Detection of carbapenemases, AmpC and ESBL genes in Acinetobacter isolates from ICUs by DNA microarray.

The accumulation of multiple inherent and acquired resistance mechanisms in Acinetobacter spp. results in emergence of "pandrug resistant" strains which is one of the major concerns in healthcare sectors worldwide. Surveillance of the carbapenemase/ extended-spectrum β-lactamases (ESBLs) genes in A. baumannii by phenotypic methods is challenging especially in developing countries, like Pakistan. In this context, a novel microarray (CT 103XL Check-MDR) assay was used for simultaneous detection of genes encoding clinically important carbapenemases and ESBLs. The results were compared with the ph

Polymorphisms of renin--angiotensin system genes as a risk factor for high-altitude pulmonary oedema.

The genes of the renin--angiotensin system (RAS) play an important role in the regulation of pulmonary vascular tone. Although studies on individual genes polymorphisms have reported association with high-altitude pulmonary oedema (HAPE), studies on multiple genes or epistasis are lacking. We therefore investigated the association of the RAS polymorphisms with HAPE. In a case-control design, we screened 163 HAPE-resistant/controls (HAPE-r) and 160 HAPEpatients (HAPE-p) of Indian origin for eight polymorphisms of four RAS genes, ACE, AGT, AGTR1 and AGTR2. Significant difference in genotype and

Comparative genomics reveals antimicrobial resistance and population structure of chicken-associated Salmonella enterica serotype Typhimurium in the United States.

Salmonella enterica serotype Typhimurium is a leading cause of foodborne outbreaks linked to chickens in the United States. To investigate the genomic landscape of this population, 1048 chicken-associated S. Typhimurium genomes collected nationwide were analyzed. Overall, 84.5% of isolates carried at least one antimicrobial resistance (AMR) determinant, and 34.3% displayed multidrug resistance (MDR). Sulfonamide and tetracycline resistance were most prevalent (1.03 and 1.01 AMR determinant per isolate, respectively). AMR gene (ARG) co-occurrence analysis identified strong associations among te

A broad-spectrum phage controls multidrug-resistant Salmonella in liquid eggs.

Salmonella is a foodborne pathogen constantly threating public health. The widespread use of antibiotics and globalization of the food industry result in rapid growth of drug-resistance. Eggs contaminated by multidrug-resistant (MDR) Salmonella are one of the riskiest factors of salmonellosis, which are frequently associated with outbreaks worldwide. Thus, there are increasing needs for the development of new technologies in controlling MDR pathogens and for the confirmation of their practical efficiency in target food matrices. In this study, 43 Salmonella phages were isolated from environmen

Emergence of a globally dominant IncHI1 plasmid type associated with multiple drug resistant typhoid.

Typhoid fever, caused by Salmonella enterica serovar Typhi (S. Typhi), remains a serious global health concern. Since their emergence in the mid-1970s multi-drug resistant (MDR) S. Typhi now dominate drug sensitive equivalents in many regions. MDR in S. Typhi is almost exclusively conferred by self-transmissible IncHI1 plasmids carrying a suite of antimicrobial resistance genes. We identified over 300 single nucleotide polymorphisms (SNPs) within conserved regions of the IncHI1 plasmid, and genotyped both plasmid and chromosomal SNPs in over 450 S. Typhi dating back to 1958. Prior to 1995, a v

Exploring the Antibacterial Properties of a Newly Isolated Microviridae Phage Against Multidrug-Resistant Escherichia coli.

In response to the alarming rise in antimicrobial resistance, bacteriophages have re-emerged as a promising alternative to conventional antibiotic therapy. The main objective of this paper was to characterize a newly isolated phage (vB_SEC_3) in the context of its suitability for phage therapy against MDR E. coli, which is considered a priority pathogen. The phage was characterized at the morphological, genomic, and biological levels relevant to phage therapy. TEM analyses revealed a non-enveloped icosahedral capsid lacking tail structure. Phylogenetic and tANI analyses placed the phage within

Gram-positive ESKAPE pathogens in Germany: A comprehensive analysis of occurrence and resistance development in animal, food, and environmental sources.

Antimicrobial resistance (AMR) is a global problem affecting both humans and animals. Multidrug resistance (MDR) infections are predominantly caused by a group of bacteria known as ESKAPE pathogens, namely Enterococcus (E.) faecium, Staphylococcus (S.) aureus, Klebsiella (K.) pneumoniae, Acinetobacter (A.) baumannii, Pseudomonas (P.) aeruginosa, and Enterobacter species. They are among the top threats to public health worldwide as their acquisition of AMR genes has significantly diminished available treatment alternatives for severe infections, increased disease burden, and elevated mortality

Antimicrobial resistance and virulence potential of non-faecium and non-faecalis enterococci isolated from canine rectal swabs.

Animals can serve as reservoirs for multidrug-resistant (MDR) enterococci with potential pathogenicity. Close contact between dogs and their owners may facilitate the interspecies transmission of these bacterial agents, potentially impacting human health. While Enterococcus faecium and Enterococcus faecalis are the main species implicated in healthcare-associated infections (HAIs), non-faecium and non-faecalis (NFF) enterococci, though less prevalent, may also contribute to infections. This study aimed to assess the antimicrobial resistance phenotypes and genotypes and virulence potential of N

Whole-genome sequencing and bioinformatic tools powered by machine learning to identify antibiotic-resistant genes and virulence factors in Escherichia coli from sepsis.

Extended-spectrum β-lactamase-producing Escherichia coli poses a global public health threat. Here, we performed a hospital-based study that reinforced the necessity for rapid antimicrobial resistance (AMR) and virulence gene mapping of clinical E. coli isolates. Whole-genome sequencing of 18 sepsis-causing E. coli strains was performed to identify multidrug resistance (MDR) and virulence factor genes and to correlate these with antibiotic use in patients with sepsis. We identified various global and emerging MDR sequence types, utilizing a supervised machine learning approach to elucidate the
