Proteolytic activities in Trypanosoma rangeli and stercorarian trypanosomes: taxonomic implications.

We analysed the proteinase profiles in nine Trypanosoma (Herpetosoma) rangeli strains from distinct geographical regions, three T. (Schizotrypanum) cruzi reference strains and in T. (Herpetosoma) lewisi, T. (Herpetosoma) musculi and T. (Megatrypanum) conorhini samples by detection of enzyme activity on SDS-PAGE containing co-polymerized gelatin as substrate. Three different profiles of cysteine proteinase activity were detected in T. rangeli within a similar range of molecular mass. T. lewisi and T. musculi displayed a similar pattern of proteolysis, quite distinct from the ones detected in T.

Effects of Corcyra cephalonica Egg Consumption on Population Fitness and Reproduction of the Whitefly Predator Serangium japonicum (Coleoptera: Coccinellidae).

Ladybird beetle, Serangium japonicum (Coleoptera: Coccinellidae), is an important predatory natural enemy of whiteflies, and its mass rearing is crucial for biological control. This study evaluated the suitability of Corcyra cephalonica (Lepidoptera: Pyralidae) eggs as an alternative diet for adult S. japonicum by directly comparing it to the natural prey, Bemisia tabaci (Hemiptera: Aleyrodidae) nymphs. Results showed that, compared to a B. tabaci diet, feeding on C. cephalonica eggs supported normal development and significantly extended the lifespan of adult S. japonicum, without compromisin…

Resistance of sorghum hybrids to sorghum aphid.

The aphid Melanaphis sorghi (Theobald) (Hemiptera: Aphididae), which infest the sorghum crop, has been an economically important pest which have been causing severe damage to sorghum crops in Brazil since 2019. These species have been observed mainly at the end of vegetative stage and beginning of reproductive stage of plants. Their high reproductive rate on sorghum raises concerns about these pests. Therefore, the present study aimed to estimate the life expectancy and fertility tables of Melanaphis sorghi fed on 15 hybrids of grain sorghum, in order to know the resistance characteristics of …

Functional and metabolic validation of CYP419A1v2 in mediating resistance of Laodelphax striatellus (Fallén) to triflumezopyrim.

Triflumezopyrim (TFM), a new type of mesoionic insecticide with good insecticidal activity, showed great value in field control of Laodelphax striatellus. However, the molecular mechanisms underlying L. striatellus resistance to TFM remain poorly understood. In the current study, a 46.67-fold resistant strain (RS31) was selected in the laboratory from a susceptible laboratory stain (SS) after 31 generations of resistance screening. Compared with the SS, the activities of cytochrome P450 monooxygenases (P450) and carboxylesterases (CarE) were significantly elevated. In addition, CYP303A1, CYP41

Functional analysis of lncRNAs regulating the TOR signaling pathway and reproduction in the white-backed planthopper, Sogatella furcifera.

BACKGROUND: long non-coding RNAs (lncRNAs), as key endogenous regulators, have been reported to be involved in regulating insect reproduction in recent years. However, little is known about their role in the reproductive regulation of insects. RESULTS: In this study, we predicted four lncRNAs of potential cis-regulated TOR signaling pathway genes using bioinformatics analysis. They were named MSTRG.5802.1, MSTRG.5803.1, MSTRG.9854.1, and MSTRG.26502.2, and we explored their biological functions in Sogatella furcifera. RT-qPCR revealed that four lncRNAs were expressed in all life stages, with

Subversion of the salicylic acid signaling pathway by the bipartite begomoviral protein BV1 promotes virus infection and vector preference to virus-infected plants.

The outbreak of vector-borne plant viruses entails both efficient in planta virus infection and productive vector-mediated transmission. Yet, the factors that concurrently regulate these two key viral traits remain understudied. Here we examine the role of viral proteins and the salicylic acid (SA) signaling pathway in modulating virus infection and vector preference to virus-infected plants. Infection of plants by the bipartite begomovirus, Sri Lankan cassava mosaic virus (SLCMV), dramatically induces the accumulation of SA, a positive regulator of antiviral defense. As countermeasures, SLCMV

Investigation into the docking of odorant binding proteins from tea green leafhoppers with host plant volatile components.

