Engineering clinically actionable nanomedicine for cancer immunometabolic ecosystem reprogramming.

Traditional metabolic inhibitors have limited clinical efficacy, with issues such as narrow window periods, and compensatory effects. These barriers indicate that effective metabolic therapy requires spatially restricted, temporally coordinated, cell-context-aware, and clinically stratifiable intervention. This review examines how glucose-lactate, lipid-redox, amino acid-immune checkpoint, and nucleotide-adenosine circuits drive tumour progression, immune suppression, and therapeutic escape. We then define the engineering advantages that distinguish nanomedicine from free metabolic drugs, incl…

Engineering organs-on-a-chip via multi-channel microfluidics.

Conventional in vitro physiological models, relying on animal studies and 2D/3D cell cultures, are fundamentally limited by interspecies biological discrepancies, ethical constraints, or inadequate replication of human physiology. Organ-on-a-chip (OoC) technology overcomes these challenges through emulating organ-specific microphysiological systems. The transformative power of this innovation lies in multi-channel microfluidic chips. These chips facilitate the formation of 3D cellular organizations and tissue interfaces via integrated porous membranes, micropillar arrays, or perfusable vascula

In vivo site-specific engineering to reprogram T cells.

Engineered T cells, reprogrammed to express chimeric antigen receptors (CAR) or T cell receptors (TCR), have transformed cancer treatment and are being explored as therapeutics for autoimmune and infectious diseases. Enhancing T cell function through genome editing, either by disrupting endogenous genes or precisely inserting DNA payloads, has shown considerable promise1. However, the ex vivo manufacturing process is lengthy and costly, limiting accessibility of these therapies. In vivo generation of CAR T cells could overcome these barriers, but current methods rely either on transient expres

Integrated Engineering of CAR-T Cells for Solid Tumours.

Chimeric antigen receptor (CAR) T-cell therapy has achieved durable efficacy in hematologic malignancies but encounters persistent obstacles in solid tumours, including antigen heterogeneity, a suppressive tumour microenvironment (TME), and intrinsic T-cell dysfunction. This review examines the transition from single-axis engineering to an integrated framework that addresses these hurdles in sequence. We delineate how next-generation CAR-T cells are designed for precise spatiotemporal activation through logic-gated and pharmacologically regulatable receptors, while being reinforced by metaboli…

Precision Engineering of Evolution-Resilient Rice against Bacterial Blight.

The persistent conflict between rice and Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of bacterial blight, exemplifies a dynamic genetic arms race in agriculture. The cyclical deployment and erosion of major resistance (R) genes highlight the high adaptive potential of Xoo and the need for strategies that are durable rather than absolute. This review synthesizes a paradigm shift from reactive, single R-gene deployment toward proactive engineering of evolution-resilient resistance. We explore the molecular-genetic basis of Xoo adaptability, including TAL effector diversification, non-T…

Systems metabolic engineering for hydroxytyrosol production in Escherichia coli.

Hydroxytyrosol (HT), the primary functional component of olive oil, presents significant challenges for microbial biosynthesis due to its inherent chemical properties and fermentation requirements. In this study, we describe the modular engineering of HT production in Escherichia coli (E. coli). We initially employed a highly efficient 4-hydroxyphenylacetic acid 3-monooxygenase (encoded by the HpaBC gene) from E. coli, phenylpyruvate decarboxylase (encoded by the ARO10 gene), and alcohol dehydrogenase (encoded by the ADH6 gene) from Saccharomyces cerevisiae (S. cerevisiae) to establish the de …
