Gαi1/3 mediate Netrin-1-CD146-activated signaling and angiogenesis.

Netrin-1 binds to the high-affinity receptor CD146 to activate downstream signaling and angiogenesis. Here, we examine the role and underlying mechanisms of G protein subunit alpha i1 (Gαi1) and Gαi3 in Netrin-1-induced signaling and pro-angiogenic activity. In mouse embryonic fibroblasts (MEFs) and endothelial cells, Netrin-1-induced Akt-mTOR (mammalian target of rapamycin) and Erk activation was largely inhibited by silencing or knockout of Gαi1/3, whereas signaling was augmented following Gαi1/3 overexpression. Netrin-1 induced Gαi1/3 association with CD146, required for CD146 internalizati

Patterns and localization of gene expression during intramembranous bone regeneration in the rat femoral marrow ablation model.

Tissue formation and repair are dependent upon cascades of biological events, but the signals involved and the possible gene coexpression patterns during intramembranous bone repair are only poorly understood. We sought to place this mode of regeneration in context by profiling quantitative gene expression for a panel of 39 genes between days 1 and 14 following rat femoral marrow ablation. In situ hybridization was employed to localize a subset of genes. Additionally, principal components analysis was conducted to identify underlying factors suggestive of coexpression patterns. During inflamma

Exercise as a Molecular Therapeutic Strategy in Metabolic Syndrome: Integrating Cellular Signaling, Organ Crosstalk, and Clinical Translation-A Narrative Review.

Metabolic syndrome (MetS) is a clinical condition defined by the coexistence of interconnected cardiometabolic risk factors, including central obesity, dyslipidemia, elevated blood pressure, and impaired glucose regulation, which collectively increase the risk of type 2 diabetes mellitus and cardiovascular disease. Beyond these clinical diagnostic features, MetS is characterized by complex pathophysiological alterations involving systemic dysregulation of metabolic signaling across adipose tissue, skeletal muscle, liver, vascular endothelium, and the immune system. Key molecular alterations in

FAM172A deletion aggravates high fat diet-induced MASLD via the eIF2α-ATF4-FGF21 loop.

AIMS: Inhibition of endoplasmic reticulum stress (ERS) can effectively improve the progression of metabolic dysfunction-associated steatotic liver disease (MASLD). In this study, we attempted to further explore the mechanism of Fam172a in high fat diet (HFD) induced-MASLD. MATERIALS AND METHODS: The correlation between FAM172A and ERS-induced MASLD was tested by WB and qRT-PCR. We utilized wild type (WT) and Fam172a gene knockout (Fam172a-/-) mice to build two models: MASLD model induced by HFD and ERS model induced by tunicamycin (Tm). We evaluated the degree of liver inflammation, steatosis

A rat model of metabolic syndrome-related heart failure with preserved ejection fraction phenotype: pathological alterations and possible molecular mechanisms.

BACKGROUND: Heart failure with preserved ejection fraction (HFpEF) represents a syndrome involving multiple pathophysiologic disorders and clinical phenotypes. This complexity makes it challenging to develop a comprehensive preclinical model, which presents an obstacle to elucidating disease mechanisms and developing new drugs. Metabolic syndrome (MetS) is a major phenotype of HFpEF. Thus, we produced a rat model of the MetS-related HFpEF phenotype and explored the molecular mechanisms underpinning the observed pathological changes. METHODS: A rat model of the MetS-related HFpEF phenotype was

7-ketocholesterol induces endothelial-mesenchymal transition and promotes fibrosis: implications in neovascular age-related macular degeneration and treatment.

Oxidized cholesterols and lipids accumulate in Bruch's membrane in age-related macular degeneration (AMD). It remains unknown what causal relationship exists between these substances and AMD pathophysiology. We addressed the hypothesis that a prevalent form, 7-ketocholesterol (7KC), promotes choroidal endothelial cell (CEC) migration and macular neovascularization in AMD. Compared to control, 7KC injection caused 40% larger lectin-stained lesions, but 70% larger lesions measured by optical coherence tomography one week after laser-injury. At two weeks, 7KC-injected eyes had 86% larger alpha sm

Inhibition of Pathological Phenotype of Hypertrophic Scar Fibroblasts Via Coculture with Adipose-Derived Stem Cells.

Hypertrophic scar (HS) is a dermal fibroproliferative disease characterized by fibroblast over-proliferation, overproduction, and deposition of the extracellular matrix. Growing evidence demonstrated that adipose-derived stem cells (ASCs) secrete a plethora of trophic and antifibrotic factors, which suppress inflammation and ameliorate fibrosis of different tissues. However, few studies investigate their effect on repressing HS activity. This study evaluated the suppressing effect of ASCs on HS fibroblast bioactivity and the possible mechanism via a coculture model. HS-derived fibroblasts (HSF

[18F]FPyQCP: A Robust FAP-Targeted PET Tracer Integrating Covalent C-18F Labeling and Radiometal Chelation for Enhanced In Vivo Performance.

Fibroblast activation protein (FAP) is a validated PET imaging target for the tumor microenvironment. Currently, most 18F-labeled FAP PET agents use Al-F chemistry, with Al18F-FAPI-74 leading clinically, whereas C-18F tracers perform less effectively. Here, we developed (4-quinolinoyl)glycyl-2-cyanopyrrolidine (QCP)-based inhibitors incorporating a 6-fluoronicotinamide prosthetic group for stable C-18F labeling. Chelator incorporation improved hydrophilicity and enabled structurally matched radiometal- and 18F-labeled analogs from a common scaffold. The compounds exhibited high FAP inhibitory

Development of Glycosylated and Dimeric FAP-Targeted Radiotracers for Preclinical Evaluation in FAP-Positive Glioblastoma Models.

