One-trial tolerance to the effects of chlordiazepoxide in the elevated plus-maze is not due to acquisition of a phobic avoidance of open arms during initial exposure.

A single exposure to the elevated plus-maze (EPM) test of anxiety reduces or abolishes the anxiolytic-like efficacy of benzodiazepines. This phenomenon called one-trial tolerance has been suggested to represent the acquisition of a phobic-like response to the open arms during trial 1. The present study was designed to examine the effects of chlordiazepoxide (5 mg/kg, ip) on the behaviour of rats in a conventional EPM apparatus after previous exposure to a four-open-arm EPM, a four-enclosed arm EPM or a conventional EPM, as well as in naive rats. Chlordiazepoxide had clear-cut anxiolytic-like e

Naloxone potentiates anxiolytic-like actions of diazepam, pentobarbital and meprobamate but not those of Ro19-8022 in the rat.

The elevated plus-maze test was used to determine if the opiate antagonist naloxone could potentiate the anxiolytic-like effects of the benzodiazepine diazepam, the barbiturate pentobarbital, the propanediol carbamate meprobamate and the partial benzodiazepine receptor agonist [R]-1-[(10-chloro-4-oxo-3-phenyl-4H-benzo[a]quinolizin-1-yl) carbonyl]-2-pyrrolidine-methanol (Ro19-8022) in the rat. A subeffective dose of each of these compounds was combined with naloxone, 10 mg/kg. Naloxone had no effect by itself, but potentiated all drugs except Ro19-8022. The proportion of entries on the open arm

Hyposalivation induced by salivary gland extraction impairs cognitive function in mice.

Hyposalivation affects cognitive function. However, its impact on hippocampus-dependent memory remains unclear. Saliva contains brain-derived neurotrophic factor (BDNF), which is also synthesized in the hippocampus and can pass through the blood-brain barrier (BBB) to influence hippocampal plasticity. Therefore, we hypothesized that hyposalivation reduces peripheral BDNF availability, leading to decreased hippocampal BDNF levels and cognitive impairment. In this study, this relationship was investigated using an in vivo model of sialadenectomy-induced hyposalivation. A total of 24 8-week-old m

The Impacts of FLASH Radiation Therapy and Conventional Radiation Therapy on the Cognitive Abilities of Mice.

PURPOSE: Preclinical studies have demonstrated that FLASH radiation therapy (RT) delivered at ultrahigh-dose rates exerts a preferential normal tissue-sparing effect. This study aimed to delineate the disparities in delayed cognitive function and biological outcomes of whole-brain irradiation in healthy mice, comparing FLASH-RT with conventional RT (CONV-RT). METHODS AND MATERIALS: Eighty adult male C57BL/6J mice were divided into 5 groups: Sham, CONV-RT10Gy, CONV-RT20Gy, FLASH-RT10Gy, and FLASH-RT20Gy. Whole-brain irradiation was conducted on mice using a miniaturized x-ray FLASH platform at

Effects of low concentration of fluoride exposure during fetal on behavior and neurotransmitters in adult mice.

Fluoride (F) naturally occurs in water in China and India, and in excess, can cause skeletal fluorosis and mottled teeth. Chronic exposure to F during gestation can affect the development of the brain, reducing intelligence quotient and inducing autism spectrum disorder-like behavior. In the present study, it was aimed to clarify the effects of chronic exposure to low concentrations of F in utero on brain function. The behavior was assessed, the levels of brain neurotransmitters were measured in mice and their relationships were analyzed. ICR mice consumed water containing sodium fluoride (F c

Withaferin A protects against epilepsy by promoting LCN2-mediated astrocyte polarization to stopping neuronal ferroptosis.

BACKGROUND: Epilepsy is among the most frequent severe brain diseases, with few treatment options available. Neuronal ferroptosis is an important pathogenic mechanism in epilepsy. As a result, addressing ferroptosis appears to be a promising treatment approach for epilepsy. Withaferin A (WFA) is a C28 steroidal lactone that has a broad range of neuroprotective properties. Nonetheless, the antiepileptic action of WFA and the intrinsic mechanism by which it inhibits ferroptosis following epilepsy remain unknown. PURPOSE: This study aimed at investigating to the antiepileptic potential of WFA in

5-HT7 receptor activation rescues impaired synaptic plasticity in an autistic-like rat model induced by prenatal VPA exposure.

