Horizontal and vertical transmission of wild-type and recombinant Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus.

Transmission plays a central role in the ecology of baculoviruses and the population dynamics of their hosts. Here, we report on the horizontal and vertical transmission dynamics of wild-type Helicoverpa armigera single-nucleocapsid nucleopolyhedrovirus (HaSNPV-WT) and a genetically modified variant (HaSNPV-AaIT) with enhanced speed of action through the expression of an insect-selective scorpion toxin (AaIT). In caged field plots, horizontal transmission of both HaSNPV variants was greatest when inoculated 3rd instar larvae were used as infectors, transmission was intermediate with 2nd instar

Herbivore-induced volatile production by Arabidopsis thaliana leads to attraction of the parasitoid Cotesia rubecula: chemical, behavioral, and gene-expression analysis.

Many plant species defend themselves against herbivorous insects indirectly by producing volatiles in response to herbivory. These volatiles attract carnivorous enemies of the herbivores. Research on the model plant Arabidopsis thaliana (L.) Heynh. has contributed considerably to the unraveling of signal transduction pathways involved in direct plant defense mechanisms against pathogens. Here, we demonstrate that Arabidopsis is also a good candidate for studying signal transduction pathways involved in indirect defense mechanisms by showing that: (1) Adult females of Cotesia rubecula, a specia

Physicochemical characterization and optimization of Tulsi-mediated silver nanocomposites with enhanced insecticidal activity against Spilosoma obliqua.

Spilosoma obliqua Walker (Lepidoptera: Erebidae) is one of the major pests of jute plants, severely reducing fibre yield and consequently lowering agricultural productivity. To address this problem through an eco-friendly approach, the green synthesis of a series of nano-biopesticide formulations, named E, P, Q, R, S, and T, based on nano-bio-composites obtained from varying proportions of Ocimum sanctum and in situ-formed silver nanoparticles, is proposed. Furthermore, their larvicidal efficacies are compared. These green-synthesised nano-bio-composites were characterised using FTIR and Raman

Global Risk Assessment of the Invasive Insect Thaumatotibia leucotreta to Citrus Based on its Potential Distribution.

The false-codling moth (FCM), Thaumatotibia leucotreta Meyrick (Lepidoptera: Tortricidae), is a globally recognized quarantine pest responsible for substantial economic losses in the southern African citrus industry. Driven by international trade and climate change, FCM poses a potential threat to regions beyond its native range. In this study, we employed the MaxEnt model to predict suitable habitats for FCM under near-current and future climate scenarios based on 141 global occurrence data and 19 bioclimatic variables. The model performed excellently with an AUC of 0.965. The key bioclimatic

Programmed meiotic errors facilitate dichotomous sperm production in the silkworm, Bombyx mori.

The goal of meiosis is typically to produce haploid gametes (eggs or sperm). Failure to do so is catastrophic for fertility. However, Lepidopteran (moths and butterflies) males produce two sperm morphs: nucleated (eupyrene) sperm and anucleated (apyrene) sperm, both of which are essential for fertilization. The meiotic differences in the two types of spermatogenesis are unclear, and our knowledge of the molecular differences between eupyrene and apyrene spermatogenesis is extremely limited in all systems. The only factor identified as being required for apyrene spermatogenesis is Sex-lethal (S

A decade of fall armyworm invasion in Africa (2016 to 2026): lessons learned, progress in management, and pathways for sustainable control.

The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), is a highly destructive pest native to the Americas that was first detected in Africa in 2016. Nearly a decade later, it continues to pose a significant threat to maize production, a staple crop for millions across the continent. The pest rapidly spread, colonizing most maize-growing regions within 2yr and causing estimated annual yield losses of up to 20 million metric tons, adversely affecting food security, rural livelihoods, and economies. Farmers who rely heavily on pesticides risk the emergence of resis

Diet quality improves survival and mediates larval response to densovirus infection in the Melissa blue butterfly.

Entomopathogenic viruses affect insect populations by reducing fitness and influencing other measures of performance such as development and longevity. Given the huge diversity of viruses, studies are needed to understand the mechanisms underlying how viruses affect their insect hosts. The Junonia coenia densovirus (JcDV) infects a wide range of Lepidoptera and can cause death in the larval and pupal stages. In this study, we used the Melissa blue butterfly (Lycaeides melissa: Lycaenidae) to describe infection progression at the individual level, the effects of infection on development, surviv

Divergent Selection Promotes Intraspecific Genomic Differentiation in Spodoptera littoralis With Possible Involvement in Detoxification.

The cotton leafworm, Spodoptera littoralis (Lepidoptera: Noctuidae), is a major agricultural pest affecting crops like cotton, maize, tomatoes, and wheat across southern Europe, Africa, the Middle East, and western Asia. Whole genome analyses have revealed adaptive evolution in chemosensation and detoxification genes in S. littoralis. However, the extent of intraspecific diversity influenced by recent adaptive evolutionary forces remains unclear. In this study, we conducted a population genomics analysis using 31 S. littoralis individuals from sub-Saharan Africa, northern Africa, and southern

Functional role of TaGSTs1 in the detoxification of spinetoram and development of Tuta absoluta.

Tuta absoluta has evolved resistance to many biological insecticides, resulting in significant annual agricultural and economic losses. Glutathione S-transferases (GSTs) are one of the major insect detoxification enzyme systems. However, the detoxification metabolism of GSTs in T. absoluta against biological insecticides remains poorly understood. In this study, We identified five key GST genes (TaGSTs1, TaGSTs2, TaGSTe1, TaGSTe3, and TaGSTd1) by screening from the comparative transcriptomes of two regional populations of T. absoluta in Xinjiang, China. Among the five GSTs, TaGSTs1 exhibited a

Larval and Larval-Pupal Parasitoids Associated with Major Owlet Moth Pests of Soybean and Maize in the Brazilian Savanna: Measures to Preserve Them in Crop Succession.

