Examination of DNAm PhenoAge as an epigenetic biomarker for perioperative risk in adult spinal deformity surgeries.

Epigenetic changes, such as DNA methylation (DNAm), offer a measure of biological age distinct from chronological age. DNAm PhenoAge is one such biomarker that is more strongly related to morbidity, mortality, and physical function than chronological age. More accurate risk stratification methods are needed for ASD surgeries, where complications remain difficult to predict with an increasingly aged population. A multicenter ASD registry was queried. DNAm PhenoAge was calculated as per Levine et al. (6). Multivariable logistic regression examined the associations of DNAm PhenoAge and chronologi

Class I histone deacetylases and their inhibitors as targets to modulate cellular senescence in osteoarthritis.

Cellular senescence plays a significant role in age-related conditions like osteoarthritis (OA) and intervertebral disc degeneration, in part due to the accumulation of senescent cells (SCs) in musculoskeletal tissues. Identifying novel therapeutics that can clear SCs is crucial for improving musculoskeletal health in the elderly. The present study aimed to elucidate the changes in Class I histone deacetylases (HDACs) and their role during senescence. All Class I HDACs except HDAC1 were downregulated during senescence in the human TC28a2 immortalized human chondrocyte cell line. Knockdown expe

PCSK9 inhibition attenuates alcohol-induced cardiovascular dysfunction and links hepatic lipid accumulation to impaired myocardial contractile reserve.

Excessive alcohol consumption accelerates cardiovascular aging by promoting oxidative stress, inflammation, lipid dysregulation, fibrotic remodeling, and loss of ventricular-vascular reserve. PCSK9, a key regulator of cholesterol metabolism, has emerged as a mediator of age-related cardiovascular dysfunction and alcohol-associated liver and neurovascular injury. We investigated whether PCSK9 inhibition protects against alcohol-induced cardiovascular dysfunction and associated cardiac-hepatic injury in rats. Male Sprague-Dawley rats were assigned to pair-fed control or 35% ethanol liquid diet g

The PERK paradox in aging: how ER stress shapes senescence and neurodegeneration.

Endoplasmic reticulum (ER) stress and activation of the unfolded protein response (UPR) are now recognized as integral components of the proteostasis network that preserves cellular and tissue function across the lifespan. With aging, increasing oxidative load, metabolic imbalance, and Ca2⁺ dysregulation elevate the burden of misfolded proteins in the ER, leading to progressive UPR engagement. When ER stress is mild or transient, UPR signaling restores folding capacity, restrains translation, and enhances redox and degradative programs, thereby promoting cellular resilience. In contrast, persi

Association between atrial fibrillation and systemic inflammation with muscle mass and strength trajectories in old age.

The long-term impact of atrial fibrillation (AF) and systemic inflammation on muscle health and, hence, functional decline remains unclear. This study investigates the link between AF and longitudinal trajectories of muscle mass and strength, considering the role played by systemic inflammation. Data were obtained from 2048 participants (≥ 60 years) in the Swedish National Study on Aging and Care in Kungsholmen (SNAC-K), followed for 12 years. Muscle mass and strength were assessed using calf circumference, handgrip strength, and chair stand test. Inflammatory marker interleukin-6 (IL-6) and t

VCD-induced estropause fails to enhance responsiveness to 17α-estradiol in adult female mice.

17α-estradiol (17α-E2) extends median lifespan and improves metabolic homeostasis in male mice through estrogen receptor α (ERα)-dependent mechanisms, but female mice are largely unresponsive unless ovariectomized or subjected to chronic high-fat feeding. Whether the gradual hormonal transition of natural reproductive aging similarly unmasks female responsiveness to 17α-E2 remains unknown. We tested whether 4-vinylcyclohexene diepoxide (VCD)-induced depletion of the ovarian reserve would render female mice responsive to 17α-E2 by treating wild-type (WT) and ERα knockout (ERαKO) littermates wit

Normal spermatogenesis in older men is associated with compensatory transcriptome changes.

