AMPHIBIAN PARASITES EXHIBIT IDIOSYNCRATIC RELATIONSHIPS WITH SPATIOTEMPORAL ENVIRONMENTAL AND HOST-COMMUNITY VARIATION.

Why parasites occur in certain hosts in certain locations has been a long-standing question among ecological and evolutionary parasitologists. Encounter and compatibility filters summarize the likelihood that a host and parasite will physically interact and establish an infection upon contact. Encounter and compatibility filters are not fixed and, among multiple locations, the abiotic environmental characteristics and biotic community composition that contribute to the filters often vary spatially and temporally. Abiotic variation may directly affect hosts or parasites-particularly parasites w

Infection with Babesia bovis alters metabolic rates of Rhipicephalus microplus ticks across life stages.

BACKGROUND: Metabolic responses to infection differ based on arthropod and pathogen. Increased metabolic rates can result in faster depletion of energetic resources, and decreases may allow for energy conservation. Babesia bovis is a protozoan pathogen transmitted by the cattle fever tick, Rhipicephalus microplus. Adult female ticks acquire B. bovis by feeding on an infected animal. Babesia bovis undergoes development and invades the ovaries where it is transmitted transovarially to tick offspring. The effects of infection on R. microplus metabolic rate are not well studied. METHODS: We teste

An ApiAP2 Family Transcriptional Factor PfAP2-06B Regulates Erythrocyte Invasion Indirectly in Plasmodium falciparum.

Obligate intracellular parasites must efficiently invade host cells to complete their life cycle and facilitate transmission. For the malaria-causing parasite Plasmodium falciparum, the invasion of an erythrocyte is a critical process, and thereby a key target for intervention strategies. In this study, we investigate the role of the ApiAP2 family transcription factor PfAP2-06B (PF3D7_0613800) in the intraerythrocytic developmental cycle of P. falciparum and focus on its regulation of genes involved in erythrocyte invasion. Conditional knockdown of PfAP2-06B resulted in a defect in asexual gro

Organ Tropism of Angiostrongylus vasorum Larval Stages in Infected African Giant Snails (Lissachatina fulica).

Angiostrongylus vasorum is a metastrongyloid lungworm causing severe cardiovascular disease in domestic and wild animals. During its heteroxenous life cycle, A. vasorum requires obligate gastropod intermediate hosts. Little is known about A. vasorum larval organ tropism and development in gastropod intermediate hosts. Thus, the aim of this study was to analyze in vivo development of A. vasorum larval stages in experimentally infected African giant snails (Lissachatina fulica). Adult L. fulica (n = 26) were orally infected with A. vasorum-L1 and thereafter continuously euthanized. Gastropod org

Understanding the significance of oxygen tension on the biology of Plasmodium falciparum blood stages: From the human body to the laboratory.

Plasmodium falciparum undergoes sequestration within deep tissues of the human body, spanning multiple organ systems with differing oxygen (O2) concentrations. The parasite is exposed to an even greater range of O2 concentrations as it transitions from the human to the mosquito host, suggesting a high level of plasticity as it navigates these different environments. In this review, we explore factors that may contribute to the parasite's response to different environmental O2 concentrations, recognizing that there are likely multiple pieces to this puzzle. We first review O2-sensing mechanisms

Climatic variations and Fasciola: a review of impacts across the parasite life cycle.

Fasciolosis, caused by the liver fluke Fasciola spp., is a significant parasitic disease of livestock and humans worldwide. Fasciola transmission and life cycle are highly dependent on climatic conditions, especially temperature and humidity. This dependency has gained significance in the context of ongoing climate change. This literature review examined evidence on the effects of temperature variability on the developmental stages of Fasciola spp. and the snail intermediate hosts. We reviewed free larval stages of Fasciola spp. development, as well as snail intermediate hosts, while investiga

Hepatitis E Virus Life Cycle.