Owlet moths (Lepidoptera: Noctuoidea) are among the most threatening pests of soybean and maize systems under intensive cropping in tropical and subtropical regions. Their high dispersal, larval polyphagia, and voracious feeding behavior often lead to severe grain production losses. Many parasitoids are associated with owlet moths; however, the ecological importance of larval parasitism in the soybean-maize crop rotation of the Brazilian savanna remains uncertain. We assessed larval parasitism in major lepidopteran pests of soybean and maize crops over three agricultural seasons in eight field

Functional Characterization and Toxicological Study of Cordyceps militaris in Weaned Pigs.

Cordyceps militaris (CM), a well-known parasitic fungus that grows on the larvae of Lepidoptera, has a variety of pharmacological activities. However, little is known about its safe dosage for animals, including pigs. To explore its effect on intestinal health and evaluate its safe dosage, 30 weaned pigs were randomly allotted to five groups and fed with a basal diet supplemented with different doses of CM for 42 days. The results showed that CM supplementation at 100 mg/kg increased the average daily weight gain (ADG) and significantly decreased the ratio of feed intake to gain (F:G) in the w

Effects of surfactin stress on gene expression and pathological changes in Spodoptera litura.

Spodoptera litura (S. litura) is a polyphagous pest of the family Lepidoptera, which causes damage and yields losses to many crops. The long-term use of chemical pesticides for control not only seriously threatens environmental health, but also causes S. litura to develop drug resistance. Therefore, there is an urgent need to develop environmentally safe and friendly biogenic pesticides. However, the mechanism of action of the secondary metabolite (surfactin) of Bacillus Vélezensis (B. vélezensis) on lepidopteran pests (S. litura) has not been reported yet. We found that several metabolites an

Bionomic responses of Spodoptera frugiperda (J. E. Smith) to lethal and sublethal concentrations of selected insecticides.

Since 2016, the invasive insect Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) from the Americas has made maize production unattainable without pesticides in parts of Sub-Saharan Africa and Asia. To counteract this pest, farmers often resort to the use hazardous pesticides. This study aimed to investigate botanicals, microbials, and semi-synthetic insecticides in Ghana for pest control without harming local ecosystems. Under laboratory and on-station conditions, the present study evaluated the acute and sublethal responses of S. frugiperda to: (i) Pieris rapae Granulovirus (PrGV)

Potential Management Options for the Invasive Moth Spodoptera frugiperda in Europe.

We here review and discuss management options that growers in Europe could take in response to the expected invasion of the fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae). The focus is put on maize but the information provided is also relevant for other crops potentially affected. A sound forecasting system for fall armyworm both on a regional as well as at local scale should be established to alert growers as early as possible. Whilst a number of cultural control methods are adopted by maize growers in different regions globally to fight fall armyworm, many of them may either n

Reduced Mythimna separata infestation on Bt corn could benefit aphids.

Use of genetically engineered plants that express insecticidal Cry proteins derived from Bacillus thuringiensis (Bt) have been proven efficacious for managing lepidopteran pests. However, in some cases herbivores that are not targeted by the Bt trait have increased in importance. It has been suggested that reduced caterpillar damage to Bt crops could lead to decreased levels of induced plant defensive compounds which might benefit other non-target herbivores. Here we investigated the potential effect of reduced damage by larvae of Mythimna separata on aphid populations in Bt corn. We compared

Spatial separation of the cyanogenic β-glucosidase ZfBGD2 and cyanogenic glucosides in the haemolymph of Zygaena larvae facilitates cyanide release.

Low molecular weight compounds are typically used by insects and plants for defence against predators. They are often stored as inactive β-glucosides and kept separate from activating β-glucosidases. When the two components are mixed, the β-glucosides are hydrolysed releasing toxic aglucones. Cyanogenic plants contain cyanogenic glucosides and release hydrogen cyanide due to such a well-characterized two-component system. Some arthropods are also cyanogenic, but comparatively little is known about their system. Here, we identify a specific β-glucosidase (ZfBGD2) involved in cyanogenesis from l

Mutations of an arylalkylamine-N-acetyltransferase, Bm-iAANAT, are responsible for silkworm melanism mutant.

Coloration is one of the most variable characters in animals and provides rich material for studying the developmental genetic basis of pigment patterns. In the silkworm, more than 100 gene mutation systems are related to aberrant color patterns. The melanism (mln) is a rare body color mutant that exhibits an easily distinguishable phenotype in both larval and adult silkworms. By positional cloning, we identified the candidate gene of the mln locus, Bm-iAANAT, whose homologous gene (Dat) converts dopamine into N-acetyldopamine, a precursor for N-acetyldopamine sclerotin in Drosophila. Isolation, synthesis and pharmacological characterization of delta-palutoxins IT, novel insecticidal toxins from the spider Paracoelotes luctuosus (Amaurobiidae).

Four novel insecticidal toxins were isolated from the venom of the spider Paracoelotes luctuosus (Araneae: Amaurobiidae) and named delta-palutoxins IT1 to IT4. The four toxins are homologous 36-37 amino acid peptides reticulated by four disulfide bridges and three have amidated C-terminal residues. The delta-palutoxins are highly homologous with the previously described mu-agatoxins and curtatoxins (77-97%). The four peptides demonstrated significant toxicity against larvae of the crop pest Spodoptera litura (Lepidoptera: Noctuidae) in a microinjection bioassay, with LD50 values in the 9-50 mi
