Powdery mildew fungi are major obligate biotrophic plant pathogens, that cause widespread disease in agricultural and natural ecosystems worldwide, but a comparative structural view of their secretomes across multiple genera has remained limited. Here, we performed computational structure prediction and comparative analysis of 7545 secretome candidates from 26 isolates representing five genera (Blumeria, Erysiphe, Golovinomyces, Parauncinula, and Podosphaera) using AlphaFold2-based structure prediction, structural annotation against CATH, SCOPe, and PDB, Foldseek-based clustering and network a

The emergence of multi-azole resistant Candida albicans strains poses a major therapeutic challenge, largely due to their biofilm formation and hyphal development. In this study, the antifungal and antivirulence properties of 18 dibenzofuran derivatives were evaluated against a multi-azole resistant C. albicans strain. Dibenzofuran-4‑boronic acid (DBFBA) and dibenzofuran-2-carboxaldehyde (DBFC) emerged as the most active compounds, with DBFBA exhibiting the MIC of 30 μg/mL. Both compounds inhibited 94-98% biofilm at 10 μg/mL, suppressed hyphal morphogenesis and impaired cell aggregation at sub

Sixty novel 5-(trifluoromethyl)-1,2,4-oxadiazole derivatives were designed by scaffold hopping from the diamide lead 6n and synthesized to examine ether, thioether, and sulfoxide linkers. Greenhouse assays against Phakopsora pachyrhizi identified FM-12818 (B5) as a leading thioether derivative, with an EC50 of 0.45 mg·L-1, comparable to flufenoxadiazam (0.49 mg·L-1). FM-12818 also provided 80% control of Puccinia triticina at 0.391 mg·L-1 under the tested conditions. Docking to a human HDAC4 surrogate and DFT calculations suggested a putative zinc-binding orientation and linker-dependent elect

In this study, a series of novel hydrazide-containing flavonol derivatives was designed, synthesized, and evaluated for antifungal activity. In the in vitro antifungal assay, most of the target compounds exhibited potent antifungal activity against seven tested phytopathogenic fungi. In particular, compound C32 showed the best antifungal activity against Rhizoctonia solani (EC50 = 0.170 μg/mL), outperforming carbendazim (EC50 = 0.360 μg/mL) and boscalid (EC50 = 1.36 μg/mL). Compound C24 exhibited excellent antifungal activity against Valsa mali, Botrytis cinerea, and Alternaria alternata with

Invasive aspergillosis is one of the most serious clinical invasive fungal infections, resulting in a high case fatality rate among immunocompromised patients. The disease is caused by saprophytic molds in the genus Aspergillus, including Aspergillus fumigatus, the most significant pathogenic species. The fungal cell wall, an essential structure mainly composed of glucan, chitin, galactomannan, and galactosaminogalactan, represents an important target for the development of antifungal drugs. UDP (uridine diphosphate)-glucose pyrophosphorylase (UGP) is a central enzyme in the metabolism of carb

Pythium insidiosum has emerged as a challenging cause of keratitis in recent decades, due to its unique microbiological and clinical features, often is mistaken for fungal keratitis. Anti-fungal therapies have proven ineffective due to the organism's distinct cellular structure. Based on in vitro and in vivo models there has been a shift towards the use of antibacterial agents like linezolid and azithromycin, which have shown promising results. This concise review provides an update on the recommended diagnostic techniques and management strategies to assist clinicians in navigating this compl

Within the past few decades, the incidence and complexity of human fungal infections have increased, and therefore, the need for safer and more efficient, broad-spectrum antifungal agents is high. In the study described here, we characterized the new tetrazole-based drug candidate VT-1598 as an inhibitor of sterol 14α-demethylase (CYP51B) from the filamentous fungus Aspergillus fumigatus VT-1598 displayed a high affinity of binding to the enzyme in solution (dissociation constant, 13 ± 1 nM) and in the reconstituted enzymatic reaction was revealed to have an inhibitory potency stronger than th

Other than repaired fractures, osteoarthritis, and periosteal reaction, the vertebrate fossil record has limited evidence of non-osseous diseases. This difficulty in paleontological diagnoses stems from (1) the inability to conduct medical testing, (2) soft-tissue pathologic structures are less likely to be preserved, and (3) many osseous lesions are not diagnostically specific. However, here reported for the first time is an avian-style respiratory disorder in a non-avian dinosaur. This sauropod presents irregular bony pathologic structures stemming from the pneumatic features in the cervical

Fungal metabolites represent an underutilized resource in the development of novel anticancer drugs. This review will focus on the promising fungal nephrotoxin orellanine, found in mushrooms including Cortinarius orellanus (Fools webcap). Emphasis will be placed on its historical significance, structural features, and associated toxicomechanics. Chromatographic methods for analysis of the compound and its metabolites, its synthesis, and chemotherapeutic potential are also discussed. Although orellanine's exceptional selectivity for proximal tubular cells is well documented, the mechanics of it

