Effects of genetic risk for depression and household socioeconomic status on emotional behavior and brain development in early adolescence.

BACKGROUND: The World Health Organization ranks depression as the leading non-fatal contributor to the global burden of disease. Research shows that early intervention before clinical onset leads to better outcomes. To study depression at this pre-onset stage, risk can be evaluated through depressive prodromal behaviors and associated brain biomarkers. Both environmental and genetic factors independently contribute to depression vulnerability, yet little work has examined how they interact. We therefore examined whether the socioeconomic status (SES) predictor of family income-to-needs ratio (

Enhancing Type 1 Diabetes Polygenic Risk Prediction Through Neural Networks and Entropy-Derived Insights.

Type 1 diabetes (T1D) is an autoimmune disease with a strong genetic component (~70% heritability). Early identification of individuals at risk is crucial for early intervention or risk assessment. Although polygenic risk scores (PRS) have shown promise in risk assessment, most current approaches remain constrained by linear assumptions and limited generalizability. We aimed to develop a neural network-driven classifier using T1D-associated single nucleotide polymorphisms (SNPs). In addition, we explored the inclusion of an entropy-derived feature as a complementary variable, representing the

Genome-wide association studies (GWAS) in 2026: From gene hunting to biological discovery in endocrine and cardiometabolic diseases.

BACKGROUND: Genome-wide association studies (GWAS) emerged with expectations borrowed from Mendelian genetics, creating a gap between what the method can and cannot deliver. This gap has fuelled both unwarranted dismissal and unwarranted enthusiasm. For endocrine clinicians, the confusion is consequential because GWAS and clinical sequencing answer fundamentally different questions and serve different purposes. PURPOSE: This review defines what GWAS is designed to do, clarifies the state of the "missing heritability" debate in 2026, and evaluates the contribution of GWAS to endocrine and card

Evaluating the heterogeneous treatment effects of major depression polygenic risk scores on self-harm using causal forest.

BACKGROUND: Self-harm, defined as non-fatal self-inflicted harm regardless of suicidal intent, is a critical global health issue influenced by the interaction between genetic and environmental factors. AIMS: This study investigates the causal effect of major depression polygenic risk scores (MD PRS) on self-harm in individuals of European ancestry and explores how environmental and personal factors moderate this effect. METHODS: Using data from 91,778 UK Biobank participants, we compared the top 10% with the highest MD PRS (the genetically vulnerable group) against the remainder. The causal

Maternal and offspring genome-wide association study of C-reactive protein reveals limited polygenic association with gestational diabetes mellitus.

BACKGROUND: C-reactive protein (CRP) is a well-established biomarker of systemic inflammation. In pregnancy, several studies show association of elevated CRP with gestational diabetes mellitus (GDM). However, the genetic contributions of CRP levels during early pregnancy and their potential association with GDM remain largely understudied. The Nulliparous Pregnancy Outcomes Study: Monitoring Mothers-to-Be (nuMoM2b) provides a unique opportunity to investigate these questions due to its rich design, including early pregnancy biospecimens, genome-wide genotype data, and detailed clinical outcome

Investigating the shared genetic architecture between dry eye disease and multisite chronic pain.

BACKGROUND: The epidemiological link between dry eye disease (DED) and chronic pain is established, yet a shared genetic etiology remains uncertain. METHODS: Utilizing GWAS summary data, we applied linkage disequilibrium score regression (LDSC) to determine genetic correlation between DED and multisite chronic pain (MCP). We investigated SNP enrichment across tissues using stratified LD score regression (S-LDSC) and multimarker analysis of genome annotation (MAGMA). Common risk SNPs were identified via Multi-Trait Analysis of GWAS (MTAG) and Cross-Phenotype Association (CPASSOC), followed by

A novel method to disentangle tightly linked risk and resilience genes for brain disorders: application to Alzheimer's disease.

Genetic risk factors for neuropsychiatric disorders are well documented. However, some individuals with high genetic risk remain unaffected, and the mechanisms underlying such resilience remain poorly understood. The presence of protective resilience factors that mitigate risk could help explain the disconnect between predicted risk and reality, particularly when genetic contributions are substantial but incompletely understood. Identifying and studying resilience factors could improve our understanding of pathology, enhance risk prediction, and inform preventive measures or treatment strategi

Consensus meta-analysis of genome-wide association studies for Alzheimer's disease and related dementia.

To better characterize the genetic architecture underlying Alzheimer's disease (AD) and related dementia (ADRD), we performed a meta-analysis of European ancestry genome-wide association studies in 128,681 cases or proxy cases of ADRD and 849,833 (proxy) controls. We identified 91 genetic loci associated with ADRD risk, including 16 which are novel, and 56 which are specifically detected in clinically diagnosed AD cases. We also provide a list of 18 loci (15 novel) requiring further external validation. A polygenic score combining the effect of the ADRD loci except APOE was primarily associate

Localization of the gene for Cowden disease to chromosome 10q22-23.

Cowden disease (CD) (MIM 158350), or multiple hamartoma syndrome, is a rare autosomal dominant familial cancer syndrome with a high risk of breast cancer. Its clinical features include a wide array of abnormalities but the main characteristics are hamartomas of the skin, breast, thyroid, oral mucosa and intestinal epithelium. The pathognomonic hamartomatous features of CD include multiple smooth facial papules, acral keratosis and multiple oral papillomas. The pathological hallmark of the facial papules are multiple trichilemmomas. Expression of the disease is variable and penetrance of the de

Relationship between Polygenic Risk Score and the Hypnotics in Bipolar I Disorder.