Hepatitis E virus (HEV) infects over 20 million people worldwide per year, leading to 30,000-40,000 deaths. In most cases HEV infection in a self-limited, acute illness. However, chronic infections could occur in immunocompromised individuals. Due to scarcity of robust cell culture models in vitro and genetic tractable animal models in vivo, the details of HEV life cycle, as well as its interaction with host cells still remain elusive, which dampens antivirals discovery. In this chapter, we present an update in the HEV infectious cycle steps: entry, genome replication/subgenomic RNA transcript

Analysis of sex-specific lipid metabolism of Plasmodium falciparum points to the importance of sphingomyelin for gametocytogenesis.

Male and female Plasmodium falciparum gametocytes are the parasite lifecycle stage responsible for transmission of malaria from the human host to the mosquito vector. Not only are gametocytes able to survive in radically different host environments, but they are also precursors for male and female gametes that reproduce sexually soon after ingestion by the mosquito. Here, we investigate the sex-specific lipid metabolism of gametocytes within their host red blood cell. Comparison of the male and female lipidome identifies cholesteryl esters and dihydrosphingomyelin enrichment in female gametocy

A new species, new host records and life cycle data for lepocreadiids (Digenea) of pomacentrid fishes from the Great Barrier Reef, Australia.

A new species of lepocreadiid, Opechonoides opisthoporus n. sp., is described infecting 12 pomacentrid fish species from the Great Barrier Reef, Australia, with Abudefduf whitleyi Allen & Robertson as the type-host. This taxon differs from the only other known member of the genus, Opechonoides gure Yamaguti, 1940, in the sucker width ratio, cirrus-sac length, position of the testes, position of the pore of Laurer's canal, and relative post-testicular distance. The new species exhibits stenoxenic host-specificity, infecting pomacentrids from seven genera: Abudefduf Forsskål, Amphiprion Bloch &

Co-aggregation and secondary nucleation in the life cycle of human prolactin/galanin functional amyloids.

Synergistic-aggregation and cross-seeding by two different proteins/peptides in the amyloid aggregation are well evident in various neurological disorders including Alzheimer's disease. Here, we show co-storage of human Prolactin (PRL), which is associated with lactation in mammals, and neuropeptide galanin (GAL) as functional amyloids in secretory granules (SGs) of the female rat. Using a wide variety of biophysical studies, we show that irrespective of the difference in sequence and structure, both hormones facilitate their synergic aggregation to amyloid fibrils. Although each hormone posse

The Basal Complex Protein PfMORN1 Is Not Required for Asexual Replication of Plasmodium falciparum.

Plasmodium falciparum, the Apicomplexan parasite that causes the most severe form of human malaria, divides via schizogony during the asexual blood stage of its life cycle. In this method of cell division, multiple daughter cells are generated from a single schizont by segmentation. During segmentation, the basal complex forms at the basal end of the nascent daughter parasites and likely facilitates cell shape and cytokinesis. The requirement and function for each of the individual protein components within the basal complex remain largely unknown in P. falciparum. In this work, we demonstrate

The impact of temperature on the life cycle of Gasterophilus pecorum in northwest China.

BACKGROUND: The departure of the mature larvae of the horse stomach bot fly from the host indicates the beginning of a new infection period. Gasterophilus pecorum is the dominant bot fly species in the desert steppe of the Kalamaili Nature Reserve (KNR) of northwest China as a result of its particular biological characteristics. The population dynamics of G. pecorum were studied to elucidate the population development of this species in the arid desert steppe. METHODS: Larvae in the freshly excreted feces of tracked Przewalski's horses (Equus przewalskii) were collected and recorded. The larv

Life Cycle Dominates the Volatilome Character of Dimorphic Fungus Coccidioides spp.

