NLR inflammasome pathways: key targets for pathogenesis and therapy of metabolic diseases.

The NOD-like receptor (NLR) inflammasome system is an evolutionarily conserved intracellular surveillance network that responds to pathogen-associated molecular patterns (PAMPs), damage-associated molecular patterns (DAMPs), and metabolism-associated molecular patterns (MAMPs). Beyond the well-characterized NLRP3 inflammasome, accumulating evidence suggests that NLRP1, NLRP6, NLRC5, NLRP12, and NLRP2 are also implicated in metabolic disorders, including obesity, type 2 diabetes mellitus (T2DM), atherosclerosis, and metabolic dysfunction-associated steatotic liver disease (MASLD). This review s

Bafetinib enhances anti-tumor immunity by activating the NLRP3 inflammasome in macrophage.

Insufficient lymphocyte infiltration in the tumor microenvironment is a major cause of resistance to immunotherapy. Previous studies have indicated that NLRP3 inflammasome activation is crucial for lymphocyte infiltration and activation, but safe, effective, and specific NLRP3 inflammasome agonists are still lacking. Here, we identified a clinical phase II drug, bafetinib, that specifically activates the NLRP3 inflammasome through high-throughput drug screening. Bafetinib directly targets and binds to the potassium channel KCNK6/TWIK2, blocking its degradation via the chaperone-mediated autoph

HDAC6 regulates BACE1 stability and NLRP3 inflammasome activation in Alzheimer's disease.

Alzheimer's disease (AD) is marked by amyloid-β (Aβ) accumulation, tau pathology, and neuroinflammation. The β-site APP cleaving enzyme 1 (BACE1) is a key driver of Aβ production, while the NLRP3 inflammasome mediates microglial inflammatory responses. Histone deacetylase 6 (HDAC6), a cytoplasmic deacetylase, is upregulated in AD, yet its role in disease mechanisms remains unclear. Here, we show that HDAC6 promotes BACE1 protein stability through direct deacetylation of its C-terminal lysine (K501), thereby increasing Aβ production. HDAC6 also facilitated NLRP3 inflammasome activation in micro

Nrf2 promotes NLRP3 inflammasome assembly and activation by Klf9-TXNIP axis.

NLRP3 inflammasome activation shows a crucial role in the innate immune response that triggers inflammation. Activation is controlled by two sequential steps: a priming step, followed by an assembly step. Nrf2 has been shown to promote NLRP3 inflammasome activation; however, the underlying mechanisms remain elusive. This study identifies Nrf2 as an encourager of NLRP3 inflammasome activation. We show that Nrf2 inhibition suppresses NLRP3 inflammasome activation in macrophages, including THP-1 cells and BMDMs. In addition, Nrf2 facilitates NLRP3 inflammasome assembly. Mechanically, Nrf2 increas

Advanced nanoformulations of NUAK1 regulate NLRP3 inflammasome for preeclampsia management in mice.

Preeclampsia (PE) is a severe pregnancy complication driven by placental dysfunction and oxidative stress, which currently lacks effective targeted therapies. Here, we evaluated the therapeutic potential of folic acid-modified lipid nanoparticles (NPs) co-loaded with polydopamine and NUAK Family Kinase 1 (NUAK1), termed FLN@(PDA + NUAK1) NPs, for mitigating PE. In vitro, these NPs effectively reduced reactive oxygen species levels and suppressed activation of the NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasome, while promoting autophagic activity and inhibiting apoptosis under oxidat

Ergosterol suppresses NLRP3 inflammasome-mediated inflammatory responses through Bmal1.

The NLRP3 inflammasome is a pivotal sensor in innate immune cells that initiates the inflammatory cascade. Upon exposure to stimuli, it follows a two-stage activation mechanism involving an initial priming phase and a subsequent activation phase. Under pathological conditions, its activation leads to undesirable inflammation that drives or contributes to the pathogenesis of various complex diseases, such as colitis and metabolic diseases. Therefore, curbing NLRP3 inflammasome activity is highly advisable, but no specific NLRP3-targeted medications are available for clinical use. We previously

Inflammasome adaptor protein ASC is a mechanistic checkpoint in IL-1β maturation.

Inflammasome activation triggers caspase-1-dependent maturation of pro-interleukin-1β (pro-IL-1β) in an apoptosis-associated speck-like prot … positions pro-IL-1β for efficient caspase-1 processing, which adds a mechanistic checkpoint within inflammasomes to license cytokine maturation independently of pyroptotic death. We propose that ASCLD may be a therapeutically tractable node for selective modulation of IL-1β-driven inflammasomopathies.

Pyroptosis-associated inflammasome activation contributes to scleral remodeling in experimental myopia.

AIM: To explore whether pyroptosis, an inflammatory type of programmed cell death, participates in the initiation and progression of myopia, and to further elucidate its regulatory role in scleral remodeling. METHODS: Scleral tissues from form-deprivation myopia (FDM) mouse models were subjected to transcriptome sequencing to screen inflammatory and cell death-related molecular characteristics. Differentially expressed gene analysis and pathway enrichment analysis were adopted to identify pyroptosis-related signaling pathways. Meanwhile, human scleral fibroblasts were treated with complement

Targeting the NLRP3 inflammasome in Alzheimer's disease: Mechanistic insights and therapeutic advances.

