CONTEXT: Polycystic ovary syndrome (PCOS) is a common endocrine disorder that is believed to have a genetic basis. However, no specific susceptibility gene or region has been conclusively identified.
OBJECTIVE: The objective of this study was to duplicate a previous study that localized a PCOS susceptibility region to chromosome 19p13.2 and to narrow the susceptibility region.
DESIGN: This study was designed to test for genetic linkage and association between PCOS and short tandem repeat polymorphisms in 367 families, by analysis of linkage and family-based association.
SETTING: The study w…

Transplantation of 5 x 10(5) DBA/2 (H-2(d)) bone marrow cells into irradiated B10 and 129-strain mice (both H-2(b)) resulted in graft failure in the first recipient strain and in graft take in the second. Transplantation of B10 (H-2(b)) cells into irradiated B10.BR and C3H mice (both H-2(k)) also resulted in failure in the congenic B10.BR recipients and take in the C3H mice. Resistance and susceptibility of B10 and 129-strain animals were specific for given H-2 alleles of donor cells. Transplantation of DBA/2 marrow into (B10 x 129)F(2) mice and of B10 marrow into (B10.BR x C3H)F(1) x C3H back

Vitiligo is a chronic stigmatizing disease, already known for millennia, which mainly affects melanocytes from epidermis basal layer, leading to the development of hypochromic and achromic patches. Its estimated prevalence is 0.5% worldwide. The involvement of genetic factors controlling susceptibility to vitiligo has been studied over the last decades, and results of previous studies present vitiligo as a complex, multifactorial and polygenic disease. In this context, a few genes, including DDR1, XBP1 and NLRP1 have been consistently and functionally associated with the disease. Notwithstandi

Otosclerosis is a common form of conductive hearing loss with a prevalence of 0.3-0.4% in white adults. It is characterized by labyrinthine endochondral sclerosis which may invade the stapedio-vestibular joint and interfere with free motion of the stapes. Both environmental factors and genetic causes have been implicated in the disease process; however, the pathogenesis of otosclerosis still remains poorly understood. To date, several loci have been mapped in families segregating autosomal dominant otosclerosis although no disease-causing mutations have been identified. In contrast, several as

Recently, a few genes have been reported to be causative in inflammatory diseases. Still, we are waiting for the vast majority to be discovered. New tools for genotyping and statistical analysis have been developed and emphasis has been put on study design. Coeliac disease (CD) is a disorder, where prolamins in dietary wheat gluten and related proteins from rye or barley are not tolerated. It is one of the most common chronic diseases in humans exceeding a population prevalence of 1%. In this article, we will summarise what is currently known about the genetics influencing CD with the emphasis

Chinese alligator, Alligator sinesis, is a critically endangered endemic species under legislative protection. Results of recent investigations revealed that the number of the alligator was continuously declining in the past 50 years and less than 150 individuals were surviving in the wild until 2000. In order to prevent the extinguishing of this species, the Reproductive Research Center of Alligator sinesis and the National Nature Reserve of Alligator sinesis were set up in early 1980s in Xuanzhou, Anhui Province. After 20 years of breeding efforts, the number of captive individuals has been

Classic genetic studies utilizing twin methods, adoption and cross-fostering data and pedigree analysis, have confirmed the hypothesis that the familial component in affective disease is biological, rather than social or environmental. Only recently, however, has it been possible to clarify the mode of inheritance of affective disorder or to identify specific DNA segments which are physically linked to genes conferring susceptibility to affective illness. At present, two susceptibility genes have been identified in individual families, and it is certain that additional vulnerability genes rema

Donor strains of Erwinia chrysanthemi ICPB EC16, a member of the soft-rot (pectolytic) section of the enterobacterial genus Erwinia, were obtained by chromosomal integration of an F'lac(+) plasmid originating from Escherichia coli. These stable donor strains, selected from an unstable F'lac(+) heterogenote by repeated platings of single Lac(+) colonies on lactose minimal agar, do not segregate (as does the parent F'lac(+) heterogenote) into Lac(-) or F(-) clones, in either the presence or absence of acridine orange. One representative donor strain (from the 12 that have been selected) has been

Autosomal dominant familial spastic paraplegias (AD-FSP) are a group of genetically heterogeneous diseases characterised by a progressive spasticity of the lower limbs. Three loci have already been identified by genetic linkage studies on chromosomes 2p, 14q and 15q. Here we present linkage data from a large German family displaying AD-FSP with anticipation which confirms the existence of the FSP2 locus on chromosome 2p. The recombination events observed in our family define the critical region for the FSP2 gene to be within a 4-cM interval, flanked by markers D2S400 and D2S367. Moreover, clin…

