Lactylation: the metabolic-immune hub in autoimmune diseases.

In recent years, lactate modification, as an emerging post-translational modification mechanism, has attracted increasing attention for its role in the regulation of the immune system. Autoimmune diseases are a category of complex disorders characterized by abnormal attacks of the immune system on self-tissues. The limitations of traditional treatments have made the search for new therapeutic targets a hot topic in research. Lactate modification plays a significant role in the development and progression of autoimmune diseases. It can modulate the activation and function of T cells, B cells, m

Multi-trait and cross-population genome-wide association studies across autoimmune and allergic diseases identify shared and distinct genetic component.

OBJECTIVES: Autoimmune and allergic diseases are outcomes of the dysregulation of the immune system. Our study aimed to elucidate differences or shared components in genetic backgrounds between autoimmune and allergic diseases. METHODS: We estimated genetic correlation and performed multi-trait and cross-population genome-wide association study (GWAS) meta-analysis of six immune-related diseases: rheumatoid arthritis, Graves' disease, type 1 diabetes for autoimmune diseases and asthma, atopic dermatitis and pollinosis for allergic diseases. By integrating large-scale biobank resources (Bioban

Renal thrombotic microangiopathy in patients with systemic lupus erythematosus and the antiphospholipid syndrome.

Current studies indicate that a thrombotic microangiopathy (TMA) identifies patients with systemic lupus erythematosus (SLE) who are at high risk of progressing to end-stage renal disease. We have observed two patients with SLE and one patient with a primary antiphospholipid syndrome (APS) who developed acute renal insufficiency with thrombocytopenia. Renal biopsies showed a TMA characterized by thrombi or by cellular and mucoid intimal hyperplasia of small arteries and arterioles. No arterial or arteriolar immune-complex deposits were detected by immunofluorescent or electron microscopy. Biop

Optineurin modulates mTORC2/AKT/STAT3 signaling to control MHC class II expression and adaptive immunity against HSV-1.

Herpes simplex virus 1 (HSV-1) infection contributes to immunopathogenic diseases and lacks an effective vaccine. Improving antigen presentation is key to better vaccine strategies and more robust immune responses. Here, we show that optineurin (OPTN), an autophagy receptor traditionally involved in protein recycling, unexpectedly stabilizes RICTOR (mechanistic target of rapamycin complex 2 [mTORC2]), a crucial step in enhancing MHC class II surface expression in dendritic cells. OPTN regulates the AKT/mTOR/signal transducer and activator of transcription 3 (STAT3) pathway, with the AKT2 isofo

Mapping the Altered Landscape of TCR Repertoire in Autoimmune Diseases.

Autoimmune diseases arise from the breakdown of immune tolerance through complex interactions between genetic predisposition, environmental exposures, and adaptive immune responses. High-throughput T-cell receptor (TCR) repertoire sequencing has transformed our ability to characterize these responses, providing unprecedented insights into clonal dynamics, antigen-driven selection, and immune history. In this review, we summarize the major alterations of TCR repertoires reported across autoimmune diseases, including changes in diversity, clonal expansion, repertoire architecture, and tissue dis

[The role of cytokines in Chlamydia-induced inflammation].

Chlamydia is an obligate intracellular pathogen that causes a wide range of diseases in humans and animals. Chlamydia infection often causes inflammatory response of the body, which seriously affects the health of the host. Cytokines, as key molecules of immune regulation, play an important role in Chlamydia-induced inflammation. Proinflammatory cytokines, such as tumor necrosis factor α (TNF-α), interleukin 1β (IL-1β), IL-6 and interferon γ (IFN-γ), are rapidly activated in the early stage of Chlamydia-induced infection, participating in the recruitment of immune cells to the site of infectio

[Research progress of the ULBP-NKG2D axis in autoimmune diseases].

The activating receptor natural killer group 2 member D (NKG2D) and its ligands, the UL16-binding protein (ULBP), play pivotal roles in autoimmune diseases, characterized by multidimensional regulatory features. This review employs a three-dimensional framework of "ligand supply-ligand fate-receptor regulation" to analyze the research progress of the ULBP-NKG2D axis in autoimmune diseases, including systemic lupus erythematosus, rheumatoid arthritis, type 1 diabetes, multiple sclerosis, and Crohn disease. In systemic lupus erythematosus, the axis involves both peripheral immune suppression (dr

GSDMD-mediated pyroptosis: molecular mechanisms, diseases and therapeutic targets.

