendritic cell bactericidal activity against Burkholderia pseudomallei.

Melioidosis sepsis, caused by Burkholderia pseudomallei, is associated with high mortality due to an overwhelming inflammatory response. Plasmacytoid dendritic cells (pDC) are potent producers of type I interferons (IFN). This study investigated whether pDC and type I IFN play a role during the early stages of B. pseudomallei infection. Human and murine pDC internalised and killed B. pseudomallei as efficiently as murine conventional DC (cDC). pDC derived from B. pseudomallei-susceptible (BALB/c) mice demonstrated poor intracellular killing and increased IFN-alpha compared to pDC derived from

An extracellular adhesion molecule complex patterns dendritic branching and morphogenesis.

Robust dendrite morphogenesis is a critical step in the development of reproducible neural circuits. However, little is known about the extracellular cues that pattern complex dendrite morphologies. In the model nematode Caenorhabditis elegans, the sensory neuron PVD establishes stereotypical, highly branched dendrite morphology. Here, we report the identification of a tripartite ligand-receptor complex of membrane adhesion molecules that is both necessary and sufficient to instruct spatially restricted growth and branching of PVD dendrites. The ligand complex SAX-7/L1CAM and MNR-1 function at…

Atlastin-1 regulates dendritic morphogenesis in mouse cerebral cortex.

Hereditary spastic paraplegias (HSPs) are human genetic disorders characterized by lower extremity spasticity and weakness. Mutations in atlastin-1 (ATL1) have been identified in patients with HSP SPG3A. However, the function of ATL1 in the mammalian brain remains unclear. Here, we found that expression of ATL1 mRNA was restricted in the deep layer of mouse cerebral cortex during the early development. We examined ATL1 functions by delivering its plasmids to the upper layer cortical neurons using in utero electroporation. The effects of ectopic expression in the pyramidal neurons were determin

Dendritic cells limit fibroinflammatory injury in nonalcoholic steatohepatitis in mice.

UNLABELLED: Nonalcoholic steatohepatitis (NASH) is the most common etiology of chronic liver dysfunction in the United States and can progress to cirrhosis and liver failure. Inflammatory insult resulting from fatty infiltration of the liver is central to disease pathogenesis. Dendritic cells (DCs) are antigen-presenting cells with an emerging role in hepatic inflammation. We postulated that DCs are important in the progression of NASH. We found that intrahepatic DCs expand and mature in NASH liver and assume an activated immune phenotype. However, rather than mitigating the severity of NASH,

Interleukin-15 dendritic cells as vaccine candidates for cancer immunotherapy.

Owing to their professional antigen-presenting capacity and unique potential to induce tumor antigen-specific T cell immunity, dendritic cells (DCs) have attracted much interest over the past decades for therapeutic vaccination against cancer. Clinical trials have shown that the use of tumor antigen-loaded DCs in cancer patients is safe and that it has the potential to induce anti-tumor immunity which, in some cases, culminates in striking clinical responses. Unfortunately, in a considerable number of patients, DC vaccination is unable to mount effective anti-tumor immune responses and, if it

Silencing TNF-α in macrophages and dendritic cells for arthritis treatment.

OBJECTIVES: Tumour necrosis factor (TNF)-α secreted by macrophages and dendritic cells (DCs) plays a predominant role in arthritis. Our previous studies suggest that a small peptide, RVG-9R (29-aa peptide derived from the rabies virus glycoprotein, fused to 9R residues), can deliver small interfering RNA (siRNA) to macrophages and DCs. We therefore tested whether knockdown of TNF-α expression in macrophages and DCs by RVG-9R/bound siRNA targeting TNF-α reduces the severity of collagen antibody-induced arthritis (CAIA) in mice. METHOD: Arthritis was induced in mice by injecting a combination o

Impact of TRPV3 on the development of allergic dermatitis as a dendritic cell modulator.

The transient receptor potential channel vanilloid subfamily V member 3 (TRPV3), which functions as a thermosensor in keratinocytes, plays an important role in the development of allergic and itchy dermatitis in rodents. Although real-time PCR analysis using lesional and non-lesional skin samples from patients with atopic dermatitis showed that TRPV3 was expressed in lesional skin, the role that TRPV3 plays in patients with dermatitis is still relatively obscure. Here, we determined whether TRPV3 was a dendritic cell (DC) modulator using DS-Nh mice with a gain-of-function mutation in TRPV3 (TR

Dendritic cell immunoreceptor regulates Chikungunya virus pathogenesis in mice.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus responsible for recent epidemic outbreaks of debilitating disease in humans. Alphaviruses are known to interact with members of the C-type lectin receptor family of pattern recognition proteins, and given that the dendritic cell immunoreceptor (DCIR) is known to act as a negative regulator of the host inflammatory response and has previously been associated with rheumatoid arthritis, we evaluated DCIR's role in response to CHIKV infection. Although we observed an increase in the proportion of dendritic cells at the site of CHIKV infectio

Plasmacytoid dendritic cells reduce HIV production in elite controllers.

HIV elite controllers (EC) are a rare group of HIV-infected patients who are able to maintain undetectable viral loads during a long period of time in the absence of antiretroviral treatment. Adaptive immunity and host genetic factors, although implicated, do not entirely explain this phenomenon. On the other hand, plasmacytoid dendritic cells (pDCs) are the principal type I interferon (IFN) producers in response to viral infection, and it is unknown whether pDCs are involved in the control of HIV infection in EC. In our study, we analyzed peripheral pDC levels and IFN-α production by peripher

Bilateral conjunctival lesions in blastic plasmacytoid dendritic cell neoplasm.

The purpose of this study is to report on a patient who developed conjunctival lesions of blastic plasmacytoid dendritic cell neoplasm (BPDCN) after radiation to skin lesions of the same pathological type. A 79-year-old man developed salmon-pink lesions in the lower fornix of the conjunctiva of both eyes and biopsy revealed BPDCN. One and a half years previously, he noticed an erythematous plaque with a 30-mm diameter, which later became multiple, on the left chest, and the biopsy revealed BPDCN. The bone marrow was negative for CD56-positive cells, but fluorescence-activated cell sorting anal
