Sperm angiotensin-converting enzyme activity in Chernobyl victims and patients with chronic prostatitis.

The method for determination of angiotensin-converting enzyme activity in human spermatozoa and seminal plasma was developed. The number of total and active-motile spermatozoa was shown to be lower in Chernobyl victims than in healthy donors. The specific activity of angiotensin-converting enzyme in spermatozoa from Chernobyl victims was 12 times higher than that in donors. Similar results, although to a lesser extent, were observed in patients with chronic prostatitis. In contrast, the enzyme activity in blood serum and seminal plasma was identical for all subjects investigated.

Alterations in mitochondrial DNA and enzyme activities in hypertrophied myocardium of stroke-prone SHRS.

To clarify the pathophysiological alteration of mitochondria in SHRSP hypertrophied heart, mitochondria-related enzyme changes were examined and compared to those in WKY. Furthermore, the structure alteration in mitochondrial DNA (mtDNA) was examined by restriction fragment length polymorphisms (RFLPs). Both isocitrate dehydrogenase (ICDH) and cytochrome c oxidase (COX), which are related to energy production or the respiratory chain in mitochondria, were significantly lower in SHRSP myocardium than in WKY. Furthermore, superoxide dismutase (SOD), a potent radical scavenger, was also lower in

Antioxidant enzyme activities in different brain areas of the neurological mutant--pt rabbit.

Cu, Zn-superoxide dismutase (Cu,Zn-SOD), Mn-superoxide dismutase (Mn-SOD), catalase (CAT), peroxidase (POX), glutathione peroxidase(GP) and glutathione reductase (GR) activities were assayed in the brains of genetically selected neurological mutant rabbits pt and their controls. Paralytic tremor (pt) is a spontaneous mutation in rabbit that affects irregular and defective myelination of CNS. Antioxidant enzyme levels were different in three brain regions: brain hemispheres, cerebellum, and brain stem. In brain hemisphere and cerebellum of pt rabbits Mn-SOD and Cu, Zn-SOD activities were elevat

Comparison of erythrocyte antioxidant enzyme activities and embryologic level of neural tube defects.

Increased exposure to oxidant-derived free radicals or inadequate systems for antioxidant defense could alter cellular response at critical points in development. We measured 5 antioxidant enzymes, glutathione peroxidase (GSH-Px), glutathione reductase, glutathione-S-transferase, catalase and superoxide dismutase in erythrocytes and their plasma cofactor trace elements (Se, Zn, Cu) in 37 children with myelomeningocele and in 37 age-matched controls. We placed the patients into 3 groups according to motor level of the lesion at birth. We found significantly lower GSH-Px activities (p = 0.007) i

Reduced activities of thiamine-dependent enzymes in brains of alcoholics in the absence of Wernicke's encephalopathy.

The relative roles of alcohol per se, thiamine deficiency, and liver disease in the pathogenesis of alcohol-related brain damage have not been fully elucidated. In particular, the extent to which alterations of brain thiamine metabolism contribute to cognitive dysfunction in alcoholism in the absence of Wernicke's encephalopathy has not been established. In the present study, thiamine diphosphate-dependent enzymes were measured using standard spectrophotometric techniques in homogenates of brain tissue obtained at autopsy from eight alcoholic patients, all of whom died in hepatic coma without

Bursal antisteroidogenic peptide alters the activity of steroidogenic enzymes in chicken granulosa cells.

We have previously reported that a peptide from chicken bursa of Fabricius, bursal antisteroidogenic peptide (BASP), inhibits luteinizing hormone-stimulated progesterone biosynthesis by chicken ovarian granulosa cells. The objective of this study was to determine the site(s) of BASP inhibition within the steroidogenic pathway of chicken granulosa cells. The effects of BASP on key steroidogenic enzymes, including adenylyl cyclase (AC), phosphodiesterase, the cholesterol side-chain cleavage enzyme complex and 3 beta-hydroxysteroid dehydrogenase were determined. Luteinizing hormone (10 ng/tube) s

Effect of subthalamic nucleus lesion on mitochondrial enzyme activity in rat basal ganglia.

The subthalamic nucleus (STN) plays a major role in the control of basal ganglia output, and its overactivity may be central to the symptoms of Parkinson's disease. In order to elucidate the functional relationship between STN and its projection nuclei, we studied the short-term (1 week) effect of a selective lesion of STN on the activity of succinate dehydrogenase (SDH) and cytochrome oxidase (CO), two markers of neuronal activity, in the basal ganglia of rats. STN ablation induced a discrete reduction of oxidative metabolism, ipsilaterally to the lesion, in substantia nigra pars reticulata a

Localization of drug-metabolizing enzyme activities to blood-brain interfaces and circumventricular organs.

The brain, with the exception of the choroid plexuses and circumventricular organs, is partially protected from the invasion of blood-borne chemicals by the specific morphological properties of the cerebral micro-vessels, namely, the tight junctions of the blood-brain barrier. Recently, several enzymes that are primarily involved in hepatic drug metabolism have been shown to exist in the brain, albeit at relatively low specific activities. In the present study, the hypothesis that these enzymes are located primarily at blood-brain interfaces, where they form an "enzymatic barrier," is tested.

[Serum activity of the lactate dehydrogenase enzyme in patients with human immunodeficiency virus infection].

FOUNDATION: Many patients with infection by the HIV show an increase in the levels of the lactodehydrogenase (LCH) enzyme. In most of these cases, such increase has been associated to the presence of pneumonia by Pneumocystis carinii. In order to analyze other possible causes of this increase, we conducted a study in which the clinical, analytical and immunological condition of a group of HIV+ patients with increased levels of LDH was compared to another group whose values were within normal limits. MATERIAL AND METHODS: The clinical records of 200 patients with HIV infection were retrospecti

[Antioxidant activity of blood serum and blood enzymes in meningococcal infections in children and adults].