Pyroptosis is a regulated form of inflammatory cell death in which Gasdermin D (GSDMD) plays a central role as the key effector molecule. GSDMD-mediated pyroptosis is characterized by complex biological features and considerable heterogeneity in its expression, mechanisms, and functional outcomes across various tissues, cell types, and pathological microenvironments. This heterogeneity is particularly pronounced in inflammation-related diseases and tumors. In the context of inflammatory diseases, GSDMD expression is typically upregulated, and its activation in macrophages, neutrophils, T cells

Classification of urticaria and the reactive inflammatory vascular dermatoses.

The common feature of all the conditions discussed in this article is that they are inflammatory vascular dermatoses that may occur as reactive processes in association with other diseases. The histopathologic characteristics of the lesions range from mild, perivascular dermal infiltration with inflammatory cells and vasodilation to various degrees of vessel damage (endothelial swelling to fibrinoid necrosis). The vessel damage is reflected in varying degrees of secondary changes including extravasation of edema fluid, extravasation of erythrocytes, epidermal necrosis, separation of the dermal

Immune signature of gene expression pattern shared by autism spectrum disorder and Huntington's disease.

Autism spectrum disorder (ASD) and Huntington's disease (HD) are complex neurological conditions with unclear causes and limited treatments, affecting individuals, families, and society. Despite ASD and HD representing two opposing stages of neuronal development and degeneration, they share similar clinical-pathological features in motor function. In this study, we leveraged transcriptomic data from the prefrontal cortex available in public databases to identify shared transcriptional characteristics of ASD and HD. Differential expression analysis revealed that the majority of differentially e

A Clinical Approach for the Use of VIP Axis in Inflammatory and Autoimmune Diseases.

The neuroendocrine and immune systems are coordinated to maintain the homeostasis of the organism, generating bidirectional communication through shared mediators and receptors. Vasoactive intestinal peptide (VIP) is the paradigm of an endogenous neuropeptide produced by neurons and endocrine and immune cells, involved in the control of both innate and adaptive immune responses. Exogenous administration of VIP exerts therapeutic effects in models of autoimmune/inflammatory diseases mediated by G-protein-coupled receptors (VPAC1 and VPAC2). Currently, there are no curative therapies for inflamm

New insight of complement system in the process of vascular calcification.

The complement system defences against pathogenic microbes and modulates immune homeostasis by interacting with the innate and adaptive immune systems. Dysregulation, impairment or inadvertent activation of complement system contributes to the pathogenesis of some autoimmune diseases and cardiovascular diseases (CVD). Vascular calcification is the pivotal pathological basis of CVD, and contributes to the high morbidity and mortality of CVD. Increasing evidences indicate that the complement system plays a key role in chronic kidney diseases, atherosclerosis, diabetes mellitus and aging-related

Inflammasomes in Common Immune-Related Skin Diseases.

The inflammasome is an important protein complex that cleaves the proinflammatory cytokines pro-IL-1β and pro-IL-18 into their active forms. Owing to its critical role in eliciting innate immune responses, IL-1β has been suggested to contribute to various skin diseases, including psoriasis, vitiligo, systemic lupus erythematosus (SLE), and atopic dermatitis (AD). Recently, several types of activators and inhibitors of different inflammasomes, as well as inflammasome-related genes and genetic susceptibility loci, have been identified in these immune-related common skin diseases. In particular,

Killing of human dermal capillary endothelial cells by antibody-dependent cellular cytotoxicity.

Damage to the microvasculature occurs commonly in autoimmune diseases affecting the skin. There has been little investigation of immune mechanisms other than immune complex deposition as a major cause of microvascular damage. We have investigated the potential of antibody-dependent cellular cytotoxicity (ADCC) to cause vascular destruction in an in vitro model consisting of human dermal microvascular endothelial cells to which were added antibody and leukocytes. Severe damage was induced by antibody and lymphocytes. Monocytes or neutrophils were not able to mediate cytotoxicity although activa

Emerging viral infections in immunocompromised patients: A great challenge to better define the role of immune response.

The immune response to invading pathogens is characterized by the rapid establishment of a complex network of cellular interactions and soluble signals. The correct balancing of activating and regulating pathways and tissue-homing signals determines its effectiveness and persistence over time. Emerging viral pathogens have always represented a great challenge to the immune system and an often uncontrolled/imbalanced immune response has been described (e.g. cytokine storm, immune paralysis), contributing to the severity of the disease. Several immune biomarkers and cell subsets have been identi

Interplay between bacteria and pulmonary immune system in respiratory infections.

The local immune system of the lungs is essential for the defense against pathogens, and respiratory bacterial infections remain a major cause of mortality in patients with lower respiratory tract diseases. However, the precise mechanisms underlying immune-pathogen interactions and the modulatory roles of commensal bacteria remain incompletely understood. This review discusses the mechanisms of immune recognition and inflammatory responses during bacterial respiratory bacterial infections, highlighting the importance of pattern recognition receptors, including toll-like receptors, nucleotide o