Aspergillus fumigatus is a deadly agent of human fungal disease where virulence heterogeneity is thought to be at least partially structured by genetic variation between strains. While population genomic analyses based on reference genome alignments offer valuable insights into how gene variants are distributed across populations, these approaches fail to capture intraspecific variation in genes absent from the reference genome. Pan-genomic analyses based on de novo assemblies offer a promising alternative to reference-based genomics with the potential to address the full genetic repertoire of

The opportunistic fungal pathogen Candida albicans forms biofilms with viscoelastic properties and rheological response to flow that can significantly impact its biology and life cycle. The rheological phenotypes of fungal biofilms, however, have been neither quantified extensively nor genetically dissected. We have developed methods to quantify the rheology of fungal biofilms, and, here, we analyzed a panel of C. albicans deletion mutants impaired in cell wall structure or extracellular matrix (ECM) production for rheological phenotypes. Increased elastic moduli, indicative of higher viscoela

Candida auris is an emerging multidrug-resistant fungal pathogen for which neither vaccines nor rapid diagnostics are available. Defined fungal glycan epitopes offer a chemically precise route toward both immunization and detection. Here, we show that a synthetic β-mannan tetrasaccharide, β-(1,2)Man-α-(1,2)Man-α-(1,2)Man-α-(1,2)Man, functions as a lead epitope against C. auris. Conjugation of this glycan to the carrier protein CRM197 afforded a glycoconjugate that elicited epitope-selective immunoglobulin M (IgM) and immunoglobulin G (IgG) responses and reduced fungal burden in kidneys and spl

The skin microbiome at lesion sites in patients with atopic dermatitis (AD) is characterized by dysbiosis. Although the administration of dupilumab, an IL-4Rα inhibitor, improves dysbiosis in the bacterial microbiome, information regarding the fungal microbiome remains limited. This study administered dupilumab to 30 patients with moderate-to-severe AD and analyzed changes in both fungal and bacterial skin microbiomes over a 12-week period. Malassezia restricta and M. globosa dominated the fungal microbiome, whereas non-Malassezia yeast species increased in abundance, leading to greater microb

The World Health Organization (WHO) recognizes Candida albicans and Cryptococcus neoformans as the critical priority fungal pathogens for which therapeutic solutions are needed. Azole-based antifungal agents, including triazoles, diazoles, and thiazoles, are widely used in the treatments for fungal infections. In light of past successes in the transformation of antibacterial kanamycin into antifungal derivatives via chemical modifications, a new library of kanamycin-azole hybrids was synthesized and tested against a panel of azole-resistant and susceptible Candida and Cryptococcus strains. Fungal natural products (NPs) comprise a vast number of bioactive molecules with diverse activities, and among them are many important drugs. However, the yields of fungal NPs from native producers are usually low, and total synthesis of structurally complex NPs is challenging. As such, downstream derivatization and optimization of lead fungal NPs can be impeded by the high cost of obtaining sufficient starting material. In recent years, reconstitution of NP biosynthetic pathways in heterologous hosts has become an attractive alternative approach to produce complex NPs. Here, we present an eff

Synergistic saccharification involving bacterial cellulosome and fungal cellulases holds significant promise for industrial applications; however, this integrated approach remains largely under-investigated. This study dissects the synergy from structural characterization, adsorption analysis, crystallinity evolution, and hydrolysis performance evaluation. The cellulosome exhibited superior enhancement of substrate accessibility after enzymatic hydrolysis of sugarcane bagasse compared with fungal cellulases. The distinct changes in crystallinity observed during the hydrolysis of microcrystalli

Infections caused by fungal species via their existence as biofilms on medical devices can cause organ damage via candidiasis and candidemia. Different Candida species like Candida albicans can pose a serious threat by resisting host's immune system and by developing drug resistance against existing antimycotic agents. Therefore, targeting of fungal membranes can be used as an alternative strategy to combat the fungal infections. Here, we present screening of different amphiphiles based on cholic acid against different Candida strains as these amphiphiles can act as potent membrane-targeting a

Virulence mechanisms of the pathogenic fungus Aspergillus fumigatus are multifactorial and depend on the immune state of the host, but little is known about the fungal mechanism that develops during the process of lung invasion. In this study, microarray technology was combined with a histopathology evaluation of infected lungs so that the invasion strategy followed by the fungus could be described. To achieve this, an intranasal mice infection was performed to extract daily fungal samples from the infected lungs over four days post-infection. The pathological study revealed a heavy fungal pro