Fibroblast activation protein (FAP) is an important target for tumor diagnosis and therapy. In this study, two FAP-targeted radioligands, the monomeric [68Ga]Ga-DOTA-T1-FAPI and the dimeric [68Ga]Ga-DOTA-T1-(FAPI)2, were designed by introducing a glucose linker into FAPI-04 and its dimer, and their FAP-targeting capability was further evaluated. In vitro studies demonstrated that [68Ga]Ga-DOTA-T1-(FAPI)2 exhibited specific uptake in FAP-positive U87 MG cells. Small animal positron emission tomography and computed tomography (PET/CT) imaging and biodistribution studies showed that the dimeric t

Development of CAR Exosomes Targeting FAP for the Treatment of Intrauterine Adhesion.

Engineered extracellular vesicles (EVs) have emerged as promising cell-free platforms for immunomodulation and tissue repair. In this study, we generated EVs derived from chimeric antigen receptor (CAR) T cells targeting fibroblast activation protein (FAP) and investigated their biological and therapeutic functions. These FAP-CAR EVs effectively inhibited intrauterine fibrosis, promoted endometrial regeneration, and increased pregnancy rates in a mouse model of intrauterine adhesion. Importantly, the exosome-based therapy did not affect embryonic development or trigger systemic inflammation, i

FAP expression as a marker of malignant transformation enabling in vivo characterization in peripheral nerve sheath tumors: a multimodal and translational study.

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive sarcomas and a major cause of mortality in neurofibromatosis type 1 (NF-1). Distinguishing MPNSTs from benign neurofibromas remains challenging. We investigated fibroblast activation protein alpha (FAP) as a malignancy biomarker and theranostic target in peripheral nerve sheath tumors. Therefore, we integrated publicly available bulk transcriptomics, spatial transcriptomics, and single-cell RNA sequencing with immunohistochemistry (IHC) on independent archival samples. We further directly assessed clinical translatability using F…

CAR Macrophages Engineered In Vivo for Attenuating Myocardial Ischemia-Reperfusion Injury.

BACKGROUND: Myocardial ischemia-reperfusion (I/R) injury induces myocardial fibrosis that compromises cardiac function and electrical conduction, yet current clinical options remain inadequate. To address this unmet need, we explored macrophage-targeted lipid nanoparticles (LNPs) encapsulating FAP CAR (FAP [fibroblast activation protein]-targeted chimeric antigen receptor) mRNA for in vivo generation of FAP CAR macrophages and evaluated their therapeutic potential in reducing myocardial fibrosis and improving cardiac function after myocardial I/R injury. METHODS: We formulated 1,2-dioleoyl-sn

[Fibroblast activation protein inhibitor PET for monitoring fibrotic diseases].

Fibroblast activation protein inhibitor (FAPI) PET was initially developed for cancer diagnostics; however, studies suggest that FAPI PET is a promising modality in various fibrotic diseases, including interstitial lung disease, fibrotic myocardial diseases, and rheumatic diseases such as rheumatoid arthritis, IgG4-related disease, and systemic scleroderma. There are limited studies on the subject, but this review finds that FAPI PET can accurately and specifically visualise active fibrosis and holds potential for monitoring treatment response and differentiation between inflammation and fibro

Dipeptidyl peptidase-IV in synovial fluid and in synovial fluid mononuclear cells of patients with rheumatoid arthritis.

BACKGROUND: Dipeptidyl peptidase-IV (DPP-IV) enzymatic activity controls biological halftime of multiple local mediators. Its deregulation is associated with pathogenesis of several autoimmune diseases, including rheumatoid arthritis (RA). Although DPP-IV is the canonical representative of the group, a number of other proteins have been shown to have similar enzymatic activity. This study was aimed to identify the molecular source of DPP-IV activity in synovial fluid (SF) and fluid mononuclear cells (FMNC) in patients with RA and osteoarthritis (OA). In addition, the association of DPP-IV and

Cancer-associated fibroblast activation predicts progression, metastasis, and prognosis of cutaneous squamous cell carcinoma.

Cutaneous squamous cell carcinoma (cSCC) is the most common metastatic skin cancer and the metastatic disease is associated with poor prognosis. Cancer-associated fibroblasts (CAFs) promote progression of cancer, but their role in cSCC is largely unknown. We examined the potential of CAF markers in the assessment of metastasis risk and prognosis of primary cSCC. We utilized multiplexed fluorescence immunohistochemistry for profiling CAF landscape in metastatic and non-metastatic primary human cSCCs, in metastases, and in premalignant epidermal lesions. Generation of electrophysiologically functional cardiomyocytes from mouse induced pluripotent stem cells.

Induced pluripotent stem (iPS) cells can efficiently differentiate into the three germ layers similar to those formed by differentiated embryonic stem (ES) cells. This provides a new source of cells in which to establish preclinical allogeneic transplantation models. Our iPS cells were generated from mouse embryonic fibroblasts (MEFs) transfected with the Yamanaka factors, the four transcription factors (Oct4, Sox2, Klf4 and c-Myc), without antibiotic selection or MEF feeders. After the formation of embryoid bodies (EBs), iPS cells spontaneously differentiated into Flk1-positive cardiac progen
