In vitro duodenal lipolysis of lipid-based drug delivery systems studied by HPLC-UV and HPLC-MS.

Oral drug delivery systems based on lipids are biodegraded in a process called lipolysis to release free fatty acids and monoglycerides. The rate of this lipolysis is usually measured by pH titration. Nevertheless, this technique has some limitations, such as not providing any information about the actual composition of the lipolytic products. In this study, we propose a method to analyze these products during and after lipolysis using HPLC. For the first time, HPLC-UV and HPLC-MS have been used to investigate in vitro duodenal lipolysis of long- and medium-chain triglycerides nanoemulsions. T…

Glucose-, pH- and thermo-responsive nanogels crosslinked by functional superparamagnetic maghemite nanoparticles as innovative drug delivery systems.

Reversibly crosslinked (RCL) nanogels made of thermo-responsive poly(vinyl alcohol)-b-poly(N-vinylcaprolactam) copolymers were combined with maghemite nanoparticles and developed as new drug delivery systems (DDS). The crosslinking was formed via boronate/diol bonding from the surface-functionalized superparamagnetic maghemite nanoparticles, endowing the DDS with thermo-, pH- and glucose-responsiveness. The capability to load a hydrophobic drug model Nile red (NR) within the RCL nanogels was evaluated, and stimuli-triggered drug release behaviours under different conditions were tested. Zero p…

Skin-compatible lecithin drug delivery systems for fluconazole: effect of phosphatidylethanolamine and oleic acid on skin permeation.

The purpose of the present study was to evaluate skin-compatible drug delivery systems for fluconazole. Pseudoternary phase diagrams were constructed, composed of different soybean lecithins/oil/isopropanol and water. The role of the various lecithin compositions was expressed in the different resulting isotropic areas. Based on these phase diagrams, two systems were chosen as drug delivery systems for fluconazole. The influence of phosphatidylethanolamine and of the oil component on the skin permeation of fluconazole was investigated. The more phosphatidylethanolamine, the greater was the flu…

Polymers-gamma ray interaction. Effects of gamma irradiation on modified release drug delivery systems for oral administration.

The aim of this work is to verify the efficiency of two kinds of matrix tablets formulations containing PEO or PVA as retarding polymer. Moreover, since in the last years the exposure to ionizing radiation is a more and more used method to reduce bacterial charge in pharmaceutical products, the effects of gamma irradiation on these two kinds of polymers has been evaluated. The study is performed on matrix tablets containing diltiazem HCl, as model drug, and polyethylene oxides (PEO) of two different molecular weights or polyvinylalchool (PVA) of medium degree of hydrolysis, as drug release mod

Redox-responsive oral drug delivery systems for inflammatory bowel disease: Analytical challenges, design strategies, and translational outlook.

Inflammatory bowel disease (IBD) is a multifactorial chronic inflammatory illness, kind a thing that involves immune dysregulation, a weakened epithelial barrier, microbiota alterations, genetic susceptibility, and of course environmental influences. In the downstream pathological cascade, oxidative stress has come up as a major driver behind mucosal injury, the whole inflammatory upscaling, and further disease progression. Because of that, higher concentrations of reactive oxygen and nitrogen species, ROS/RNS, inside the inflamed intestinal tissues have started getting attention as natural en

Closed-loop systems for automated hypnotic drug delivery during general anaesthesia: a systematic review and meta-analysis.

BACKGROUND: Closed-loop systems for automated hypnotic delivery integrate continuous bispectral index (BIS) monitoring with controller algorithms to adjust infusion rates in real time. Unlike earlier syntheses that focused on aggregate control metrics, we frame anaesthetic depth as an asymmetric risk problem by separating exposure to potentially inadequate anaesthesia from exposure to excessive depth of anaesthesia. METHODS: This PROSPERO-registered systematic review (CRD420251208039) searched PubMed, Embase, Cochrane CENTRAL, and Web of Science through November 2025. Randomised trials compar

Honeysuckle-derived vesicle-like nanoparticle and their hybrid vesicle as novel drug delivery systems for glioma therapy.

Gliomas present a formidable challenge in oncology due to their immunosuppressive tumor microenvironment and the restricted delivery of therapeutics across the blood-brain barrier. Here, we report a novel hybrid nanoplatform (HEV) for synergistic chemo-immunotherapy, constructed by integrating honeysuckle-derived vesicle-like nanoparticles (HDVN) with paclitaxel (PTX)-loaded liposomes via PEG-mediated fusion. HDVN, extracted from Lonicera japonica Flos using sucrose gradient ultracentrifugation, measured 104 ± 2.1 nm in diameter and carried functional miRNAs, including miRNA2911, capable of mo

Nanoparticle based drug delivery systems improve antimalarial efficacy and vaccine performance in African research and clinical settings.

BACKGROUND: Malaria is a significant public health problem in sub-Saharan Africa, responsible for a large proportion of the world's malaria cases and deaths. Antimalarial drug resistance and the low efficacy of existing drugs, due to poor solubility, bioavailability, short half-life and non-specific distribution, highlight the importance of the development of better delivery strategies. The current systematic review focuses on the platforms that were developed or evaluated in Africa (2015-2025) involving nanoparticles. METHODS: PubMed, Web of Science, and Scopus were searched for original exp

Polyglycerol-Based Lipids: A Next-Generation Alternative to PEG in Lipid Nanoparticles for Advanced Drug Delivery Systems.

Polyethylene glycol (PEG) is widely used to provide a stealth effect in various therapeutic applications. However, an increased occurrence of anti-PEG antibodies in patients can lead to immunological side effects and accelerated blood clearance, reducing the efficiency of PEG-based delivery vectors. One of these PEG-containing vectors is lipid nanoparticles (LNPs), which effectively deliver nucleic acids. Their high potential for treating a great number of diseases in the future was already showcased in the mRNA vaccines developed during the COVID-19 pandemic. Thus, the aim of our study was to
