Influence of metal ions on flavonoid protection against asbestos-induced cell injury.

Influence of metal ions (Fe2+, Fe3+, Cu2+, Zn2+) on the protective effect of rutin, dihydroquercetin, and green tea epicatechins against in vitro asbestos-induced cell injury was studied. Metals have been found to increase the capacity of rutin and dihydroquercetin to protect peritoneal macrophages against chrysotile asbestos-induced injury. The data presented here show that this effect is due to the formation of flavonoid metal complexes, which turned out to be more effective radical scavengers than uncomplexed flavonoids. At the same time epicatechins and their metal complexes have similar a…

Dihydroflavonoids outperform their flavonoid counterparts in lipid-lowering efficacy based on pairwise analyses.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide, affecting approximately 25% of the adult population, yet no approved pharmacotherapy is currently available. Flavonoids have shown promise in improving MASLD, but the structural determinants underlying their lipid-lowering activities remain unclear. This study aimed to systematically investigate whether the presence or absence of the C-2,3 double bond affects the efficacy of flavonoids in ameliorating MASLD. Two pairs of C-2,3 positional isomeric flavonoids-eriodictyol (Compo…

Engineered flavonoid disrupts mitochondrial AIF/CHCHD4 complex for targeted cancer therapy.

Cancer metabolism depends on multifaceted mechanisms, including bidirectional inter-organelle communication between mitochondria and the nucleus, facilitating cellular adaptation at the transcriptomic, proteomic, and metabolomic levels. The mitochondrial protein complex composed of apoptosis-inducing factor (AIF) and coiled-coil-helix-coiled-coil-helix domain-containing protein 4 (CHCHD4) is essential for this mitochondrio-nuclear communication. The AIF/CHCHD4 complex mediates the mitochondrial import of cysteine-enriched nuclear gene-encoded proteins, thereby adapting the mitochondrial proteo

Possible applicability of flavonoid hesperetin for the treatment of dental biofilm-forming Candida glabrata.

BACKGROUND: Biofilm formation is a key virulence factor for Candida glabrata because it forms an extracellular matrix that prevents antifungal passage, which resists antifungal agents and causes treatment failure. To overcome this, the present study aimed to investigate the therapeutic potential of phytocompounds as an alternative choice in eliminating dental biofilm-forming C. glabrata. METHOD: The antifungal potential of various phytocompounds against C. glabrata was evaluated through zone of inhibition (ZOI) and minimum inhibitory concentration (MIC) studies. The antibiofilm potential of p

Flavonoids enhance tomato plant resistance to whitefly by interfering with the expression of a salivary effector.

Plants can deploy chemical defenses that poison herbivorous insects or deter their feeding; however, insects can counter by secreting effect … high-flavonoid-producing tomato to B. tabaci could mainly be attributed to the inhibitory effect of flavonoids on the expression of BtE3. Artificial diet feeding assays suggested that quercetin and rutin inhibited the expression of BtE3 gene. Combining with exogenous stem applications and CRISPR/Cas9-generated S. lycopersicum flavonol synthase mutants, we fu …

Antioxidant flavonoid baicalein as a dual purpose radiomodifier with promising bench to bedside potential.

Despite the potential of drug-radiotherapy combinations, a limited number of oncology drugs are approved by FDA for use along with radiotherapy. Currently, there is no agent available in the clinic that can exhibit dual radiomodifying behavior. Despite availability of sophisticated, high-precision techniques in radiotherapy for accurate targeting of tumors, normal tissue toxicity impedes its use in curative settings. Combining radiotherapy with an agent that can protect normal tissue and augment tumor killing can improve therapy outcomes. Other than combining drugs for therapeutic radiation ex