OBJECTIVE: Bipolar disorder (BD) is marked by significant change in mood and energy levels with sleep disturbance a common feature, resulting in diminished quality of life and impaired daily functioning. This study assessed the association between BD-polygenic risk scores (PRS) and hypnotics in bipolar I disorder (BD-I) patients. METHODS: Large-sample data were collected from the genome-wide association study of a multicenter Bipolar Genomic Study, and 1,394 BD-I patients with available medication information were divided into two groups depending on whether they used hypnotics or not. The Di

A machine learning-derived polygenic risk score reveals that healthy lifestyle counteracts obesity-related mortality.

Obesity is influenced by genetic predisposition and lifestyle. The associations among genetic susceptibility to obesity, lifestyle, and all-cause mortality remain unexplored. Our goal is to develop and validate a machine learning model to assess the genetic risk of obesity and examine its association with lifestyle and all-cause mortality. We integrated genetic data from 482,700 UK Biobank participants and 8,607 Nanfang Hospital participants to create and validate a stacked machine learning model, which generates an obesity-related polygenic risk score (OPRS), to evaluate the relationships amo

Diabetic Phenotypes and Late-Life Dementia Risk: A Mechanism-specific Mendelian Randomization Study.

BACKGROUND: Mendelian Randomization (MR) studies have reported that type 2 diabetes (T2D) was not associated with Alzheimer disease (AD). We adopted a modified, mechanism-specific MR design to explore this surprising result. METHODS: Using inverse-variance weighted MR analysis, we evaluated the association between T2D and AD using data from 39 single nucleotide polymorphisms (SNPs) significantly associated with T2D in DIAbetes Genetics Replication And Meta-analysis (DIAGRAM) and the corresponding associations of each SNP with AD risk obtained from the International Genomics of Alzheimer's Pro

Frailty and genetic risk predict fracture after lung transplantation.

Fractures negatively impact quality of life and survival. We hypothesized that recipient frailty score and genetic profile measured before transplant would predict risk of fracture after lung transplant. We conducted a retrospective cohort study of bone mineral density (BMD) and fracture among lung transplant recipients at a single center. The association between predictors and outcomes were assessed by multivariable time-dependent Cox models or regression analysis. Among the 284 participants, osteoporosis and fracture were highly prevalent. Approximately 59% of participants had posttransplant

Shared Genetic Etiology between Alzheimer's Disease and Blood Levels of Specific Cytokines and Growth Factors.

Late-onset Alzheimer's disease (AD) has a significant genetic and immunological component, but the molecular mechanisms through which genetic and immunity-related risk factors and their interplay contribute to AD pathogenesis are unclear. Therefore, we screened for genetic sharing between AD and the blood levels of a set of cytokines and growth factors to elucidate how the polygenic architecture of AD affects immune marker profiles. For this, we retrieved summary statistics from Finnish genome-wide association studies of AD and 41 immune marker blood levels and assessed for shared genetic etio

Genetic overlap between Alzheimer's disease and blood lipid levels.

Late-onset Alzheimer's disease (AD) has a significant genetic component, but the molecular mechanisms through which genetic risk factors contribute to AD pathogenesis are unclear. We screened for genetic sharing between AD and the blood levels of 615 metabolites to elucidate how the polygenic architecture of AD affects metabolomic profiles. We retrieved summary statistics from genome-wide association studies of AD and the metabolite blood levels and assessed for shared genetic etiology, using a polygenic risk score-based approach. For the blood levels of 31 metabolites, all of which were lipid

Fine-mapping of retinal vascular complexity loci identifies Notch regulation as a shared mechanism with myocardial infarction outcomes.

There is increasing evidence that the complexity of the retinal vasculature measured as fractal dimension, Df, might offer earlier insights into the progression of coronary artery disease (CAD) before traditional biomarkers can be detected. This association could be partly explained by a common genetic basis; however, the genetic component of Df is poorly understood. We present a genome-wide association study (GWAS) of 38,000 individuals with white British ancestry from the UK Biobank aimed to comprehensively study the genetic component of Df and analyse its relationship with CAD. We replicate

The risk of venous thromboembolism in oral contraceptive users: the role of genetic factors-a prospective cohort study of 240,000 women in the UK Biobank.

BACKGROUND: More than 150 million women worldwide use oral contraceptives. Women with inherited thrombophilia and carriers of certain thrombophilia gene variants, such as factor V Leiden and the prothrombin, are at an increased risk for venous thromboembolism, especially when combined with oral contraceptive use. Venous thromboembolism is a complex disorder involving many genetic risk factors, and recently, polygenic risk scores have been proposed to capture a significant proportion of the genetic risk of venous thromboembolism. OBJECTIVE: The aim of this study was to estimate the risk for de

[Association of Rest-Activity Rhythm With the Risk of Rheumatoid Arthritis and Effect Modification by Genetic Susceptibility].

OBJECTIVE: To investigate the association between rest-activity rhythm (RAR) and the risks of rheumatoid arthritis (RA), and to evaluate whether genetic susceptibility modifies this relationship. METHODS: This prospective cohort study utilized data from the UK Biobank, including 88060 participants who did not have RA at baseline. RAR parameters (e.g., relative amplitude) were calculated using data obtained through wrist-worn accelerometers. The participants' genetic susceptibility to RA was assessed using a polygenic risk score. Cox proportional hazards models were employed to analyze the ass