Insects possess an intricate and efficient olfactory system that plays a critical role in navigating their environment, facilitating host location, defensive maneuvers, and reproductive behaviors. In the present study, 12 biological samples were successfully sequenced, yielding a total of 79.92 G data. This led to the assembly of 46,291 Unigenes, with a cumulative length of 47,102,395 base pairs. These Unigenes were subsequently annotated across seven functional databases, including NR (22,104), Swissprot (16,203), KEGG (7,652), KOG (14,597), eggNOG (19,129), GO (14,852), and Pfam (15,267). Identification and analysis of microRNAs responsible for brown planthopper resistance in BPH14 and BPH15 pyramiding rice.

BACKGROUND: Rice farming faces a significant challenge from the brown planthopper (BPH), a destructive pest that threatens crop yields. Developing BPH-resistant rice varieties is critical for ensuring food security. The pyramiding of BPH resistance genes, BPH14 and BPH15, has proven effective in providing protection in elite rice strains. MicroRNAs (miRNAs) play a pivotal role in plant defense by fine-tuning resistance responses through the modulation of genes involved in various signaling pathways and metabolic processes. B1415, the pyramiding line containing BPH14 and BPH15, exhibits stronge

Comparative transcriptomic and metabolomics analysis of ovary in Nilaparvata lugens after trehalase inhibition.

The fecundity of Nilaparvata lugens (brown planthopper) is influenced by trehalase (TRE). To investigate the mechanism by which trehalose affects the reproduction of N. lugens, we conducted a comparative transcriptomic and metabolomic analysis of the ovaries of N. lugens following injection with dsTREs and validamycin (a TRE inhibitor). The results revealed that 844 differentially expressed genes (DEGs) were identified between the dsGFP and dsTREs injection groups, with 317 up-regulated genes and 527 down-regulated genes. Additionally, 1451 DEGs were identified between the water and validamyci

wStri spread dynamics in Nilaparvata lugens via discrete mathematical models.

Wolbachia, an intracellular bacterium, is well-known for inducing cytoplasmic incompatibility, which has become a promising and environmentally sustainable strategy for controlling pest populations. The strain wStri, specifically identified in Nilaparvata lugens (brown planthopper), has shown potential for such biocontrol applications. In this study, we develop a comprehensive discrete mathematical model to analyze the dynamics of wStri spread in a mixed population of wStri-infected, wLug-infected, and uninfected Nilaparvata lugens under both constant and periodically varying environmental con

MicroRNA response in insect salivary glands to plant virus infection.

UNLABELLED: Most arboviruses rely on insect vectors for transmission, with salivary glands serving as a critical gateway for viral spread to new hosts. MicroRNAs (miRNAs) are key regulators of gene expression, yet their roles in salivary glands during virus infection remain poorly understood. Using the small brown planthopper (SBPH)-rice stripe virus (RSV) system, we sequenced small RNAs from salivary glands of nonviruliferous and viruliferous insects and identified 5,909 known miRNAs from 201 families. Of these, 1,143 miRNAs were differentially expressed upon RSV infection, including 1,090 up

The down-regulation of salivary protein gene expression by etofenprox partially contributed to reducing the risk of increased fecundity in the brown planthopper.

Etofenprox is a pyrethroid insecticide that acts on the nervous system of insects. Due to its low toxicity to aquatic animals, it is permitted for use in controlling insect pests in rice fields. The brown planthopper (BPH), Nilaparvata lugens, a significant piercing-sucking pest feeding on rice exclusively, secretes various salivary components when feeding. Salivary proteins are essential for BPH feeding, but their response to etofenprox is not well understood. The application of etofenprox down-regulated the expression of 21 salivary protein genes, among which 9 genes (NlShpa, Salivap 3, CA,

Beet curly top virus affects vector biology: the first transcriptome analysis of the beet leafhopper.