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), is an enduring public health problem globally, particularly in sub-Saharan Africa. Several studies have suggested a role for host genetic susceptibility in increased risk for TB but results across studies have been equivocal. As part of a household contact study of Mtb infection and disease in Kampala, Uganda, we have taken a unique approach to the study of genetic susceptibility to TB, by studying three phenotypes. First, we analyzed culture confirmed TB disease compared to latent Mtb infection (LTBI) or lack of Mtb infection. Sec

A study was conducted to identify the major candidate chromosome and to detect the region which included the candidate gene causing gutter-shaped root (GSR) in an inbred strain of mice. The candidate chromosomal analysis was performed on genetic crosses of C57L/J strain mice, which have GSR, and C57BL/6J strain mice, which have normal roots. Linkage analysis suggested that mouse chromosome 5 was one of the major candidates and therefore this chromosome was investigated in detail by individual genotyping of all backcross mice. The highest linkage was found at D5Mit161, and other high linkages w

The developing mouse brain exhibits sex-biased microRNA (miRNA) expression driven by complex genetic, regulatory, and hormonal mechanisms. This study explored six hypotheses contributing to sex-biased miRNA expression in the E15.5 mouse brain: Y-chromosome linkage, escape from X to inactivation, regulation by sex-biased transcription factors, co-regulation of sex-biased miRNAs with their host genes, interaction of biological sex and miRNA processing, and transcriptional regulation by the estrogen receptor (ER) pathway. Evidence supports all of the mechanisms tested, except Y-linkage, as contri

PURPOSE: Hypospadias, when the urethra opens on the underside of the penis, is a common malformation that occurs in about 1/300 boys. The etiology of hypospadias is unknown but several lines of evidence indicate that it is caused by the combined influence of genetic and environmental factors. To identify chromosomal loci involved in the pathogenesis of hypospadias we performed genome-wide linkage analysis.
MATERIALS AND METHODS: A total of 69 families with at least 2 members with hypospadias were genotyped with 360 microsatellite markers and analyzed with a nonparametric affected relative pai

Autosomal dominant optic atrophy (OPA, MIM 165500) is an eye disease characterised by variable optic atrophy and reduction in visual acuity. It has an insidious onset in the first decade of life and is clinically highly heterogeneous. It is associated with a centrocecal scotoma of varying size and density and an acquired blue-yellow dyschromatopsia. Recent studies of three large Danish pedigrees have mapped a gene for dominant optic atrophy (OPA1) to a 10 cM region on chromosome 3q, between markers D3S1314 and D3S1265 (3q28-qter). Genetic linkage analysis in five British pedigrees confirms map…

Forty families submitted to Genetic Analysis Workshop 8 are described. These are the families in the Seattle data set which have been typed for at least one genetic marker from the centromeric region of chromosome 21 or the q arm of chromosome 19. Thirteen of these families have average ages of onset below age 61 and are therefore considered to be early onset families. Seven of the families are of Volga German descent. Thirty-four of the families have autopsy documented Alzheimer's disease, including all 13 of the early onset pedigrees. The data set includes both the clinical and pedigree info

Myoclonus-dystonia (M-D) is an autosomal dominant disorder characterized by myoclonic and dystonic muscle contractions that are often responsive to alcohol. The dopamine D2 receptor gene (DRD2) on chromosome 11q has been implicated in one family with this syndrome, and linkage to a 28-cM region on 7q has been reported in another. We performed genetic studies, using eight additional families with M-D, to assess these two loci. No evidence for linkage was found for 11q markers. However, all eight of these families showed linkage to chromosome 7 markers, with a combined multipoint LOD score of 11

Autosomal dominant osteogenesis imperfecta (OI) is a heterogeneous group of disorders. Molecular haplotypes associated with the pro alpha 2(I) gene of human type I procollagen were used for genetic linkage studies in a group of 10 families with OI. The clinical phenotypes of the families studied were those of OI type I and OI type IV. Evidence for linkage was highly suggestive in the four families with OI type IV (Z = 3.91 for theta = 0). In contrast, little or no indication for linkage was found in the six families with OI type I (Z = .055 for theta = .415). Heterogeneity between the two grou…
