Cortical functional connectivity changes in amnestic mild cognitive impairment.

OBJECTIVE: Amnestic mild cognitive impairment (aMCI) is characterized by episodic memory deficits and is defined as the prodromal phase of Alzheimer's disease. Therefore, it is critical to reveal the dysfunction in large-scale networks during the dementia phase of the disease. This study aimed to examine the resting-state functional connectivity changes between aMCI and healthy control groups. METHODS: MRI and clinical data from 25 individuals with aMCI and 25 healthy controls (HC) were used. Seed-to-seed functional connectivity analyses were performed between all histologically classified Br

Oxytocin Reduces Subjective Fear in Naturalistic Social Contexts via Enhancing Top-Down Middle Cingulate Amygdala Regulation and Brain-Wide Fear Representations.

Accumulating evidence from animal and human studies suggests a fear-regulating potential of the neuropeptide oxytocin (OT), yet the clinical translation into novel interventions for pathological fear requires a behavioral and neurofunctional characterization under close-to-real-life conditions. Here, we combined a naturalistic fMRI-design that elicited strong and immersive fear experience in social and non-social contexts with a preregistered between-subjects randomized double-blind placebo-controlled intranasal OT trial (24 IU, n  =  67 healthy men). OT selectively reduced subjective fear in

Investigating Neural Dynamics in Tinnitus Using Constrained Independent Component Analysis.

Background: Tinnitus is a neurological condition characterized by the perception of ringing or other phantom sounds in the absence of external auditory stimuli. It affects an estimated 10%-15% of adults worldwide and can significantly affect sleep and mood. Neuroimaging techniques, particularly functional Magnetic Resonance Imaging (fMRI), have been widely used to investigate the auditory system and brain networks in tinnitus. Resting-state fMRI (rs-fMRI), a noninvasive approach, is particularly effective in examining spontaneous neural activity and functional connectivity (FC) across brain re

Diminished reward circuit response underlies pain avoidance learning deficits in problem drinkers.

Individuals engaging in problem drinking show impaired proactive pain avoidance. As successful pain avoidance is intrinsically rewarding, this impairment suggests reward deficiency, as hypothesized for those with alcohol and substance misuse. Nevertheless, how reward circuit dysfunctions impact avoidance learning and contribute to drinking behavior remains poorly understood. Here, we combined functional imaging and a probabilistic learning go/nogo task to examine the neural processes underlying proactive pain avoidance learning in 103 adult drinkers. We hypothesized that greater drinking sever

Blunted anterior midcingulate response to reward in opioid users is normalized by prefrontal transcranial magnetic stimulation.

Abnormalities in goal-directed behavior, mediated by mesocorticolimbic reward system, contribute to worse clinical outcomes including higher risk of treatment dropout and drug relapse in opioid users (OU). Despite efforts to counteract such neural alterations, brain-based interventions for this disorder remain ineffective. In this sham-controlled randomized study, we report the initial results on the efficacy of transcranial magnetic stimulus (TMS) in normalizing reward functioning in this population. During a reward-based choice task, we applied robot-assisted 10-Hz TMS to the prefrontal cort

[Electroacupuncture alleviates facial nerve injury-induced anxiety-like behavior by raising the excitability of glutamatergic neurons in the anterior cingulate cortex of mice].

OBJECTIVES: To investigate the effect and mechanism of electroacupuncture (EA) at "Siguan" acupoints (bilateral Hegu [LI4] and bilateral Taichong [LR3]) in ameliorating facial nerve injury-induced anxiety-like behavior in mice. METHODS: Thirty male C57BL/6J mice were randomly divided into control, model (unilateral facial nerve injury), and EA groups, with 10 mice in each group. The facial nerve injury model was established by lightly clamping the isolated facial nerve for about 30 s, twice altogether after operation. The mice of the EA group were subjected to EA stimulation (2 Hz, 0.5 mA) of

A Meta-Analysis of Magnetic Resonance Spectroscopy Studies on Glutamatergic Neurometabolite Levels in Major Depressive Disorder.

Glutamatergic neurometabolite dysregulation has recently garnered attention in the pathophysiology of major depressive disorder (MDD). However, studies have reported heterogeneous results for changes in neurometabolite levels across brain regions and whether these changes are related to antidepressant intervention. Herein, we performed a meta-analysis to investigate consistent findings by searching PubMed, Embase, MEDLINE, PsycINFO, and SinoMed from the start date of these databases to May 2023 (PROSPERO#CRD42023405205). Among the 2529 publications screened, 55 studies were included in the met

The relationship between structural properties of frontal cortical regions and response inhibition in 6-14-year-old children.

Development of attentional skills and inhibitory control rely on maturational changes in the brain across childhood and youth. However, both brain anatomy and different components of attention and inhibition show notable individual variation. Research on ADHD and inhibitory training and control have shown that variations in the thickness and surface area of particularly inferior cortical structures are associated with attentional control. However, the intricacies of how the development of inhibitory control is associated with the anatomical variations beyond the general age- and gender-depende

Altered activation of the dorsal anterior cingulate cortex during oddball performance in individuals at risk for Alzheimer's disease.