Autism spectrum disorder (ASD) is a severe life-long neuropsychiatric disorder. Alterations and imbalance of several neurochemical systems may be involved in ASD pathophysiology, of them, serotonergic neurotransmission dysfunction and deficiency may underlie behavioral abnormalities associated with ASD. However, the functional importance of serotonergic receptors, particularly 5HT7 receptors in ASD pathology remains poorly defined. Serotonin receptor subtype 7 (5-HT7R) plays a direct regulatory role in the development and also for the mature function of the brain, therefore, further studies ar

Synergistic effects of capsaicin and ketamine on the depression-like behaviors of animals subjected to the chronic unpredictable mild stress protocol.

A significant percentage of depression patients are resistant to pharmacological treatments, highlighting the need for novel antidepressant strategies. Ketamine, an NMDA receptor antagonist, produces rapid antidepressant effects and is effective in treatment-resistant cases, but its association with cognitive and behavioral abnormalities resembling schizophrenia limits broader clinical utility. Capsaicin, a TRPV1 agonist, alone or combined with standard antidepressants, reduces immobility in the forced swim test (FST) in rodents. This study evaluated the effects of capsaicin and ketamine, isol

Functional Remodeling of the Maternal Brain: Linking Brain c-Fos Dynamics to Behavioral Changes in Pregnancy: An Exploratory Study.

As an exploratory study with a limited sample size, this work systematically examined pregnancy-induced changes in neural activity by analyzing c-Fos expression across the whole brain of pregnant mice. The results showed that 36 brain regions-including the cortex, hippocampus, thalamus, and hypothalamus-exhibited dynamic changes in c-Fos expression throughout pregnancy. Behavioral tests revealed that pregnant mice displayed increased anxiety-like behavior in the elevated plus maze. Group-level descriptive correlation analysis indicated nominal directional trends between regional c-Fos expressi

Anxiolytic-like effects of a new 1-N substituted analog of melatonin in pinealectomized rats.

In spite of the wide variety of drugs available for treating anxiety, this disorder continues to represent a worldwide health problem that is classified within the first 10 causes of disability. Therefore, the search continues for new antianxiety agents, particularly those not related to benzodiazepines. Even though melatonin has been prescribed as an anxiolytic drug, its use is currently limited due to its short half-life and photo-sensitivity, among other disadvantages. The present study explores the antianxiety properties of a new 1-N substituted melatonin analog, M3C, in pinealectomized ra

Modeling stress-induced social and anxiety-like behaviors in mice: Towards a preclinical framework for Munchausen-like traits.

Munchausen syndrome is a psychiatric condition in which individuals feign or induce illness to gain attention, care, and sympathy. Despite being clinically recognized, its biological underpinnings remain largely unexplored, highlighting the need for suitable animal models. Male mice were subjected to social isolation and restraint stress (2 h daily for 7 days) to induce chronic stress. Behavioral changes were assessed using a social interaction test with freely moving or confined juvenile and adult male conspecifics. Anxiety-like behaviors were evaluated by the open field test and elevated plu

Hesperidin mitigates cognitive and anxiety-like deficits by enhancing hippocampal antioxidant defenses, reducing neuroinflammation, and preventing neuronal apoptosis in a third-trimester-equivalent rat model of developmental ethanol neurotoxicity.

Ethanol exposure during brain development has been associated with hippocampal oxidative stress, neuroinflammation, and apoptosis, resulting in lasting cognitive and emotional deficits characteristic of fetal alcohol spectrum disorders (FASD). This study evaluated the neuroprotective potential of Hesperidin, a citrus flavanone with antioxidant and anti-inflammatory actions, against developmental ethanol neurotoxicity. Neonatal Wistar rats received ethanol (5.25 g/kg/day; 11.9% v/v) from postnatal days 2-10 and were subsequently treated intraperitoneally with Hesperidin (25, 50, or 100 mg/kg).

Introducing Female Black Swiss Mice: Minimal Effects of Sex in a Strain-Specific Battery of Tests for Mania-Like Behavior and Response to Lithium.

Black Swiss (BS) mice were shown to be an advantageous strain to model behavioral domains of mania, but to date only male mice were tested, whereas bipolar disorder (BPD) is equally prevalent in women and men. This study was therefore designed to examine the possibility of using both male and female BS mice in future studies. Groups of male and female BS mice were compared with each other, with or without lithium treatment, in tests for domains of mania-like behavior including activity in an open field, sweet solution preference, elevated plus maze, forced swim and amphetamine-induced hyperact
