Dysregulated microvascular reactivity in hippocampus and cortex in CVN Alzheimer's disease mouse model.

Microvascular reactivity in acute cortical and hippocampal brain slices and hippocampal synaptic- evoked cerebral blood flow (CBF) in vivo were analyzed in a mouse model of Alzheimer's disease (AD, CVN). Microvessels underwent initial vasoconstriction (2 µM noradrenaline) then treatment with either 0.5 mM glutamate or 100 µM NMDA. In acute brain slices from young mice (<20 weeks) the glutamate and NMDA treatment led to dilation of capillaries in cortex and hippocampus, but not in aged CVN mice (>30 weeks, with AD pathology). Furthermore, 1 mM adenosine restored pre-constricted capillaries to c…

Hippocampus and basal forebrain degeneration differentially impact cognition in Lewy body spectrum disorders.

Cognitive impairment is a major contributor to the burden in Parkinson's disease and dementia with Lewy bodies, both of which make up the Lewy body disease spectrum, with dementia affecting up to 80% of patients over the course of the disease. Macroatrophy and microstructural neurodegenerative alterations are typically assessed separately in MRI, although neuropathologically they represent the same mechanism-the loss of functional tissue. To gain a deeper insight into the differential impact of neurodegeneration in the basal forebrain and hippocampus on cognition, we have developed a combined …

Functional Connectivity Between Human Hippocampus and Orbitofrontal Cortex Supports Inference-Based Decision Making.

Internal representations of the relationships between entities or events, usually known as cognitive maps, enable individuals to make inferences from past experiences to guide decisions. Previous studies found both the hippocampus (HIP) and orbitofrontal cortex (OFC) are involved in this process. However, it is still unclear how the interaction between them responds to the inference-based decision. We recruited healthy adult subjects to perform an inference task during the functional magnetic resonance imaging (fMRI) scanning. Subjects were required to use their learned knowledge of hierarchic…

Flavokawain B ameliorates depressive-like behaviors by inhibiting neddylation in the ventral hippocampus.

Depression is a widespread and serious mental illness. Recent evidence suggests that post-translational modifications, particularly neddylat … lation-related protein expression (NEDD8, NAE1, UBA3, UBC12) was measured in the dorsal and ventral hippocampus (DH, VH), prelimbic cortex (PrL), and nucleus accumbens (NAc). Results showed that FKB reversed the activation of neddylation anxiety- and depressive-like behaviors in both LPS- and CUMS-treated mice. The LPS and CUMS experimental paradigms only imp …

PD-1 deficiency disrupts in vivo neural activity in mouse Hippocampus and cortex.

Programmed cell death 1 (PD-1), encoded by the Pdcd1 gene, was identified as a target in cancer immunotherapy but may result in the overactivation of T cell function and emotional changes such as anxiety. The dynamic changes in neuronal activity related to the anxious status caused by Pdcd1-/- remain unclear. In this study, we addressed these physiological issues by simultaneously recording neuronal activity (spikes) and local field potentials (LFPs) in the medial prefrontal cortex (mPFC) and hippocampal CA3 region using in vivo multi-channel electrodes. Our results demonstrate that PD-1 defic

Effects of stress-related neuromodulators on amygdala and hippocampus resting state functional connectivity.

BACKGROUND: The human stress response is characterized by increases in neuromodulators, including norepinephrine (NE) and cortisol. Both neuromodulators can enter the brain and affect neurofunctional responses. Two brain areas associated with stress are the amygdala and the hippocampus. The precise influence of NE and cortisol on the amygdala and hippocampal resting state functional connectivity (RSFC) is poorly understood. AIMS: To investigate the influence of NE and cortisol on the amygdala and hippocampal RSFC. METHODS: We recruited 165 participants who received 10 mg yohimbine and/or 10 …

Exercise, memory, and the hippocampus: Uncovering modifiable lifestyle reserve factors in refractory epilepsy.

Physical exercise is an emerging target for improving cognition in aging and neurological disease. Due to the beneficial impact of exercise on hippocampal health and the vulnerability of the hippocampus in medication-resistant temporal lobe epilepsy (TLE), exercise could present a promising intervention in TLE. We investigated whether exercise engagement is associated with verbal memory function and hippocampal integrity in 29 young to middle-aged adults with refractory TLE and 21 demographically matched controls. Participants completed a self-reported questionnaire of weekly exercise, three t…

Hippocampus under Pressure: Molecular Mechanisms of Development of Cognitive Impairments in SHR Rats.

