This study describes the antispasmodic activity of some fractions and cynaropicrin, a sesquiterpene lactone from Cynara scolymus, cultivated in Brazil, against guinea-pig ileum contracted by acetylcholine. The dichloromethane fraction showed the most promising biological effects, with an IC(50) of 0.93 (0.49-1.77) mg/ml. We herein report a copper-catalyzed 1,3-aminocyclization of cyclopropanes as a direct and versatile entry into important heterocycles. This reaction was initiated by a copper-catalyzed, NFSI-promoted ring opening of cyclopropanes, followed by nucleophilic cyclization. A variety of nucleophiles successfully participate in this transformation, including alcohols, carboxylic acids, sulfonamides, and amides, for the construction of diverse cyclic ethers, pyrrolidines, lactones, and iminolactones.

An inexpensive iron-catalyzed alkoxyl radical-induced C-C bond cleavage/gem-difluoroalkylation cascade is presented. Regulated by the structure of alkoxyl radical precursors, fluorinated distal diketones were synthesized through a ring-opening strategy and difluoroalkylated medium-sized lactones and macrolactones were constructed via a ring-expansion strategy. Both protocols proceeded under mild and redox neutral conditions with a broad substrate scope and good functional group compatibility.

We recently reported that parthenolide and related sesquiterpene lactones are able to prevent and reverse behavioral responses in planarian worms induced by acute cocaine exposure. Previous reports indicate that when planarians are chronically exposed to microM concentrations of cocaine, they display stereotypical withdrawal-like behaviors when the cocaine is removed. Here we report that parthenolide prevents this cocaine-induced expression of planarian withdrawal-like behaviors.

Some recent publications concerning perfume contact sensitivity are briefly reviewed. Japanese workers have identified some perfume sensitizers and have devised an allergen replacement system. Costus absolute of perfumery has been found to sensitize guinea-pig and man; sesquiterpene lactones are responsible for cross-sensitivity with some other plant products. Balsam of Peru is a useful screen for perfume sensitivity; terpenoids of balsams require study.

A photoredox-catalyzed radical heterocyclization of alkenoic acids and sulfonamides with α-iodonium diazo compounds is reported. The reaction delivers diazomethyl-containing lactones and pyrrolidines in good yields with full retention of the diazo group. The synthetic utility is demonstrated by a gram-scale synthesis and downstream derivatizations under mild, redox-neutral conditions. A plausible radical-polar crossover mechanism is proposed for this transformation.