Aspartate transaminase activity (AST) was measured in the serum of children and adults suffering from generalized meningococcal infection. Activation of AST is only rarely detected in children, which is associated with higher protection from lipid peroxidation. The ratio between antioxidants and prooxidants was higher in children than in adults.

Increased mucosal antioxidant enzyme activities in chronic gastritis and benign gastric polyps.

Gastroscopy with gastric biopsy was performed in 109 individuals aged 25-71 years. Activities of three antioxidant enzymes were assayed in biopsy specimens: Cu/Zn superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px). Patients were classified according to the endoscopic and histological findings in the following groups: normal findings (N), superficial gastritis (SG), mild (MAG) and severe (SAG) atrophic gastritis, gastritis after partial gastrectomy (PGG), hyperplastic polyp (HP), and gastric adenoma (A). Compared with the N group, increased activity of SOD was found

The in vivo inhibition of transport enzyme activities in different organs of rat by chlorpromazine is reversible.

Chlorpromazine, an antipsychotic drug is found to inhibit Na+K(+)-ATPase, Ca(2+)-ATPase and acetylcholinesterase activities in the microsomal membranes of rat in vivo, when the drug is injected for certain periods of time. The inhibition seems to be due to the changes in fatty acid composition of lipid and microviscosity of the membranes. However, once the drug has been withdrawn, the enzyme activities are found to return to the normal level in three to five weeks, suggesting that the drug effect is reversible.

Effects of thiamine deficiency on hepatic cytochromes P450 and drug-metabolizing enzyme activities.

To elucidate the mechanisms by which thiamine deficiency affects hepatic microsomal monooxygenase activities, the effect of thiamine deficiency on two constitutive cytochrome P450 isozymes, P450IIE1 and P450IIC11, was investigated, using weanling male Sprague-Dawley rats. The clinical signs of thiamine deficiency were apparent after feeding a thiamine-deficient diet for 3 weeks. Thiamine deficiency caused an increase in P450IIE1, which was determined by N-nitrosodimethylamine demethylase assay and immunoquantitation of P450IIE1. This increase in the P450IIE1 level was mainly attributed to thia

[Prevention of cAMP stimulation of activity of various oxidative mitochondrial enzymes with lidocaine and aminazine].

Lidocaine or aminazine (chlorpromazine) injected to rats or incubated with liver mitochondria prevented NAD-isocitrate dehydrogenase and transhydrogenase activation during the subsequent incubation of mitochondria with cAMP. Integrity of mitochondrial membranes is obligatory for manifestation of the drugs effect. Lidocaine and aminazine-induced desensitization of mitochondria to the cAMP action was apparently realized via alteration in the state (and/or composition) of lipid component of mitochondria. The significance of mitochondria sensitivity to cAMP stimulation is discussed.

Acute in vitro thyrotropin regulation of lysosomal enzyme activity in the thyroid follicular cell.

The acute effect of a physiological concentration (1 mU/l) of thyrotropin (TSH) on the activity of four lysosomal enzymes in the thyroid follicular lining cell has been studied by quantitative cytochemical techniques. N-acetyl-beta-glucosaminidase (NAG) activity was increased by 14% after 10 min TSH stimulation and NAG and beta-galactosidase activities were increased by 24% and 25% respectively (P less than 0.05) after 20 min stimulation and by 40% and 45% (P less than 0.05) respectively after 30 min stimulation with TSH, indicating an early processing of these carbohydrate residues in thyrogl

Prenatal ethanol exposure alters steroidogenic enzyme activity in newborn rat testes.

We have examined the in utero effects of ethanol exposure on testicular steroidogenesis in newborn male pups. Pregnant Sprague-Dawley rats were fed a liquid ethanol diet (35% ethanol-derived calories), a pair-fed isocaloric liquid diet, or a standard laboratory rat chow and water diet beginning on Day 12 of gestation and continuing through parturition. Although there were no significant differences in the enzymatic activity of 5-ene-3 beta-hydroxysteroid dehydrogenase/isomerase or C17,20-lyase, the enzymatic activity of 17 alpha-hydroxylase was significantly (p less than 0.01) reduced (i.e., a

Heterogeneity of glycolytic enzyme activity and isozyme composition of pyruvate kinase in breast cancer.

In 6 patients with breast cancer - of whom specimens of the primary tumor as well as one of its metastases were available for examination - we demonstrated intratumoral and intertumoral heterogeneity in expression of activity of the glycolytic enzymes hexokinase, phosphofructokinase, aldolase, enolase and pyruvate kinase. Heterogeneity also existed in isozyme composition of pyruvate kinase. The transition of the tumors towards normal surrounding breast tissue showed either a sharp drop in activity, or a gradual decrease in activity, corresponding to pushing margins or infiltrative growth of th

Chlorinated hydrocarbon-induced peroxisomal enzyme activity in relation to species and organ carcinogenicity.

Trichloroethylene (TCE), perchloroethylene (PER), and pentachloroethane (PENT) are widely used industrial chemicals that cause an increased incidence of hepatocellular carcinoma in mice and a very low incidence of renal tubular adenocarcinoma in rats. A recent study (C. R. Elcombe, M. S. Rose, and I.S. Pratt (1985), Toxicol. Appl. Pharmacol. 79, 365-376) suggested that the species difference in the hepatocarcinogenicity of TCE seen between rats and mice was due to a species difference in peroxisomal proliferation and cell proliferation. The purpose of the present investigation was to understan