Male reproductive ageing is a complex process involving progressive and detrimental histological and physiological alterations to the testis and beyond. Age-related morbidities often confound reproductive function, making it difficult to disentangle systemic from reproductive male ageing. We have previously shown that healthy ageing is associated with full spermatogenesis, normal sperm production and hormonal secretion. However, the molecular mechanisms allowing the human testis to age without major loss of function remained elusive. In this study, we investigated the transcriptomic dynamics o

Cardiovascular health, as per life's essential 8, and unhealthy aging trajectories.

BACKGROUND: Cardiovascular health (CVH) is associated with individual age-related health outcomes, but its relation to unhealthy aging trajectories remains uncertain. We assessed whether CVH is associated with the baseline burden of unhealthy aging and its progression over time, and whether these associations differ by sex. METHODS: We analyzed 2,487 adults aged 65 years or older enrolled in the Seniors-ENRICA-2 cohort (Madrid, Spain). CVH was assessed at three timepoints over a 5.2-year follow-up using the American Heart Association's Life's Essential 8 (LE8) score (range: 0-100; higher valu

Age-adjusted neurofilament light chain cutoffs for diagnosing neurodegenerative dementia: a two-threshold machine learning approach.

Clinical implementation of neurofilament light chain (NfL), a biomarker of neurodegeneration, remains challenging due to absence of reliable cutoffs and influence of confounding factors, particularly age. We aimed to develop an age-adjusted, two-threshold classification framework to support clinical interpretation of NfL in the neurodegenerative dementia diagnosis. We retrospectively enrolled subjects with cognitive/behavioral disturbances and cognitively unimpaired controls (CTR). Participants underwent a baseline diagnostic workup, including at least a neuropsychological assessment, blood te

Recovery strategies during trip-like support surface perturbations while walking in older adults with mild cognitive impairment.

Older adults with mild cognitive impairment (OAwMCI) fall 2 × more than cognitively intact older adults (CIOA), possibly due to impaired reactive stepping. However, most studies in OAwMCI have focused on slip-like perturbations (backward loss of balance), rather than trip-like perturbations (forward loss of balance). This study examined reactive stepping strategies following a novel support surface perturbation during treadmill walking, and compared fall rate, margin of stability (MOS), step length, and trunk angle between OAwMCI and CIOA during unperturbed walking (i.e., pre-trip) and recover

Muscle Ageing and Sarcopenia Study (MASS) Lifecourse: a valuable resource for understanding skeletal muscle ageing.

Advances in our understanding of the biology of skeletal muscle ageing are being made at pace, with great potential for these findings to inform the identification of novel treatments for sarcopenia. However, translation of findings from animal models to humans has been hampered by limitations of existing human muscle biopsy studies. Devised to directly address this challenge, the Muscle Ageing and Sarcopenia Study (MASS) Lifecourse is a novel resource for the study of human muscle ageing. This deep-phenotyped observational study of 260 community-dwelling men and women aged 18 to 85 years livi

Amyloid and tau pathologies are drivers of white matter damage in aging and Alzheimer's disease.

White matter hyperintensities (WMHs) are increasingly recognized as neuroimaging biomarkers of cerebrovascular pathology in Alzheimer's disease (AD), yet their temporal relationship with amyloid and tau accumulation remains unclear. While previous studies suggest bidirectional associations between WMHs and AD pathology, regional associations between WMHs and AD pathology have yet to be examined. This study investigated the temporal and regional associations between PET measures of amyloid (Aβ) and tau pathology and WMH burden in older adults. Data from the Alzheimer's Disease Neuroimaging Init

Analysis of second-generation epigenetic clocks reveals further associations between disproportionate biological ageing and hippocampal volume.

Age acceleration, or the difference between biological and chronological age, is a research area of growing interest, particularly in the field of age-related and neurodegenerative disorders, including Alzheimer's disease (AD). First-generation epigenetic clocks that predict chronological age utilising DNA methylation data were the first to derive estimates of age acceleration, which have been associated with several age-related conditions. More recently, second-generation epigenetic clocks that are predictors of specific traits such as age-related health, disease or morbidity and mortality ph

The choice stepping reaction task: how prefrontal network topology reconfigures in aging and dementia.