Alzheimer's disease (AD) is the leading cause of dementia, yet current therapies provide limited clinical benefit. Neuroinflammation, as an early and sustained driver of AD, places the NLRP3 inflammasome at the center of pathological and therapeutic focus. In this review, we synthesize recent advances in the structure, assembly, and activation of the NLRP3 inflammasome, and evaluate its contribution to AD using evidence from human brain tissues, cerebrospinal fluid, and diverse AD animal models. Available data consistently support aberrant NLRP3 activation in AD brain, where it is closely asso

TLR9-mediated activation of the NLRP3 inflammasome in Fusarium solani-induced keratitis.

Toll-like receptor 9 (TLR9) is a crucial innate immune receptor that recognises pathogenic DNA and initiates downstream inflammatory signaling, yet its function in fungal keratitis remains poorly defined. This study aimed to clarify the specific role of TLR9 in Fusarium solani (F. solani) -induced keratitis and to provide mechanistic insights for targeted therapy. A murine model of F. solani keratitis was established to examine the expression and regulation of TLR9 and NLRP3. RNA sequencing, quantitative real-time polymerase chain reaction (qRT-PCR), Western blotting, and immunofluorescence we

TKI-mediated inhibition of NLRP1 inflammasome restores erythropoiesis in DBA syndrome.

Diamond-Blackfan anemia syndrome (DBAS) is marked by defective erythropoiesis caused by impaired ribosome biogenesis and aberrant signaling. Here, we investigate how ribosomal stress-induced activation of the NLRP1 inflammasome affects erythroid differentiation in DBAS. We demonstrate that FDA/EMA-approved tyrosine kinase inhibitors (TKIs) effectively mitigate defective erythropoiesis by inhibiting NLRP1 inflammasome activation. In K562 cells, nilotinib suppresses the ZAKα/P38/NLRP1/CASP1 axis, leading to increased GATA1 levels and upregulation of key erythroid genes. These effects were valida

The mitophagy-inflammasome axis: a shared pathological hub in Alzheimer's and Parkinson's diseases.

Alzheimer's disease (AD) and Parkinson's disease (PD) represent the most prevalent chronic neurodegenerative disorders, characterized by progressive loss of neurons as a core pathological feature. Despite discrepancies in their clinical phenotypes and signature pathological proteins, accumulating evidence has validated a common molecular pathogenic mechanism: dysfunctional bidirectional crosstalk between mitophagy and inflammasomes. As the central hub of neuronal energy metabolism, mitochondrial impairment triggers the release of damage-associated molecular patterns such as reactive oxygen spe…

Targeting the NLRP3 inflammasome with antibody-based therapeutics for chronic neurodegenerative diseases.

INTRODUCTION: The NLRP3 inflammasome is a central regulator of innate immunity that becomes aberrantly activated by amyloid-β, hyperphosphorylated tau, and α-synuclein aggregates in chronic neurodegenerative diseases, such as Alzheimer's (AD) and Parkinson's disease (PD). Sustained activation drives neuroinflammation, synaptic dysfunction, and neuronal loss, making NLRP3 a compelling therapeutic target. TOPICS COVERED: This review summarizes current insights into NLRP3 inflammasome biology in AD and PD, with emphasis on antibody-based interventions. Emerging delivery approaches, such as recep

Identification of Chlamydia pneumoniae and NLRP3 inflammasome activation in Alzheimer's disease retina.

Chlamydia pneumoniae is an intracellular bacterium implicated in Alzheimer's disease (AD), but its role in retinal pathology and disease progression is unclear. Here we identify Chlamydia pneumoniae inclusions in the retina, showing higher burden in AD retina and brain, increasing with APOEε4, disease stage, and cognitive deficit. Retinal and cortical proteomics reveal bacterial-infection and related NLRP3-inflammasome pathways. Retinal NLRP3 is elevated in mild cognitive impairment and activated in AD dementia, evidenced by increased caspase-1, cleaved interleukin-1β, and cleaved N-terminal g

Inflammasome Dynamics in the High-Risk Clone ST235 with Altered Pyoverdine Structures.

Pseudomonas aeruginosa ST235 isolates drive a distinct macrophage inflammatory program characterized by IL-1β axis dominance and suppression of canonical NF-κB-associated cytokine responses. In THP-1-derived macrophages, ST235 isolates induce robust inflammasome activation, resulting in elevated IL-1β production and altered cell death responses, while eliciting markedly lower TNFα, IL-6, IL-8, and IL-10 responses than non-ST235 isolates. In contrast, nonST235 isolates preferentially stimulate NF-κB-dependent cytokine production with weaker inflammasome activation. Mechanistically, ST235 infect

Inflammasome activation dictates the efficacy of antimycobacterial activity of frontline TB drugs.

Recent developments in tuberculosis (TB) treatment have identified an enormous potential of host-directed therapies (HDT)) in achieving better and faster control of infection. We have previously demonstrated the synergistic effect of sertraline (SRT) with frontline TB drugs in clearing infection in murine tissues. Our attempts to uncover the mechanistic basis of this enhancement, using sertraline as a probe, help identify host signalling pathways critical for controlling Mycobacterium tuberculosis (Mtb). We identify a significant role for sertraline-mediated modulation of mitochondrial physiol

Repurposing Nelfinavir: AIM2 inflammasome-associated anti-tumor effects in Glioblastoma.

Nelfinavir (NFR), originally developed as an antiretroviral agent for the human immunodeficiency virus, has demonstrated anti-cancer properties across various malignancies; however, its therapeutic potential in Glioblastoma (GBM) remains largely unexplored. In the present study, we investigated the anti-tumor effects of NFR in GBM using a comprehensive panel of experimental models, including established GBM cell lines, GBM cell line-derived spheroids, patient-derived primary glioma cells, and patient-derived glioma organoids. We further evaluated the anti-tumor potential of NFR in combination