Valley fever (coccidioidomycosis) is an endemic fungal pneumonia of the North and South American deserts. The causative agents of Valley fever are the dimorphic fungi Coccidioides immitis and C. posadasii, which grow as mycelia in the environment and as spherules within the lungs of vulnerable hosts. Current diagnostics for Valley fever are severely lacking due to poor sensitivity and invasiveness, contributing to a 23-day median time to diagnosis, and therefore, new diagnostic tools are needed. We are working toward the development of a breath-based diagnostic for coccidioidomycosis, and in t

Dengue Virus Infection: A Tale of Viral Exploitations and Host Responses.

Dengue is a mosquito-borne viral disease (arboviral) caused by the Dengue virus. It is one of the prominent public health problems in tropical and subtropical regions with no effective vaccines. Every year around 400 million people get infected by the Dengue virus, with a mortality rate of about 20% among the patients with severe dengue. The Dengue virus belongs to the Flaviviridae family, and it is an enveloped virus with positive-sense single-stranded RNA as the genetic material. Studies of the infection cycle of this virus revealed potential host targets important for the virus replication

The endoplasmic reticulum-resident serpentine receptor SR10 has important functions for asexual and sexual blood stage development of Plasmodium falciparum.

Serpentine receptors (SRs) are transmembrane proteins generally acting as mediators to facilitate the communication between a cell and its environment. At least six putative SR-like proteins are encoded in the genome of the malaria parasite Plasmodium falciparum. For two of them, roles in cell stress control were reported; however, for most of the SR-like proteins the functions are not yet known. In this study, we provide a first phenotypic analysis of the plasmodial SR10. The transmembrane protein is expressed in the asexual and sexual blood stages of P. falciparum. Co-localization and co-imm

New Antiparasitic Bis-Naphthoquinone Derivatives.

A series of bis-naphthoquinone derivatives prepared by condensation of aryl aldehydes with lawsone were tested for antiparasitic activities against Toxoplasma gondii and Trypanosoma brucei parasites. Monofluorophenyl derivative 1a, 3,4-difluorophenyl analog 1c and furyl compound 1l exhibited significant activity against T. gondii cells and appear to be new promising drug candidates against this parasite. The 3,4,5-trifluorophenyl derivative 1g and the isovanillyl derivative 1j displayed selective activity against Leishmania major amastigotes.

Effect of host fruit, temperature and Wolbachia infection on survival and development of Ceratitis capitata immature stages.

The Mediterranean fruit fly, Ceratitis capitata (Diptera: Tephritidae), holds an impressive record of successful invasions promoted by the growth and development of international fruit trade. Hence, survival of immatures within infested fruit that are subjected to various conditions during transportation seems to be a crucial feature that promotes invasion success. Wolbachia pipientis is a common endosymbiont of insects and other arthropods generating several biological effects on its hosts. Existing information report the influence of Wolbachia on the fitness traits of insect host species, in

Globally Regionalized Monthly Life Cycle Impact Assessment of Particulate Matter.

This work provides a globally regionalized approach for quantifying particulate matter (PM2.5) health impacts. Atmospheric transport and pollutant chemistry of primary particulate matter, sulfur dioxide (SO2), nitrogen oxide (NOx), and ammonia (NH3) from stack emissions were modeled and used to calculate monthly high-resolution maps of global characterization factors that can be used for life cycle impact assessment (LCIA) and risk assessment. These characterization factors are applied to a global data set of coal power emissions. The results show large regional and temporal differences in hea

Pesticide selectivity to the parasitoid Trichogramma pretiosum: A pattern 10-year database and its implications for Integrated Pest Management.

Trichogramma pretiosum is one of the main egg parasitoids used in the control of lepidopteran pests in Brazil. This natural enemy can be negatively affected by the use of insecticides, herbicides, and fungicides. The present work used a systematic review and meta-analysis to group information from multiple studies on the selectivity of pesticides (279 commercial products) in rice, corn, soybean, apple and peach crops for immature stages (egg-larva, pre-pupa, and pupa) and adult parasitoids. The selected studies used the International Organization for Biological and Integrated Control (IOBC) me