Data from clinical trials and animal experiments demonstrate relationship between chronic hypertension and development of cognitive impairments. Here, we review structural and biochemical alterations in the hippocampus of SHR rats with genetic hypertension, which are used as a model of essential hypertension and vascular dementia. In addition to hypertension, dysfunction of the hypothalamic-pituitary-adrenal system observed in SHR rats already at an early age may be a key factor of changes in the hippocampus at the structural and molecular levels. Global changes at the body level, such as hype…

L-DOPA ameliorates hippocampus-based mitochondria respiratory dysfunction caused by GCI/R injury.

Mitochondrial dysmorphology/dysfunction follow global cerebral ischemia-reperfusion (GCI/R) injury, leading to neuronal death. Our previous researches demonstrated that Levodopa (L-DOPA) improves learning and memory impairment in GCI/R rats by increasing synaptic plasticity of hippocampal neurons. This study investigates if L-DOPA, used in Parkinson's disease treatment, alleviates GCI/R-induced cell death by enhancing mitochondrial quality. Metabolomics and transcriptomic results showed that GCI/R damage affected the Tricarboxylic acid (TCA) cycle in the hippocampus. The results of this study …

Quinic acid contributes to neurogenesis: Targeting Notch pathway a key player in hippocampus.

Coordinated proliferation and differentiation of neural stem cells (NSCs) results in continuous neurogenesis. The present study provides novel insights into the Notch intracellular signaling in neuronal cell proliferation, maintenance, migration, and differentiation regulated by naturally based Quinic acid (QA) in primary hippocampal cell culture. Further, this study might help in the discovery and development of lead molecules that can overcome the challenges in the treatment of neurodegenerative diseases. The growth supporting effect of QA was studied using Alamar Blue assay. The migratory p

Elevated Astrocytic NFAT5 of the hippocampus increases epilepsy susceptibility in hypoxic-ischemic mice.

OBJECTIVE: Hypoxic-ischemic brain damage (HIBD) is a leading cause of neonatal mortality, resulting in brain injury and persistent seizures that can last into the late neonatal period and beyond. Effective treatments and interventions for infants affected by hypoxia-ischemia remain lacking. Clinical investigations have indicated an elevation of nuclear factor of activated T cells 5 (NFAT5) in whole blood from umbilical cords of severely affected HIBD infants with epilepsy. Experimental research has demonstrated that NFAT5 has ambivalent effects on neuroprotection and neurologic damage. However

The hippocampus associated GABAergic neural network impairment in early-stage of Alzheimer's disease.

Alzheimer's disease (AD) is the commonest neurodegenerative disease with slow progression. Pieces of evidence suggest that the GABAergic system is impaired in the early stage of AD, leading to hippocampal neuron over-activity and further leading to memory and cognitive impairment in patients with AD. However, the precise impairment mechanism of the GABAergic system on the pathogenesis of AD is still unclear. The impairment of neural networks associated with the GABAergic system is tightly associated with AD. Therefore, we describe the roles played by hippocampus-related GABAergic circuits and …

Hippocampus shapes cortical sensory output and novelty coding through a direct feedback circuit.

To extract behaviorally relevant information from our surroundings, our brains constantly integrate and compare incoming sensory information with those stored as memories. Cortico-hippocampal interactions could mediate such interplay between sensory processing and memory recall1-4 but this remains to be demonstrated. Recent work parsing entorhinal cortex-to-hippocampus circuitry show its role in episodic memory formation5-7 and spatial navigation8. However, the organization and function of the hippocampus-to-cortex back-projection circuit remains uncharted. We combined circuit mapping, physiol…

What does the hippocampus do during working-memory tasks? A cognitive-neuropsychological perspective.

In this commentary, we highlight the role of the hippocampus as a binding device that may explain its recruitment during associative working-memory paradigms. Furthermore, we argue that both functional neuroimaging research, as presented in Slotnick (this issue), and carefully designed lesion studies in patients with selective bilateral hippocampal damage are crucial for advancing our understanding of the neural structures and processing involved in human memory in general and disentangling the role of the hippocampus proper and other medial temporal lobe structures in working-memory function …