Curly top disease, caused by beet curly top virus (BCTV), is among the most serious viral diseases affecting sugar beets in western USA. The virus is exclusively transmitted by the beet leafhopper (BLH, Circulifer tenellus) in a circulative and non-propagative manner. Despite the growing knowledge on virus-vector interactions, our understanding of the molecular interactions between BCTV and BLH is hampered by limited information regarding the virus impact on the vector and the lack of genomic and transcriptomic resources for BLH. This study unveils the significant impact of BCTV on both the pe

Enabling biocontained plant virus transmission studies through establishment of an axenic whitefly (Bemisia tabaci) colony on plant tissue culture.

Whiteflies (Bemisia tabaci) and the diseases they transmit are a major detriment to crop yields and a significant contributor to world hunger. The highly evolved interactions of host plant, phloem-feeding insect vector with endosymbionts and persistently transmitted virus represent a tremendous challenge for interdisciplinary study. Presented here is the establishment of a colony of axenic whiteflies on tissue-cultured plants. Efficient colony establishment was achieved by a surface sterilization of eggs laid on axenic phototrophically tissue-cultured plants. The transfer of emerging whiteflie

Flavonoid production in tomato mediates both direct and indirect plant defences against whiteflies in tritrophic interactions.

BACKGROUND: The role of plant flavonoids in direct defences against chewing and sap-sucking herbivorous insects has been extensively characterized. However, little is known about flavonoid-mediated tritrophic interactions between plants, herbivorous insects and natural enemies. In this study, we investigated how flavonoids modulate plant-insect interactions in a tritrophic system involving near-isogenic lines (NILs) of cultivated tomato (Solanum lycopersicum) with high (line NIL-purple hypocotyl [PH]) and low (line NIL-green hypocotyl [GH]) flavonoid levels, with a generalist herbivore whitefl

The extraction and identification of active components of the sex pheromones of Asian citrus psyllid, Diaphorina citri.

In this study, we extracted and identified the active components of the Asian citrus psyllid, Diaphorina citri sex pheromones to provide a basis for further development of sex attractants. Under laboratory conditions, mating activity in D. citri started 3 d after emergence, which peaked at 6-7 d, and mating activity had no obvious peak during the observed period 7:00-21:00 h. Additionally, D. citri males were attracted to the emanations from conspecific females, especially to the n-hexane extracts of the pheromone. A total of 17 compounds were identified from the n-hexane extracts of female an

Portiera Gets Wild: Genome Instability Provides Insights into the Evolution of Both Whiteflies and Their Endosymbionts.

Whiteflies (Hemiptera: Sternorrhyncha: Aleyrodidae) are a superfamily of small phloem-feeding insects. They rely on their primary endosymbionts "Candidatus Portiera aleyrodidarum" to produce essential amino acids not present in their diet. Portiera has been codiverging with whiteflies since their origin and therefore reflects its host's evolutionary history. Like in most primary endosymbionts, the genome of Portiera stays stable across the Aleyrodidae superfamily after millions of years of codivergence. However, Portiera of the whitefly Bemisia tabaci has lost the ancestral genome order, refle…

Brassinosteroids mediate susceptibility to brown planthopper by integrating with the salicylic acid and jasmonic acid pathways in rice.

Improved knowledge of the interactions between plants and insects will facilitate better insect control in crops. Brassinosteroids (BRs) play a vital role in plant growth, developmental processes, and responses to pathogen infection, but the role of BRs in interactions between plants and insects remain largely unknown. In this study, we characterized a negative role of BRs in rice defense against brown planthopper (BPH, Nilaparvata lugens) and examined its underlying mechanisms. We found that BPH infestation suppressed the BR pathway while successively activating the salicylic acid (SA) and ja

Qualitative and Quantitative Differences in Herbivore-Induced Plant Volatile Blends from Tomato Plants Infested by Either Tuta absoluta or Bemisia tabaci.

Plants release a variety of volatile organic compounds that play multiple roles in the interactions with other plants and animals. Natural enemies of plant-feeding insects use these volatiles as cues to find their prey or host. Here, we report differences between the volatile blends of tomato plants infested with the whitefly Bemisia tabaci or the tomato borer Tuta absoluta. We compared the volatile emission of: (1) clean tomato plants; (2) tomato plants infested with T. absoluta larvae; and (3) tomato plants infested with B. tabaci adults, nymphs, and eggs.