The ability to initiate stepping promptly and accurately is critical for safe mobility in later life. Choice stepping reaction time (CSRT) is an independent predictor of recurrent falls in older adults and is often prolonged in dementia. Although the prefrontal cortex (PFC) is crucial for rapid response execution, existing studies primarily focus on regional activation, leaving the topological reorganization of prefrontal functional networks poorly understood. In this study, twenty-six healthy older adults (HOAs) and 25 older adults with mild-to-moderate dementia (MMD) completed four blocks ea

Plasma metabolomic and inflammatory profiles associated with low physical function in heart failure: an integrated cardiac-metabolic-muscular phenotype.

Dysregulated metabolomic profiles (dysmetabolism) have been observed in patients with heart failure (HF). In this study, we investigated metabolomic profiling differences in HF with reduced physical-functional capacity, hereafter termed reduced "muscle health" for brevity (HF-RM) compared to preserved muscle health (HF-PM) and healthy adults (NonHF). The HF-RM label denotes a screening-level functional phenotype consistent with EWGSOP2 "probable sarcopenia" and Fried physical-frailty constructs and is not intended to imply primary skeletal myopathy independent of cardiac limitation. Association of high-intensity evening light exposure with risk of incident age-related macular degeneration, cataract, and glaucoma: a prospective cohort study of 82,826 participants.

Excessive artificial light at night (ALAN) disrupts circadian rhythms and may accelerate ocular aging; however, personal objective dosimetry data linking evening light exposure to specific age-related eye diseases (AREDs) remain sparse. The objective of the study is to determine whether personal high-intensity light exposure during the evening transition window (20:00-23:30) is associated with the risk of incident age-related macular degeneration (AMD), cataracts, and glaucoma. This prospective cohort study utilized data from the UK Biobank. Participants (n = 82,826) were monitored for 7 days

Age-dependent changes in lipid droplet distribution and vascularization in naked mole rat vs. mouse hippocampus.

The accumulation of lipid droplets in microglia has been reported to increase in response to ageing and age-related diseases like Alzheimer's disease. The present study investigates changes in lipid droplet dynamics with increasing age in the mouse and the similarly sized long-lived rodent, the naked mole rat (Heterocephalus glaber). We show that the naked mole rat contains large amounts of microglial lipid droplets in the CA1 and CA3 region of the hippocampus with an age-dependent increase in the CA3 region. Neuronal lipid droplet size is unaffected by age in the naked mole rat. The mouse hip

Intestinal oxysterol remodeling underlies severe COVID-19 in an aged hamster model.

Age is a critical determinant of COVID-19 severity, yet the underlying metabolic mechanisms driving disease progression remain poorly understood. Using a well-characterized Syrian hamster model, we investigated how aging shapes intestinal and systemic sterol metabolism during SARS-CoV-2 infection. Young (7-10 weeks) and aged (12 months) hamsters were intranasally infected, and clinical outcomes were integrated with virological, histopathological, and lipidomic profiles across intestinal tissues, lungs, and serum. Despite comparable pulmonary pathology, aged hamsters exhibited significantly hig

Long-term outcomes of transcatheter mitral valve repair in patients with cancer: a systematic review and meta-analysis.

Surgical and transcatheter mitral valve interventions are the mainstay of treatment for mitral regurgitation (MR). Their impact in patients with cancer has recently gained attention in observational studies but remains poorly characterized. We performed a systematic review and meta-analysis to evaluate outcomes of mitral valve interventions, with particular focus on M-TEER, in patients with active or prior cancer. PubMed and Scopus were systematically searched according to the PRISMA 2020 Statement. This systematic review was prospectively registered in PROSPERO (CRD420261368392). Studies repo

Energetic pathway to lower urinary tract symptoms in older adults: associations with cardiorespiratory fitness and walking energetics in the study of muscle, mobility and aging.

Lower urinary tract symptoms (LUTS) are associated with older age and mobility limitations. We evaluated whether energetic determinants of mobility-maximum oxygen consumption (VO2peak), energetic cost of walking (ECw), and cost-capacity ratio (CCR) were associated with LUTS in community-dwelling older adults. We used baseline data from 629 participants aged ≥ 70 years in the Study of Muscle, Mobility and Aging. The 10-item LURN Symptom Index (LURN SI-10; range: 0-38, higher = worse) assessed LUTS in the past week. A treadmill cardiopulmonary exercise test using a symptom-limited modified Balke