INTRODUCTION: The neural mechanisms underlying neurodegenerative disorders in the elderly remain elusive, despite extensive neuroimaging research in recent decades. Amnestic type mild cognitive impairment (aMCI) and late-life major depressive disorder (MDD) are two such conditions characterized by intersecting cognitive and affective symptomatology, and they are at a higher risk for Alzheimer's disease. MATERIALS AND METHODS: This study analyzed the neural underpinnings of cognitive and depressive symptoms in a cohort comprising 12 aMCI subjects, 24 late-life MDD patients, and 26 healthy cont

Localized alterations in cortical thickness and sulcal depth of the cingulo-opercular network in relation to lower reading fluency skills in children with dyslexia.

The traditional models of reading development describe how language processing and word decoding contribute to reading comprehension and how impairments in word decoding, a defining feature of dyslexia, affect reading comprehension outcomes. However, these models do not include word and sentence reading (contextual reading) fluency, both of which engage executive functions, with notably decreased performance in children with dyslexia. In the current study, we compared cortical thickness and sulcal depth (CT/SD) in the cingulo-opercular (CO) executive functions brain network in children with dy

Altered whole brain functional activity in patients with fibromyalgia.

OBJECTIVES: Fibromyalgia (FM) is a chronic pain disorder that takes a severe physical and psychological toll on patients and severely reduces their quality of life. In recent years, an increasing number of studies have used functional magnetic resonance imaging (fMRI) to investigate its pathogenesis. However, a recent summary analysis of functional connectivity in patients with FM is lacking. METHODS: We searched bibliographic databases, including PubMed, Web of Science (from inception until September 1st, 2022). Two separate researchers assessed the bias and quality of the studies. In order

Developmental Differences in a Hippocampal-Cingulate Pathway Involved in Learned Safety Following Interpersonal Trauma Exposure.

OBJECTIVE: Nearly 65% of youth experience trauma, and up to one-third of youth with trauma exposure face profound mental health sequelae. There remains a need to elucidate factors that contribute to psychopathology following trauma exposure, and to optimize interventions for youth who do not benefit sufficiently from existing treatments. Here, we probe safety signal learning (SSL), which is a mechanism of fear reduction that leverages learned safety to inhibit fear in the presence of threat-associated stimuli and has been shown to attenuate fear via a hippocampal-cingulate--specifically, a dor

Brain circuits for maternal sensitivity and pain involving anterior cingulate cortex among mothers receiving buprenorphine treatment for opioid use disorder.

Opioid-induced deficits in maternal behaviors are well-characterized in rodent models. Amid the current epidemic of opioid use disorder (OUD), prevalence among pregnant women has risen sharply. Yet, the roles of buprenorphine replacement treatment for OUD (BT/OUD) in the brain functions of postpartum mothers are unclear. Using functional magnetic resonance imaging (fMRI), we have developed an evolutionarily conserved maternal behavior neurocircuit (MBN) model to study human maternal care versus defensive/aggressive behaviors critical to mother-child bonding. The anterior cingulate gyrus (ACC)

Shared and Distinct Neurobiological Bases of Bipolar Disorder and Attention-Deficit/Hyperactivity Disorder in Children and Adolescents: A Comparative Meta-Analysis of Structural Abnormalities.

OBJECTIVE: Pediatric bipolar disorder (PBD) and attention-deficit/hyperactivity disorder (ADHD) frequently co-occur and share dysfunctions in affective and cognitive domains. As the neural substrates underlying their overlapping and dissociable symptomatology have not been well delineated, a meta-analysis of whole-brain voxel-based morphometry studies in PBD and ADHD was conducted. METHOD: A systematic literature search was performed in PubMed, Web of Science, and Embase. The seed-based d mapping toolbox was used to identify altered clusters of PBD or ADHD and obtain their conjunctive and com

Impaired self-awareness of cognitive deficits in Parkinson's disease relates to cingulate cortex dysfunction.

BACKGROUND: Impaired self-awareness of cognitive deficits (ISAcog) has rarely been investigated in Parkinson's disease (PD). ISAcog is associated with poorer long-term outcome in other diseases. This study examines ISAcog in PD with and without mild cognitive impairment (PD-MCI), compared to healthy controls, and its clinical-behavioral and neuroimaging correlates. METHODS: We examined 63 PD patients and 30 age- and education-matched healthy controls. Cognitive state was examined following the Movement Disorder Society Level II criteria. ISAcog was determined by subtracting z-scores (based on

Greater reading gain following intervention is associated with low magnetic resonance spectroscopy derived concentrations in the anterior cingulate cortex in children with dyslexia.

BACKGROUND/OBJECTIVE: The "neural noise" hypothesis suggests that individuals with dyslexia have high glutamate concentrations associated with their reading challenges. Different reading intervention programs have showed low GLX (a combined measure for glutamine and glutamate obtained with in vivo magnetic resonance spectroscopy) in association with reading improvement. Several studies demonstrated improved reading and increased activation in the anterior cingulate cortex following an-executive-function (EF)-based reading intervention. The goals of the current study are two-fold: 1) to determi
