The clinical efficacy of granulocyte transfusions: studies on the oral cavity.

Patients with severe granulocytopenia of different origin received granulocyte transfusions. The migration and phagocytic ability of the transfused granulocytes in the oral cavity ("orogranulocytes") served as a parameter for measuring the viability of the granulocytes in vivo. There was no difference between granulocytes collected by continuous flow centrifugation and those collected by filtration leukapheresis. Under the influence of the transfusion treatment mucous membrane ulcers (MMU) subsided and healed. Where ulcers were present the number of orogranulocytes (OG) increased after granulo…

Effect of bone marrow suppression on granulocyte opsonin levels.

Levels of serum opsonin for neutrophilic granulocytes were measured in dogs made neutropenic by cyclophosphamide administration. Heat-labile opsonin became elevated within 24 hr following cyclophosphamide (P less than 0.005) and remained elevated over the 4-5 day period of observation (P less than 0.005). In contrast, heat stable opsonin was not significantly effected. Bone marrow suppression by X-ray and busulfan also caused serum opsonin levels to increase. Changes in the levels of IgG, C3, and total hemolytic complement during the course of bone marrow suppression did not correlate with the…

Felty's syndrome: granulocyte-bound immunoglobulin G and splenectomy.

A quantitative antiglobulin consumption technique was used to measure immunoglobulin G (IgG) present on human granulocytes. Granulocytes from 50 normal subjects had less than 20 X 10(-14) g IgG per cell. Patients with granulocytopenia due to bone-marrow failure, patients with IgG multiple myeloma, patients with splenomegaly, and patients with rheumatoid arthritis without granulocytopenia had granulocyte-bound IgG within the range of normal. Four patients with rheumatoid arthritis, splenomegaly, and severe granulocytopenia (Felty's syndrome) had granulocyte-bound IgG between 30 and 220 X 10(-14…

Granulocyte reactive antinuclear factors in juvenile rheumatoid arthritis.

The prevalence of mouse liver reactive antinuclear factors (L-ANF) and human granulocyte reactive antinuclear factors (G-ANF) was determined in sera from 34 age- and sex-matched pairs of patients with juvenile rheumatoid arthritis (JRA) and controls. Neither type of antinuclear factor was found in any of the control sera, whereas of the patient sera, 35% were L-ANF positive and 59% were G-ANF positive. 90% of the G-ANF positive sera were of the IgG class and 50% were able to fix complement. L-ANF was most frequent in female patients. Several types of ANF were associated with the polyarticular

[Granulocyte dysfunction. I. Inborn defects (author's transl)].

The insight in the function and dysfunction of granulocytes lately arouses more and more interest. This report summarises our present knowledge. In the first of two chapters the authors review the molecular basis of granulocyte function and the inborn defects of chemotaxis, opsonisation, phagocytosis and intracellular killing of bacteria and fungi.

[Editorial: Discrepancies in granulocyte survival time (author's transl)].

Important discrepancies between the renewal rate of mature granulocytes measured by in vitro labelling with DF 32P and with sodium chromate 51Cr are observed. The possible explanations are reviewed: errors made in the drawing of the experimental curve of the radioactivity disappearance; labelling of long-living lymphocytes with chromium; elution of 32P; toxicity of DFP for the tagged cells. If the renewal-rate measured by using 51Cr is the real one, as it seems to be, currently accepted models of granulopoiesis will need reappraisal.

2-Deoxy-D-glucose reshapes the sepsis-trained stressed granulocytes by comprehensive alterations of the transcriptional landscape.

The secondary infections following sepsis pose a significant clinical challenge due to hyperinflammatory organ injury, which is primarily driven by maladaptive trained immunity (TRIM) in stressed granulocytes. Glucose metabolic reprogramming serves as a crucial mechanism underlying TRIM and is increasingly acknowledged as a promising therapeutic target for preventing cytokine storms following reinfections. Here, we employed recovery mice subsequent to the cecal ligation and puncture (CLP) procedures to model reinfection following sepsis and confirm the beneficial effects of 2-Deoxy-D-glucose (…

Protocol for quantification of circulating immature granulocytes in patients with a chronic myeloid malignancy using flow cytometry.

Immature granulocytes (iGRANs) circulating in the peripheral blood of patients with a chronic myeloid malignancy, such as chronic myelomonocytic leukemia, are immunosuppressive cells that promote disease progression. Here, we present a multiparameter flow cytometry-based protocol to quantify iGRANs among peripheral blood mononucleated cells (PBMCs). We describe steps for collecting and labeling PBMCs. We describe the gating strategy to quantify iGRANs as a fraction of myeloid cells or as an absolute number of circulating cells, which supports patient stratification and monitoring. For complete…

Whole-blood model reveals granulocytes as key sites of dengue virus propagation, expanding understanding of disease pathogenesis.

Dengue virus (DENV) infection poses a significant global health threat, yet our understanding of its immunopathogenesis remains incomplete due to limitations of existing models. Here, we establish an in vitro whole-blood model using hirudin, an anticoagulant that preserves complement activity and cellular interactions, to study DENV infection. Our model reveals the susceptibility of all major leukocyte populations to DENV infection, with monocytes and granulocytes demonstrating high permissiveness and production of infectious virus progeny. Notably, granulocytes emerge as previously unrecogniz…

Granulocytes-Rich Thrombi in Cerebral Large Vessel Occlusion Are Associated with Increased Stiffness and Poorer Revascularization Outcomes.

We aim to identify a profile of intracranial thrombus resistant to recanalization by mechanical thrombectomy (MT) in acute stroke treatment. The first extracted clot of each MT was analyzed by flow cytometry obtaining the composition of the main leukocyte populations: granulocytes, monocytes, and lymphocytes. Demographics, reperfusion treatment, and grade of recanalization were registered. MT failure (MTF) was defined as final thrombolysis in cerebral infarction score IIa or lower and/or need of permanent intracranial stenting as a rescue therapy. To explore the relationship between stiffness …

Autosomal recessive hyposegmentation of granulocytes in Australian Shepherd Dogs indicates a role for LMBR1L in myeloid leukocytes.

Pelger-Huët anomaly (PHA) in humans is an autosomal dominant hematological phenotype without major clinical consequences. PHA involves a characteristic hyposegmentation of granulocytes (HG). Human PHA is caused by heterozygous loss of function variants in the LBR gene encoding lamin receptor B. Bi-allelic variants and complete deficiency of LBR cause the much more severe Greenberg skeletal dysplasia which is lethal in utero and characterized by massive skeletal malformation and gross fetal hydrops. HG phenotypes have also been described in domestic animals and homology to human PHA has been cl…

[Functional activity of blood neutrophilic granulocytes in patients with opisthorchiasis, depending on the severity of liver fibrosis].

AIM: To study the activity of neutrophilic granulocytes in patients with opisthorchiasis, depending on the severity of liver fibrosis. MATERIALS AND METHODS: A total of 74 patients with chronic opisthorchiasis (39 men and 35 women, average age 42.3 years) and 32 practically healthy patients (17 men and 15 women, average age 41.5 years) aged 24 to 60 years were examined. Diagnosis of opisthorchiasis was carried out by two methods: coprooscopy and identification of eggs or bodies of adult parasites in duodenal content. Liver fibrosis was determined by the method of elastometry according to the …
